Loss of sugars contributes to accelerate postharvest senescence of broccoli. Several treatments have been developed to delay senescence, but in many cases their effects on sugar metabolism were not analyzed. We studied the effect of harvest at different times of day (08:00, 13:00 and 18:00 h) and of several postharvest treatments as heat treatment (HT), modified atmosphere (MA) and 1-methylcylcopropene (1-MCP) on sugar levels and activities of enzymes related to sucrose and starch degradation. Harvesting at the end of day delayed the loss of chlorophylls and caused the lowest decrement in sugars, although no differences in invertase, sucrose synthase and β-amylase activities were detected among samples. Treatments of MA and 1-MCP caused a lower loss of glucose and fructose, while HT caused a lower decrement of sucrose. Treated samples maintained higher levels of chlorophylls. The treatments reduced the activity of invertase and sucrose synthase and induced higher levels of β-amylase activity. Harvesting at the end of day and performing simultaneously a MA treatment could be a good combination to maintain the green color of the inflorescence and sugar levels during postharvest of broccoli.
La cosecha y comercialización del brócoli se realiza en momentos en que la inflorescencia se encuentra en pleno desarrollo. La separación de la inflorescencia de la planta madre detiene el suministro de agua, hidratos de carbono provenientes de la fotosíntesis, otros nutrientes y hormonas, principalmente citocininas, las cuales regulan el proceso de senescencia en la planta, retardándolo. Estos factores, sumados a una alta tasa respiratoria conducen a una senescencia acelerada y rápido deterioro postcosecha del producto. Entre los principales síntomas de la senescencia en brócoli se destaca la ocurrencia de un amarilleamiento de la inflorescencia, consecuencia del inicio del catabolismo de las clorofilas. Se produce degradación de proteínas, disminución del nivel de azúcares solubles y de reserva como el almidón, lo que conlleva al inicio de nuevas vías de obtención de energía como la proveniente de los ácidos grasos de membrana, por lo que las células van perdiendo la permeabilidad selectiva de sus membranas. Asimismo, aumenta el número de especies reactivas de oxígeno y disminuye el poder antioxidante. Dada la importancia de los hidratos de carbono en el inicio de la senescencia y la variación existente en el contenido de los mismos en la planta a lo largo del día, dependiente de los ciclos de luz/oscuridad, en el presente trabajo de tesis doctoral se estudió el efecto de la cosecha a diferentes horas del día (8, 13 y 18 h) sobre la senescencia postcosecha. Se encontraron diferencias en el cambio de color, contenido de clorofilas y de azúcares solubles, así como también en la expresión de genes relacionados al catabolismo de clorofilas. Las muestras cosechadas a la tarde mantuvieron mayores contenidos de clorofila, lo que se reflejó en el mantenimiento del color verde por más tiempo, lo que correlacionó con una menor expresión de genes relacionados a la degradación de clorofila como BoCLH2, BoPPH y BoPaO. Las muestras cosechadas tardíamente fueron las que mostraron mayores niveles de almidón y una disminución menos marcada en el nivel de azúcares solubles. La actividad de enzimas relacionadas tanto al catabolismo de sacarosa como de almidón fue superior en las muestras cosechadas a las 18 h sólo en el día inicial, y se mantuvieron sin diferencias entre las muestras cosechadas a diferentes tiempos durante el almacenamiento. Teniendo en cuenta los resultados anteriores, y para determinar el efecto de los azúcares sobre la senescencia, se realizaron ensayos con un suministro externo de glucosa y sacarosa en brócolis cosechados por la mañana. Se halló que las muestras tratadas mantuvieron un nivel de azúcares más alto hacia el final del período postcosecha, manteniendo el color verde por más tiempo, a la vez que retuvieron mayores niveles de clorofilas, contenido de fenoles y antioxidantes. Se estudiaron también diferentes métodos aplicados en postcosecha para retardar la senescencia, como ser las atmósferas modificadas (AM), tratamientos térmicos (TT) y con 1-metilciclopropeno (1-MCP), de modo de evaluar su efecto sobre la actividad de enzimas involucradas en la degradación de almidón y sacarosa. Se encontró un retraso en el amarilleamiento de las muestras almacenadas en AM, las tratadas térmicamente y aquellas a las que se les aplicó 1-MCP, respecto de las utilizadas como controles. Estas últimas mostraron un descenso más acelerado en el contenido de azúcares, con mayor actividad invertasa y sacarosa sintasa durante el período almacenamiento, y con una menor actividad β-amilasa hacia el final de dicho período.
