This study was conducted to evaluate the dietary α‐tocopherol (vitamin E) requirement in juvenile sea cucumber, Apostichopus japonicus. Sea cucumbers averaging 1.48 ± 0.07 g (mean ± SD) were randomly distributed into 18 rectangular plastic tanks of 20 L capacity in a recirculating system (20 animals per tank). Six semi‐purified experimental diets with average protein and crude lipid levels (dry matter) of 29.7 ± 0.36% and 4.39 ± 0.23% (mean ± SD), respectively were formulated to contain 0 (E4), 15 (E12), 30 (E23), 60 (E44), 120 (E77) and 600 (E378) mg α‐tocopherol/kg diet, supplied as dl‐α‐tocopheryl acetate. Diets were analyzed for α‐tocopherol content by HPLC and the α‐tocopherol levels were 4.01, 12.4, 23.1, 44.3, 77.4 and 378 mg α‐tocopherol/kg diet for E4, E12, E23, E44, E77 and E378 diets, respectively. Casein and defatted fish meal were used as the protein sources in the diets while wheat flour was the carbohydrate source. Sea cucumbers were fed each of the six experimental diets in triplicate groups. At the end of the 14‐week feeding trial, weight gain (WG), specific growth rate (SGR) and feed efficiency (FE) of sea cucumbers fed on E23, E44, E77 and E378 diets were significantly (P < 0.05) higher than those of animals fed on E4 and E12 diets. However, there were no significant differences in WG, SGR and FE among sea cucumbers fed on E23, E44, E77 and E378 diets or among those fed on E4 and E12 diets. Survival of sea cucumbers fed on E44, E77 and E378 diets were significantly higher than those of animals fed on E4, E12 and E23 diets. However, there were no significant differences among sea cucumbers fed on E4, E12 and E23 diets or among those fed on E44 and E77 diets. Whole‐body vitamin E concentration increased with α‐tocopherol content of the diets. Broken line analysis of WG showed an optimum dietary α‐tocopherol requirement of 41 mg α‐tocopherol/kg diet in sea cucumber. These results indicated that the optimum dietary α‐tocopherol requirement in sea cucumber in the form of dl‐α‐tocopheryl acetate could be higher than 23.1 mg α‐tocopherol/kg diet but lower than 44 mg α‐tocopherol/kg diet.
Cytogenetic analyses of the river puffer, Takifugu obscurus, the tiger puffer, Takifugu rubripes and hybrids produced between female T. obscurus and male T. rubripes and their hybrid triploids (produced by cold shock treatment at 4°C) were performed.T. obscurus had 2n = 44 chromosomes and 1.84 AE 0.019 pg DNA/cell, T. rubripes had 2n = 44 and 2.64 AE 0.015, the hybrids had 2n = 44 and 2.15 AE 0.010 and the hybrid triploids had 3n = 66 and 3.22 AE 0.010. The erythrocyte values of the hybrids were more similar to those for T. obscurus, whereas the hepatocyte, midgut and proximal tubule kidney cell values of the hybrids fell down between those for the parental species (p < .05). The erythrocyte, proximal tubule, hepatocyte and midgut epithelial cell sizes for the hybrid triploids were 1.5-fold larger than those for the hybrids (p < .05). The thickness of retina and each layer in the hybrid triploids were 1.1-fold larger than those of the hybrids (p < .05) and did not differ significantly among T. obscurus, T. rubripes and the hybrids (p > .05); however, the hybrid triploids had fewer cell nucleus outer layers than the hybrids (p < .05). Gonad development in the hybrids and hybrid triploids was less matured than in T. obscurus and T. rubripes. The metaphase nucleolus organizer regions (NORs) and the gill cells of T. obscurus, T. rubripes and the hybrids contained two satellite telocentrics, whereas the hybrid triploids contained three satellite telocentrics.
A comparison of the growth, hematological values, fatty acids, and gonadal and growth hormonal changes of river puffer, Takifugu obscurus, tiger puffer, T. rubripes, their hybrids (river puffer × tiger puffer) and hybrid triploids was performed during 3 months of their early growth period. Several features were observed during these 3 months: hybrids showed the highest levels of specific growth rate, 1.48%; hybrid triploids showed the smallest change in viscera fat (P<0.05), but GSI was not significantly different among groups (P>0.05). Considering hematological parameters, hybrid triploids had increased mean corpuscular volume and mean corpuscular hemoglobin (P<0.05), but other parameters were not significantly different between groups (P>0.05). With respect to fatty acids, puffer fish, hybrids and hybrid triploids contained fatty acids such as SFAs, MUFAs, n-3 PUFAs and n-6 PUFAs. There were significantly different amounts of total fatty acids between groups (P<0.05), however, rates of changes in fatty acids did not differ significantly between groups (P>0.05). Gonadal hormone (estradiol and testosterone) changes in the river puffer and tiger puffer were significantly higher than that observed in hybrids and hybrid triploids. The hybrids and tiger puffers had higher amounts of growth hormone (thyroid stimulating hormone and thyroxine) than the hybrid triploids and river puffers (P<0.05).
For the 2 years of farming, at the indoor circulating aquaculture system, four kinds of skeletal deformities were found among 60 Far Eastern catfish, Silurus asotus. Deformities saw jawbone’s luxation, abnormality of upper lip and malocclusion. Spinal deformity was most fatal deformities with low weight and small length. Jawbone’s luxation had 1 maxilla and 2 mandibles. Abnormality of upper lip had just lip was back over. Malocclusion’s left maxilla and right maxilla were not balanced. This experiment was any deformities in this species through the deformity can grasp how it affects.
This experiment was conducted to evaluate the long term effects of dehulled soybean meal (DHSM) as fish meal (FM) replacer in the diet for olive flounder, Paralichthys olivaceus, for 42 wk at a commercial fish farm. The four experimental diets were MP (moist pellet based on raw fish), DHSM 0 (fish meal based dry pellet), DHSM 30 (30% fish meal was replaced by DHSM) and COMD (commercial dry pellet). Fish were fed one of the four diets containing 50% crude protein and 16.7 kJ available energy/kg diet for 42 wk. Fish averaging 3.20±0.04 g (mean±SD) were initially distributed to each tank as a group of 6,000 fish reared in a flow-through system. At the end of the experiment, average body weight of fish was 315±9.3 g (mean±SD). Weight gain (WG) and feed efficiency (FE) of fish fed MP, DHSM 0 and DHSM 30 showed no significant difference among the diet treatments. However, WG and FE of fish fed MP were significantly higher than those of fish fed COMD (p<0.05). Specific growth rate (SGR), protein efficiency ratio (PER) and survival of fish fed all diets were not significantly different among the dietary treatments. Therefore, these results indicate that DHSM could replace up to 30% white fish meal with amino acid supplementation in olive flounder diets for long-term feeding practice at the commercial farm.
