Acid and alkaline deoxyribonuclease (EC 3.1.4.6, EC 3.1.4.5) activities were demonstrated in the human seminal plasma. By means of a micro-disc-electrophoretic deoxyribonuclease test six different fractions are detectable. They were characterized with respect to their optimal substrate and optimal incubation conditions. The various enzyme activities were tested at pH 5.0 in 0.1 ᴍ Na-acetat buffer or at pH 7.4 in 0.1 ᴍ Tris-HCl buffer in the presence of different combinations of MgCl2, CaCl2, EDTA, NaSO4, NaF and mercaptoethanol with native or denatured herring sperm DNA as substrate
