The authors have previously tested many chemical colour reactions for protein and amino-acids and demonstrated that plenty of such substances as an amino-acid containing an imidazol group or at least an analogical substance of it are contained in the subnuclear portion of the gastric surface cells, where the productin vacuoles are seen (Arch. hist. jap. 13, 1957). As this fact will play a great role in the determination of the true nature of productin, the authors researched here if the positive colour of the reactions was surely related to the productin vacuoles, and tested one of the most remarkable reactions on dogs' stomaches when the diet was administered and productin vacuoles were discharged from the surface cells.Adult and healthy dogs were accustomed to regular feeding. Then the dogs were used for the experiment as follows; 1. 24hrs. after the administration of the last diet one dog was killed, 2. then diet was administered and at periods of 1/2, 1, 2, 3, 6hrs. afterwards these dogs were killed respectively.To take samples, the dogs were narcotized by ether, the abdominal cavity was opened and the small pieces of the gastric wall were taken.
