L’objectif de ce travail, en plus de l’étude phyto-chimique et antifongique, est d’étudier la cinétique des interactions anti-candidosiques de l’huile essentielle (H.E) de Melaleuca alternifolia et de deux antifongiques : l'amphotéricine B (Amph B) et le fluconazole (Fluc). L’huile essentielle de Melaleuca alternifolia a été extraite des feuilles et des tiges terminales à l’aide d’un dispositif de type Clevenger, analysée par chromatographie en phase gazeuse couplée à la spectrométrie de masse. L’activité anti-candidosique a été évaluée selon deux techniques ; la technique de l’aromatogramme et la technique de microdilution. La souche microbienne est un lot de l’ATCC (American Type Culture Collection 90029) de Candida albicans. Le pourcentage et la zone d’inhibition, les concentrations minimales inhibitrices (CMI) et fongicides (CMF) et les indices de concentrations fractionnaires inhibitrices (ICFI) et fongicides (ICFF) ont été déterminés. Le rendement de l’extraction était de 1,98 %. Les composés majoritaires étaient : Le terpinène-4-ol (31,15 %), l’-terpinène (12,02 %) et le -terpinéol (8,12 %). Le diamètre d’inhibition de l’huile essentielle était de 24,52 ± 0,9 mm. Le taux d’inhibition de la pousse de Candida albicans était de 97,35 ± 2,16 %. On a noté une variation des valeurs des CMI et des CMF en fonction des concentrations d’association entre l’huile essentielle et les antifongiques étudiés. Les plus faibles valeurs des ICFI des associations H.E/Amph B et H.E/Fluc ont été respectivement 0,493 et 0,432, celles des ICFF ont été de l’ordre de 0,862 et 0,798. L’huile essentielle de Melaleuca alternifolia est un excellent fongicide. Son activité anti-candidosique majeure in vitro pourrait faire l’objet d’autres études plus soutenues in vivo afin d’enrichir l’arsenal antifongique tout en réduisant les effets médicamenteux indésirables.
