En el presente estudio se evaluó en qué medida el extracto hidroalcohólico de las hojas del Smallanthus sonchifolius "yacón" y el extracto acuoso de las semillas de Vitis vinifera "uva" son seguras y eficaces en el efecto sinérgico frente a la actividad hipoglucemiante en ratas con inducción experimental de diabetes mellitus tipo 2. La seguridad en animales vía oral se realizó a dosis única en Rattus norvegicus albinus machos de la cepa Holtzman y a dosis repetidas durante 28 días en Rattus norvegicus albinus machos y hembras de la misma cepa. La inducción de diabetes mellitus tipo 2 a Rattus norvegicus albinus se realizó con el aloxano monohidratado (Sigma, Saint Louis, MO, EE. UU.) a dosis única de 100 mg/kg de peso. Los resultados indicaron una dosis letal 50 (DL50) por encima de los 2000 mg/kg para ambos extractos, por lo que son sustancias no tóxicas y, en el ensayo de toxicidad a dosis repetidas, se halló ligero incremento de alanino aminotransferasa; no se encontraron lesiones microscópicas ni macroscópicas relacionadas con los tratamientos. En el ensayo sobre la diabetes mellitus tipo 2, los grupos de glibenclamida, Smallanthus sonchifolius "yacón", Vitis vinifera "uva" y la sinergia de Smallanthus sonchifolius + Vitis vinifera disminuyeron la glucemia, siendo significante comparado con el grupo control positivo SSF (p<0.05). La sinergia de los extractos de Smallanthus sonchifolius + Vitis vinifera representa mayor disminución de la glucemia (p<0.05). El posible mecanismo de acción de la sinergia de los extractos es que mejoraría la secreción de insulina por el páncreas. En las condiciones experimentales, la sinergia del extracto hidroalcohólico de las hojas del Smallanthus sonchifolius "yacón" y el extracto acuoso de las semillas de Vitis vinifera "uva" ha evidenciado ser segura y tener efecto hipoglucemiante en Rattus norvegicus albinus diabéticas inducidas por aloxano.
