Amaç: Bu çalışmada, naso-pharyngeal myiasis ile enfekte bir insandan elde edilmiş bot sineği larvalarının identifikasyonu ve moleküler karakterizasyonu amaçlanmıştır. Yöntemler: Adana bölgesinde naso-pharyngeal myiasis enfeksiyonu olan 49 yaşındaki bir kadından elde edilen sekiz adet bot sineği larvası çalışmanın materyalini oluşturmuştur. Larvaların teşhis anahtarlarına göre morfolojik teşhisleri yapılmıştır. Larva örneklerinin genomik DNA ekstraktlarında barkod CO1 geni amplifiye edilmiştir. CO1 sekanslarında haplotip ve genetik farklılık analizleri yapılmış ve ilişkileri göstermek için filogenetik ağaç oluşturulmuştur. Bulgular: Morfolojik özelliklerine göre tüm bot sineği larvaları O. ovis'in ikinci dönem larvası olarak teşhis edilmiştir. İzolatların CO1 sekansları arasında polimorfizm belirlenmemiş olup bu sonuç O. ovis için yeni bir haplotipin varlığını ortaya çıkarmıştır. Çalışmada yeni karakterize edilen haplotip Bosna-Hersek, Hırvatistan, Brezilya ve İran'dan rapor edilmiş O. ovis haplotipleriyle ortalama %99,5 identiklik göstererek kümelenme göstermiştir. Oestridae alt aileleri arasındaki interspesifik genetik farklılıklar %19,8-%30,8 olarak belirlenmiştir. Sonuç: Bu çalışma Türkiye'de rastlantısal bir insan konakta belirlenen O. ovis larvaları üzerine CO1 barkod sekansları temelinde ilk moleküler karakterizasyon verilerini sağlamıştır.

In this study, it was aimed to determine the molecular prevalence and genotypes of Enterocytozoon in healthy cattle. Methods: Fecal samples were collected from 50 cattle in Sivas between October 2017 and March 2018 and genomic DNA (gDNA) isolations were performed. gDNA isolates were processed by Nested PCR specifically amplifying ITS rRNA gene region to identify E. bieneusi. ITS rRNA region of E. bieneusi positive isolates were sequenced for genotyping and phylogenetic analyzes. Obtained sequences were assembled with appropriative genetic software, then phylogenetic relationships were revealed. Results: According to Nested PCR analyses, 29 (19.3%) out of totally examined samples were found positive for E. bieneusi. As a result of the sequence analyses, five distinct genotypes were determined. The most frequent genotype ERUSS1 and the other ERUSS2-4 genotypes were characterized as close to each other, which was reported for the first time in the world. Two isolates were determined in N genotype that was reported from cattle in Germany and were more different from the other genotypes. Phylogenetic analysis revealed that all the genotypes characterized in the study belonged to the genogroup 2. Conclusion: First molecular epidemiological data on E. bieneusi in cattle from Turkey were obtained with this study.

Hysterothylacium fabri is commonly reported in various Mediterranean fish. However, there is no data on the mitochondrial ribosomal RNA (rrnS) gene sequence of H. fabri larvae in the Mediterranean Sea waters. Therefore, we aimed to reveal molecular characterization of H. fabri based on the rrnS gene sequence in the current study. Firstly, the fourth stage of H. fabri larvae were identified at parasitological examination from Mullus barbatus in the Mediterranean Sea. Morphologically identified H. fabri larvae herein were also supported by the sequencing of the nuclear ribosomal ITS gene regions. In the next step, the rrnS gene of these larvae was molecularly analysed. Our H. fabri isolate (MK886659) showed 96.42% identity with H. fabri reported from China (MF140349) based on the rrnS gene. The nucleotide difference based on the rrnS gene between Chinese isolate (MF140349) and our H. fabri isolate (MK886659) was determined as 3.2%. Consequently, for the first time, the mitochondrial rrnS gene sequencing of H. fabri from the Mediterranean waters were performed in the current study.
