BackgroundBcl-2 plays a major role in the pathobiology and drug resistance of ovarian cancer, and inhibition of bcl-2 was useful for OC therapy. It has previously reported that TW-37, a small-molecule inhibitor of Bcl-2 family proteins, inhibited cell growth and induced apoptosis in many cancer cells. In the present study,we investigate the effect of TW-37 or / and in combination with cisplain on several ovarian cancer (OC) cell lines with high bcl-2 expression.MethodsThe bcl-2 mRNA and protein expression, and the cisplain (DDP) sensitivity of OC cell lines SKOV3, OVCAR3, OV-90 and 3AO and SKOV3DDP were determined by Quantitative real-time RT-PCR,Western blot, and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and fluorescence-activated cell sorting (MTT) assays. The effects of TW-37 alone or combined with cisplain on growth and apoptosis in bcl-2 overexpressed OVCAR3, OV-90 and SKOV3DDP cells was detected by MTT,clonogenic assay, ELISA and terminal deoxynucleotidyl transferase-mediated nick end labeling (TUNEL) assay.ResultsThe cell lines SKOV3 and 3AO were sensitive, whereas OVCAR3, OV-90 and SKOV3DDP were resistant to cisplain. Significant positive correlation was observed between basal bcl-2 mRNA and protein and cisplain sensitivity. Cisplain treatment did not activate bcl-2 in vitro. Treatment with TW-37 inhibited bcl-2 expression in bcl-2 overexpressed OVCAR3, OV-90 and SKOV3DDP cells , and inhibited growth and induced apoptosis ,and increased cisplain killing of the bcl-2 overexpressed cells in a does and time-dependant manner in vitro.ConclusionBcl-2 level positively correlated with sensitivity to cisplain. Treatment with TW-37 was effective alone and in combination with cisplain in bcl-2 overexpressed OC cell lines in vitro. Thus, TW-37 may be a useful therapeutic agent for OCs.

BackgroundAirway management of the obese patient presenting for surgery is more likely to be a challenging problem. Supraglottic airway device has been adopted as a bridge to connect ventilation and tracheal intubation in obese patients who would be suffered with difficult intubation. The optimum sevoflurane concentration for supraglottic airway device insertion allowing spontaneous breathing in 50% of obese patients (ED50) is not known. The purpose of this study was to determine the ED50 of sevoflurane for supraglottic airway device Blockbuster™ insertion with spontaneous breathing in obese patients requiring general anesthesia.MethodsThirty elective obese patients (body mass index 30-50 kg/m2) undergoing bariatric surgery were recruited in this study. The predetermined target sevoflurane concentration (initiating at 2.5% with 0.5% as a step size) was sustained for >5 min using a modified Dixon’s up-and-down method, and then the supraglottic airway device Blockbuster™ was inserted. The patient’s response to supraglottic airway device insertion was classified as either ‘movement’ or ‘no-movement’. The ED50 of sevoflurane were determined by calculating the midpoint concentration of crossover point from ‘movement’ or ‘no-movement’ response.ResultsThe ED50 of sevoflurane for supraglottic airway device Blockbuster™ insertion in obese patients calculated using up-and-down method were 2.50 ± 0.60%. The ED50 and ED95 (95% confidence interval) obtained by probit regression analysis were 2.35 (1.28–3.42) % and 4.03 (3.16–17.83) % for supraglottic airway device Blockbuster™ insertion, respectively.ConclusionWe conclude that the optimum end-tidal sevoflurane concentration required for the supraglottic airway device Blockbuster™ insertion allowing spontaneous breathing in 50% of obese patients (ED50) is 2.5 ± 0.6%.Trial registrationChinese Clinical Trial Registry, ChiCTR-IPR-16009071, Registered on 24 August 2016.

Initial clinical feasibility with the small-diameter chitosan artificial vascular prosthesis has been reported previously. Here, we present the results of mechanical properties of artificial vascular prosthesis with 2, 3, and 4 mm inner diameter (ID) and compare some of the properties with the native blood vessel of dog femoral artery. Thickness wall measurement demonstrated the average wall thickness of the artificial vascular prosthesis with 2, 3, and 4 mm ID and the native blood vessel of the dog femoral artery with 3 mm ID were 0.54 ± 0.022 mm, 0.71 ± 0.032 mm, 0.79 ± 0.026 mm, and 0.67 ± 0.22 mm (n = 20). Water absorption rate of 226.02% ± 8.17%, 216.13% ± 4.86%, and 205.69% ± 4.34% were obtained from 2-, 3-, and 4-mm-diameter artificial vascular prosthesis (n = 12), respectively. Water osmotic pressure of the 2-, 3-, and 4-mm-diameter artificial vascular prosthesis was 39.25 ± 3.35 mmHg, 34.2 ± 4.54 mmHg, and 28.00 ± 2.72 mmHg (n = 20), respectively. Water osmotic amount of the 2-, 3-, and 4-cm-diameter artificial vascular prosthesis (n = 20) was 4.90 ± 0.47 mL/(min cm(2) ), 5.51 ± 0.21 mL/(min cm(2) ), and 6.24 ± 0.71 mL/(min cm(2) ), respectively. The ruptured stretching rate of the artificial vascular prosthesis (n = 20) with 2, 3, and 4 mm ID and the native blood vessel was 1.59% ± 0.14%, 1.99% ± 0.24%, 2.52% ± 0.21%, and 32.16% ± 2.15%, respectively. The longitudinal tensile strength of the artificial vascular prosthesis (n = 20) with 2, 3, and 4 mm ID and the native blood vessel was 8.58 ± 1.98 N, 19.75 ± 4.07 N, 22.92 ± 3.85 N, and 18.76 ± 2.05 N, respectively. The suture retention of the artificial vascular prosthesis (n = 20) with 2, 3, and 4 mm ID and the native blood vessel in dry condition is 5.80 ± 0.51 N, 7.01 ± 0.32 N, 8.49 ± 0.56 N, and 7.92 ± 0.39 N, respectively. The suture retention of the artificial vascular prosthesis (n = 20) with 2, 3, and 4 mm ID in wet condition is 3.87 ± 0.43 N, 4.73 ± 0.37 N, 5.63 ± 0.36 N, and 7.92 ± 0.39 N, respectively. The compliance of the artificial vascular prosthesis with 2, 3, and 4 mm ID and the native blood vessel was 6.5% ± 2.6%/100 mmHg, 5.2% ± 1.5%/100 mmHg, 4.7% ± 1.3%/100 mmHg, and 10.3% ± 2.3%/100 mmHg, respectively. The data reported here fulfill the quality requirement of clinical use of this kind of biodegradable small diameter artificial vascular prosthesis.
