Das AusmaD an gewiinschtcr 1,4-Addition 5ndert sich mit der Natur der Addenden und ist von ciner Reihe experimentellcr Parameter abhhgig, wie etwa der Art des Cu(I)-Salzes, dcr Anwcsenheit komplexierender Liganden. dem Liisungsmittel sowie der Temperatur.a$-Ungesattigte Aminosaurederivate 1 lJ sind Bestandteile zahlreicher Naturstoffe2) und bilden dariiber hinaus wertvolle Zwischenprodukte in der Aminosauresynthese ' I. Interessant sind die hochreaktiven Schiff-Basen 2 solcher Dehydroaminosauren, die zusatzlich ein 2-Azabutadien-System enthalten. In der Pyridoxalphosphat-katalysierten Biosynthese einiger Aminosauren wird das N-(Pyridoxy1iden)dehydroalanin (3) als reaktive Zwischenstufe formuliert4); so erfolgt z. B. der Aufbau des Tryptophans in vivo durch Michael-Addition des Enamins Indol an 3.Samtliche Syntheseversuche von 3 scheiterten bisher an der extremen Labilitat dieser Verbindung 536J. Ein Ersatz des Pyridoxalrestes durch einfachere aromatische Substituenten sollte die Zuganglichkeit verbessern. Es konnten bisher jedoch nur P-substituierte Verbindungen 2 (R', R2 = Aryl oder Alkyl) dargestellt werden6-'OJ. Die unsubstituierte Dehydroalaninverbindung 6 wurde lediglich durch 'H-NMR-spektroskopische Untersuchungen lo) und durch Folgereaktionen *J'J~) nachgewiesen.
N‐(Arylmethylen)dehydroalaninester sind hochreaktive Verbindungen. Sie dimerisieren unter [4+2]‐Cycloaddition (Diels‐Alder‐Reaktion) zu rac‐3,4,5,6‐Tetrahydropyridin‐Derivaten 2, die unter geeigneten Bedingungen isoliert werden können. In Lösung tautomerisieren die Dimeren 2 zu Enaminen 3, die schnell intramolekular zum Bicyclus 4 reagieren. Diese 3,6‐Diazabicyclo‐[3.2.1]oct‐2‐en‐Derivate 4 werden diastereoselektiv gebildet. Ihre Struktur wurde durch Röntgenstrukturanalyse geklärt. Die bicyclischen Diaminodicarbonsäureester 4 stellen die stabilen Endprodukte der gesamten Reaktionsfolge dar.
