BackgroundInterferon-gamma is a key cytokine in the protective responses against intracellular pathogens. A single nucleotide polymorphism (SNP) located in the first intron of the human IFN-γ gene can putatively influence the secretion of cytokine with an impact on infection outcome as demonstrated for tuberculosis and other complex diseases. Our aim was to investigate the putative association of IFNG+874T/A SNP with American tegumentary leishmaniasis (ATL) and also the influence of this SNP in the secretion of IFN-γ in vitro.MethodsBrazilian ATL patients (78 cutaneous, CL, and 58 mucosal leishmaniasis, ML) and 609 healthy volunteers were evaluated. The genotype of +874 region in the IFN-γ gene was carried out by Amplification Refractory Mutational System (ARMS-PCR). Leishmania-induced IFN-γ production on peripheral blood mononuclear cell (PBMC) culture supernatants was assessed by ELISA.ResultsThere are no differences between +874T/A SNP frequency in cases and controls or in ML versus CL patients. Cutaneous leishmaniasis cases exhibiting AA genotype produced lower levels of IFN-γ than TA/TT genotypes. In mucosal cases, high and low IFN-γ producers were clearly demonstrated but no differences in the cytokine production was observed among the IFNG +874T or A carriers.ConclusionOur results suggest that +874T/A polymorphism was not associated with either susceptibility or severity to leishmaniasis. Despite this, IFNG +874T/A SNP could be involved in the pathogenesis of leishmaniasis by influencing the amount of cytokine released by CL patients, although it could not prevent disease development. On the other hand, it is possible that in ML cases, other potential polymorphic regulatory genes such as TNF-α and IL-10 are also involved thus interfering with IFN-γ secretion.
O objetivo deste estudo foi identificar e analisar compreensões mobilizadas por estudantes sobre aspectos dos modelos científicos. Para isso, foram realizadas entrevistas semiestruturadas individuais contendo perguntas gerais — sem exemplos — e específicas — com exemplos, envolvendo temas de Biologia. O quadro teórico teve como base as categorias e os níveis de complexidade utilizados por Grünkorn et al. (2014) para a análise e atribuição de significação às respostas obtidas nas entrevistas. O corpus foi analisado e categorizado nos aspectos sobre a compreensão de modelos: natureza, multiplicidade, objetivos, mutabilidade e testagem dos modelos. Os resultados mostraram uma presença expressiva da ideia de modelo como sendo uma cópia do original, assim como a associação entre modelo e método científico, entre outras características das concepções dos participantes. Adicionalmente, foi possível notar que essas concepções variam de acordo com o tipo de pergunta (geral ou específica) e com o conteúdo dos exemplos (no caso das perguntas específicas). Com base nesses resultados, levantamos a hipótese de que essa variabilidade possa ser explicada à luz da Teoria dos Campos Conceituais de G. Vergnaud.
A biorremediação consiste na utilização de organismos vivos, plantas ou micro-organismos, com vistas a reduzir ou eliminar poluentes presentes no ambiente. Tal temática, assim, pode ser considerada de grande potencial para ensino de conteúdos e competências presentes dentro das Ciências da Natureza, na Educação Básica. Dentre as abordagens utilizadas em ensino com o objetivo de instigar os alunos, destaca-se a aprendizagem baseada em problemas. No presente estudo, essa metodologia foi associada aos jogos educacionais que também são apontados como ferramentas didáticas que trazem o lúdico para sala de aula. O presente trabalho discute a criação e o emprego de um simulador/jogo que emprega situações-problemas ambientais reais, assim como as ferramentas para que o estudante consiga solucioná-las. Sendo assim, o objetivo é oportunizar aos educandos o aprendizado de conceitos importantes sobre biorremediação e áreas correlatas; assim como o desenvolvimento de competências relacionadas a resoluções de situações-problemas pautados no emprego do método científico aplicado. Para isso foi conduzida uma etapa de validação do jogo em uma sequência didática com um grupo de 5 alunos com um protótipo do jogo com vistas a analisar os resultados preliminares relativos a viabilidade de sua utilização como ferramenta de ensino. Os resultados se mostraram satisfatórios, indicando que os participantes conseguiram adquirir competências importantes relativas a biorremediação e conseguiram mobilizar os conhecimentos necessários para sua aplicação prática.
