The structure and absolute configuration of Chaetocin, a metabolite of the fungus Chaetomium minzltum with antibacterial and cytostatic activity has been elucidated by chemical and X-ray methods.
ZUSAMMENFASSUNG 1235 4,5,6-Trimethoxy-N, N-dimethyl-tryptamin (XI) und zwei dem Mescalin nahestehende Verbindungen, fi-(3,4,5-Trimethoxyphenoxy)-athyl-amin (VI) und ,4,5-Trimethoxyphenoxy)-athyl-dimethylamin (III), wurden synthetisiert. Keine der drei Substanzen zeigte in vorlaufigen Humanversuchen psychotrope Aktivitat. Vor etwa 28 Jahren haben OXFORD et ~1 .~) aus dem Kulturfiltrat von Penicillium brefeldianum DODGE die Fulvinsaure (I) z, 3, isoliert. Der gleiche Pilz produziert auch Grise~fulvin~). Bei der Untersuchung eines Stammes (S 464) dieses Pilzes haben wir als Hauptprodukt Palitantin (11) 4, gefunden. In kleinerer Menge konnten wir das mit Palitantin eng verwandte Frequentin (111) 5, 6 , sowie zwei neue Stoffwechselprodukte isolieren, die wir Brefeldin A und Brefeldin B nennen. Fulvinsaure und Griseofulvin liessen sich bisher in keiner unserer Ziichtungen, die unter den verschiedensten Bedingungen durchgefiihrt wurden, nachweisen. Dagegen konnten wir Brefeldin A auch aus dem Kulturfiltrat eines weiteren Penicillium-Stammes (S 498) als Hauptmetabolit gewinnen. Als Nebenprodukt erhielten wir aus diesem Stamme noch 1-0-Stearylglycerin (cc-Monostearin)

Ces phCnomilnes seraient particulibrement intenses dans le cas du composC chloral.SbC1, (AT = -0,41), oh la complexation entrainerait une forte variation de l'anisotropie du groupe carbonyle, alors que le dCplacement Clectronique produit par l'addition de l'accepteur se porterait assez fortement sur l'atome d'hydrogkne.On remarquera dans le tableau 2 un parallClisme limit6 entre les variations des dkplacements chimiques et des frCquences dues B la prCsence de la liaison C=O, en ce sens que, pour un m&me accepteur AT et AOJ varient de faGon inverse. Mais le nombre restreint de coniposCs Ctudiks n'a pas permis d'ktablir une corrklation numCrique.Dans les spectrogrammes de RMN. enregistrds pour H,CO * SbCl,, nous n'avons observC qu'un seul pic, alors que l'angle C=O-A mesurC pour des composCs daddition de type semblable &ant toujours sensiblement diffkrent de z, on aurait pu s'attendre B deux valeurs tg pour le cas d'un composC de symbtrie C, , modille que nous avons adopt6 dans la note prCcCdente [l]. La rCsolution du spectrombtre utilisk Ctant limitCe B f 0,03 ppm, nous ne pouvons pas conclure d'une maniilre dkfinitive. Summary: Sordarin (C2,H4008), an antifungal mould metabolite of Sordaria uruneosu, has been degraded to the aglykon sordaricin (C2,H,,04) and G-desoxy-4-O-methyl-~-altrose, a hitherto unknown sugar. I , Isolierung und Charakterisierung. Aus Kulturfiltrat und Mycel von Sordaria araneosa Gain (Stamm S 2266) haben wir einen neuen Metaboliten mit antifungischer Aktivitat, den wir Sordarin (1) nennen, isoliert. Reinigung der rohen Extrakte durch Chromatographie an Kieselgel oder mittels Gegenstromverteilung lieferte amorphes, optisch aktives ( 1 . 1 2 = -45" in Methanol) Sordarin, das auf Grund von Analyse und thermoelektrischer Molekulargewichtsbestimmung, sowie von Massenspektren verschiedener Derivate die Bruttoformel C,,H,,,O,, aufweist. Sordarin (1) ist eine monobasische Saure (pK& = 7,6) (1R.-Spektrum: s. Fig. 1) ; ihr Kaliumsalz (2) laisst sich leicht aus Essigester-Methanol kristallisieren und schmilzt bei 253-255".

On the basis of X-ray crystal structure analysis, CD. measurements, and asymmetric synthesis the absolute configuration of brefeldin A has been determined.Some years ago we reported the isolation [l] and the elucidation of the constitution 121 of brefeldin A (1) (identical with ascotoxin [3], decumbin [4] and cyanein [5]). At that time the (S)-chirality of C15, the trans connection of the two rings and the trans relationship at the conjugated double bond had been established, but the stereochemistry of the other asymmetric centers and of the isolated double bond remained to be clarified.A recent publication by SuzMki et al.[3] prompts us to describe our own results concerning relative and absolute configuration of brefeldin A, which are not fully in agreement with those of the Japanese authors. . -; H 3 0~c 0 0 c H 3 10 "COOCH, 1 R = H X = H, OH 2 R = AC X = H, OH 3 R = AC X = 0 4The NMR. spectrum of brefeldin A (1) (SO(CD,)&DCI, 7:3, 100MHz) shows signals for the protons on the isolated double bond at 5.22 and 5.60ppm (tetramethylsilane = 0) with a coupling constant of 15.5 cps, which indicates their trans relationship. The method of Horeau [6] allowed1) to specify the chirality at C4 as ( S ) in brefeldin A (1) and ( R ) in tetrahydro brefeldin A [l]. l)The resulting a-phenyl butyric acid had a = -0.091O in the case of brefeldin 4 and M = +0.087" in the case of tetrahydro-brefeldin A. The optical yield in both cases was 23%. Owing to its symmetrical position the hydroxyl group in the five-membered ring appears to be without influence on the asymmetric synthesis.
