A new kind of bio-inspired terpolymer was synthesized by a conventional free radical terpolymerization of dopamine methacrylamide (DMA), 2-(dimethylamino)-ethyl methacrylate (DMAEMA) and 2-methacryloyloxyethyl phosphorylcholine (MPC) with azobisisobutyronitrile (AIBN) as an initiator.
Suspended sediment dynamics are still imperfectly understood, especially in the loess hilly region on the Loess Plateau, with strong temporal variability, where few studies heretofore have been conducted. Using a dataset up to eight years long in the Lower Chabagou Creek, the variability in suspended sediment load at different temporal scales (within-flood variability, monthly-seasonal and annual) is analyzed in this paper. The results show that, on the within-flood scale, most of the sediment peaks lag behind peak discharges, implying that slope zones are the main sediment source area; independent of the occurring sequences of the peaks of sediment and discharge, all the events could present an anti-clockwise hysteresis loop resulting from the abundant material and the influence of hyperconcentrated flows on suspended sediment concentration. At monthly and seasonal scales, there is a 'store-release' process, i.e. sediment is prepared in winter, spring and late autumn, and exported in summer and early autumn. At the annual scale, the high variability in concentration and sediment yield are highly correlated with water yield, resulting from the number and magnitude of floods recorded yearly, and almost all the suspended load is transported during these events.
Runoff was measured from seven plots with different slopes nested in Tuanshangou catchment on the Loess Plateau to study effect of slopes on runoff in relation to rainfall regimes. Based on nine years of field observation and K-mean clusters, 84 rainfall events were grouped into three rainfall regimes. Rainfall regime A is the group of events with strong rainfall intensity, high frequency, and short duration. Rainfall regime C consists of events with low intensity, long duration, and infrequent occurrence. Rainfall regime B is the aggregation of events of medium intensity and medium duration, and less frequent occurrence. The following results were found: (1) Different from traditional studies, runoff coefficient neither decreased nor increased, but presented peak value on the slope surfaces; (2) For individual plot, runoff coefficients induced by rainfall regime A were the highest, and those induced by rainfall regime C were the lowest; Downslope, the runoff coefficients induced by three rainfall regimes presented the same changing trend, although the peak value induced by regime A occurred on a shorter slope length compared to those by regime B and C; (3) Scale effect on runoff induced by rainfall regime A was the least, and that induced by rainfall regime C was the largest. These results can be explained by the interactions of crusting, soil moisture content, slope length and gradient, and erosion units, etc., in the context of different rainfall regimes.
The bone morphogenetic protein receptor IB (BMPR-IB) was studied as a candidate gene for the prolificacy of sheep. Nine pairs of primers (P1-P9) were designed to detect single nucleotide polymorphisms (SNPs) of exons 1-4 and 6-10 of the BMPR-IB gene in both high (Small Tail Han and Hu sheep) and low prolificacy breeds (Texel and Chinese Merino sheep) by polymerase chain reaction (PCR)-single strand conformation polymorphism (SSCP). Only the products amplified by primers P2, P5, P6, P7, P8 and P9 displayed polymorphisms. The present study identified 22 SNPs in partial coding regions of ovine BMPR-IB, in which 20 SNPs were reported for the first time. In total of the 22 mutations, 18 DNA variations were originated from the Hu breed, three were found in the Small Tail Han breed (two of them were found in other sheep breeds), three in the Chinese Merino breed, and none in the Texel breed. These results preliminarily demonstrated that BMPR-IB is a major gene affecting the hyperprolificacy in Small Tail Han and Hu sheep, and could be used as a molecular genetic marker for early auxiliary selection for hyperprolificacy in sheep.
The maturation and developmental potential on cumulus-cell-free oocytes is of great importance theoretically and practically. The present study was to investigate the effects of l-ascorbic acid, alpha-tocopherol and co-culture on in vitro developmental potential of porcine denuded oocytes (DOs). Porcine DOs were cultured in maturation medium supplemented with vitamin C (0, 50, 100, 250, 500, 750 microM) and vitamin E (0, 10, 20, 50, 100, 250 microm), respectively. And they were also co-cultured with dispersed cumulus cells (group CCscoculture), intact cumulus cells oocyte complexes (COCs) (group COCscoculture), and COCs whose oocytes were removed (group OOXcoculture), respectively. After 44 h incubation, the maturation rates, cleavage rates and blastocyst rates after parthenogenetic activation in three experiments mentioned above were collected and analysed, respectively. L-Ascorbic acid promoted porcine DOs in vitro maturation and blastocyt development after parthenogenetic activation while alpha-tocopherol did not increase the in vitro maturation rates, but improved the blastocyst rate. None of the three co-culture manner promoted the in vitro maturation and the cleavage of porcine DOs after parthenogenetic activation, but all the co-culture manners improved the blastocyst rates. Both Vitamin C and E enhance the in vitro developmental potential of porcine DOs. Co-culture increases the developmental potential of porcine DOs.
The ongoing coronavirus disease (COVID-19) pandemic is caused by a new coronavirus (severe acute respiratory syndrome coronavirus type 2 (SARS-CoV-2)) first reported in Wuhan City, China. From there, it has been rapidly spreading to many cities inside and outside China. Nowadays, more than 110 million cases with deaths surpassing 2 million have been recorded worldwide, thus representing a major health and economic issues. Rapid development of a protective vaccine against COVID-19 is therefore of paramount importance. Here, we demonstrated that the recombinantly expressed receptor-binding domain (RBD) of the spike protein can be coupled to immunologically optimized virus-like particles derived from cucumber mosaic virus (CuMVTT). The RBD displayed CuMVTT bound to ACE2, the viral receptor, demonstrating proper folding of RBD. Furthermore, a highly repetitive display of the RBD on CuMVTT resulted in a vaccine candidate that induced high levels of specific antibodies in mice, which were able to block binding of the spike protein to ACE2 and potently neutralize SARS-CoV-2 virus in vitro.
The recently ermerging disease COVID-19 is caused by the new SARS-CoV-2 virus first detected in the city of Wuhan, China. From there it has been rapidly spreading inside and outside China. With initial death rates around 4%, COVID-19 patients at longer distances from Wuhan showed reduced mortality as was previously observed for the SARS coronavirus. However, the new coronavirus spreads more strongly, as it sheds long before onset of symptoms or may be transmitted by people without symptoms. Rapid development of a protective vaccine against COVID-19 is therefore of paramount importance. Here we demonstrate that recombinantly expressed receptor binding domain (RBD) of the spike protein homologous to SARS binds to ACE2, the viral receptor. Higly repetitive display of RBD on immunologically optimized virus-like particles derived from cucumber mosaic virus resulted in a vaccine candidate (RBD-CuMVTT) that induced high levels of specific antibodies in mice which were able to block binding of spike protein to ACE2 and potently neutralized the SARS-CoV-2 virus in vitro.
