Abstract. China presently contributes the largest amount of anthropogenic mercury (Hg) emission into the atmosphere in the world. Over the past decade, numerous studies have been conducted to characterize the concentration and forms of atmospheric Hg in China, which provide insights into the spatial and temporal distributions of atmospheric Hg through ground-based measurements at widely diverse geographical locations and during cruise and flight campaigns. In this paper, we present a comprehensive review of the state of understanding in atmospheric Hg in China. Gaseous elemental mercury (GEM) and particulate-bound mercury (PBM) measured at the remote sites in China are substantially elevated compared to the background values in the Northern Hemisphere. In Chinese urban areas, the highly elevated GEM, PBM and gaseous oxidized mercury (GOM) were mainly derived from local anthropogenic Hg emissions, whereas regional anthropogenic emissions and long-range transport from domestic source regions are the primary causes of the elevated GEM and PBM concentrations at remote sites. Using 7-9 years of continuous observations at a remote site and an urban site, a slight increase in atmospheric GEM (2.4-2.5 % yr −1 ) was identified (paired samples test: p < 0.01), which is in agreement with the increasing domestic anthropogenic emissions. Anthropogenic GEM emission quantity in China estimated through the observed GEM / CO concentration ratios ranged from 632 to 1138 t annually over the past decade, 2-3 times larger than published values using emission activity data. Modeling results and filed measurements show dry deposition is the predominant process for removing Hg from the atmosphere, 2.5-9.0 times larger than wet deposition, due to the elevated atmospheric GEM and PBM concentrations that facilitate dry deposition to terrestrial landscapes. Further studies to reconcile the observed and simulated Hg concentrations, to understand the impact of domestic emission reduction on Hg concentration and deposition and to delineate the role of Hg emission and deposition of China in the global Hg biogeochemical cycle, are needed.
Abstract. China is regarded as the largest contributor of mercury (Hg) to the global atmospheric Hg budget. However, concentration levels and depositions of atmospheric Hg in China are poorly known. Continuous measurements of atmospheric gaseous elemental mercury (GEM) were carried out from
We report on near-GeV electron beam generation from an all-optical cascaded laser wakefield accelerator (LWFA). Electron injection and acceleration are successfully separated and controlled in different LWFA stages by employing two gas cells filled with a He/O2 mixture and pure He gas, respectively. Electrons with a Maxwellian spectrum, generated from the first LWFA assisted by ionization-induced injection, were seeded into the second LWFA with a 3-mm-thick gas cell and accelerated to be a 0.8-GeV quasimonoenergetic electron beam, corresponding to an acceleration gradient of 187 GV/m. The demonstrated scheme paves the way towards the multi-GeV laser accelerators.
Heterochromatin is a tightly packed form of chromatin that is associated with DNA methylation and histone 3 lysine 9 methylation (H3K9me). Here, we identify an H3K9me2-binding protein, Agenet domain (AGD)-containing p1 (AGDP1), in Arabidopsis thaliana. Here we find that AGDP1 can specifically recognize the H3K9me2 mark by its three pairs of tandem AGDs. We determine the crystal structure of the Agenet domain 1 and 2 cassette (AGD12) of Raphanus sativus AGDP1 in complex with an H3K9me2 peptide. In the complex, the histone peptide adopts a unique helical conformation. AGD12 specifically recognizes the H3K4me0 and H3K9me2 marks by hydrogen bonding and hydrophobic interactions. In addition, we find that AGDP1 is required for transcriptional silencing, non-CG DNA methylation, and H3K9 dimethylation at some loci. ChIP-seq data show that AGDP1 preferentially occupies long transposons and is associated with heterochromatin marks. Our findings suggest that, as a heterochromatin-binding protein, AGDP1 links H3K9me2 to DNA methylation in heterochromatin regions.
Abstract. Total gaseous mercury (TGM) was continuously monitored at a remote site (CBS) in Mt. Changbai area, Northeastern China from 24 October 2008 to 31 October 2010. The overall mean TGM concentration was 1.60±0.51 ng m −3 , which is lower than those reported from remote sites in Eastern, Southwestern, and Western China, indicating a relatively lower regional anthropogenic mercury (Hg) emission intensity in Northeastern China. Measurements at a site in the vicinity (∼1.2 km) of CBS station from August 2005 to July 2006 showed a significantly higher mean TGM concentration of 3.58±1.78 ng m −3 . The divergent result was partially attributed to fluctuations in the relatively frequencies of surface winds during the two study periods and moreover an effect of local emission sources. The temporal variation of TGM at CBS was influenced by regional sources as well as long-range transported Hg. Regional sources frequently contributing to episodical high TGM concentrations were pin-pointed as a large iron mining district in Northern North Korea and two large power plants and urban areas to the southwest of the sampling site. Source areas in Beijing, Tianjin, southern Liaoning, Hebei, northwestern Shanxi, and northwestern Shandong were found to contribute to elevated TGM observations at CBS via longrange transport. Diurnal pattern of TGM at CBS was mainly controlled by regional sources, likely as well as intrusion of air masses from the free troposphere during summer season. There are no consistent seasonal pattern of TGM at CBS, and the monthly TGM variations showed links with the patterns of regional air movements and long-range transport.
DNA methylation is an epigenetic event involved in a variety array of processes that may be the foundation of genetic phenomena and diseases. DNA methyltransferase is a key enzyme for cytosine methylation in DNA, and can be divided into two functional families (Dnmt1 and Dnmt3) in mammals. All mammalian DNA methyltransferases are encoded by their own single gene, and consisted of catalytic and regulatory regions (except Dnmt2). Via interactions between functional domains in the regulatory or catalytic regions and other adaptors or cofactors, DNA methyltransferases can be localized at selective areas (specific DNA/nucleotide sequence) and linked to specific chromosome status (euchromatin/heterochromatin, various histone modification status). With assistance from UHRF1 and Dnmt3L or other factors in Dnmt1 and Dnmt3a/ Dnmt3b, mammalian DNA methyltransferases can be recruited, and then specifically bind to hemimethylated and unmethylated double-stranded DNA sequence to maintain and de novo setup patterns for DNA methylation. Complicated enzymatic steps catalyzed by DNA methyltransferases include methyl group transferred from cofactor Ado-Met to C5 position of the flipped-out cytosine in targeted DNA duplex. In the light of the fact that different DNA methyltransferases are divergent in both structures and functions, and use unique reprogrammed or distorted routines in development of diseases, design of new drugs targeting specific mammalian DNA methyltransferases or their adaptors in the control of key steps in either maintenance or de novo DNA methylation processes will contribute to individually treating diseases related to DNA methyltransferases.
Lightweight and high strength sheet metal such as aluminium alloy sheets are used in a variety of industrial applications. Due to the limited weldability of these materials, mechanical joining techniques such as clinching are of interest. The challenge in this regard is that conventional round clinch forming locally induces large plastic deformations which potentially cannot be accommodated by materials with limited ductility. Ductile damage models are used to predict the occurrence of bottom cracks during conventional round clinch forming of EN AW-6082 T6 sheet. It is shown that cracks in the bottom of a clinched joint and the final static strength of a single lap shear specimen can be numerically reproduced provided that the post-necking strain hardening behavior and damage behavior of the base material are properly identified. The observed bottom cracks did not have a detrimental effect on the static strength and fatigue life of single shear lap specimens. It is hypothesised that fatigue cracks in single shear lap tests initiate due to fretting.
