RESUMO.-[Caracterização do ciclo estral de Galea spixii (Wagler, 1831).]Os Galea spixii habitam a vegetação semiá-rida da Caatinga, no Nordeste brasileiro. Eles são criados em cativeiro para realização de pesquisas relacionadas a biologia da reprodução. Sendo assim, o objetivo deste trabalho foi caracterizar o ciclo estral de G. spixii para obtenção de informações que melhorem o conhecimento do manejo reprodutivo da espécie em cativeiro. O ciclo estral foi monitorado por citologia esfoliativa vaginal em doze fêmeas adultas. Após a detecção de dois ciclos completos em cada animal, os mesmos foram eutanasiados. Em seguida foi realizado estudo histológico do epitélio vaginal com três fêmeas em cada fase do ciclo estral; cinco foram pareadas com machos para realização do grupo controle e outras três fêmeas foram utilizadas para monitorar a formação e ruptura da membrana de oclusão vaginal. Através de citologia esfoliativa vaginal, constatou-se predomínio de células superficiais em estro, células intermediárias grandes em proestro, células intermediárias pequenas e células parabasais com presença de neutrófilos em diestro e metaestro, respectivamente. O es- The Galea spixii inhabits semiarid vegetation of Caatinga in the Brazilian Northeast. They are bred in captivity for the development of researches on the biology of reproduction. Therefore, the aim of this study is characterize the estrous cycle of G. spixii, in order to provide information to a better knowledge of captive breeding of the species. The estrous cycle was monitored by vaginal exfoliative cytology in 12 adult females. After the detection of two complete cycles in each animal, the same were euthanized. Then, histological study of the vaginal epithelium, with three females in each phase of the estrous cycle was performed; five were paired with males for performing the control group for estrous cycle phases, and three other were used to monitor the formation and rupture of vaginal closure membrane. By vaginal exfoliative cytology, predominance of superficial cells in estrus, large intermediate cells in proestrus, intermediate and parabasal cells, with neutrophils, in diestrus and metestrus respectively was found. Estrus was detected by the presence of spermatozoa in the control group. By histology, greater proliferation of the vaginal epithelium in proestrus was observed. We conclude that the estrous cycle of G. spixii lasts 15.8 ± 1.4 days and that the vaginal closure membrane develops until complete occlusion of the vaginal ostium, breaking after few days. O preá do semiárido nordestino (Galea spixii Wagler, 1831) é um roedor pertencente à família Caviidae. Pouca literatura é encontrada sobre essa espécie em relação a sua morfologia e seu comportamento ambiental e reprodutivo. Com o objetivo de entender a morfologia geral, em foco, na inervação do membro pélvico dessa espécie, neste trabalho, foi explorado o nervo isquiático, o qual é o maior de todos os nervos do organismo. Foram utilizados 10 preás (cinco machos e cinco fêmeas) que vieram a óbito por causas naturais, oriundos do Centro de Multiplicação de Animais Silvestres da Universidade Federal Rural do Semiárido (CEMAS/UFERSA). Os animais foram fixados após o óbito em solução aquosa de formaldeído 10% e, após 48 horas de imersão nessa solução, foram dissecados para exposição do nervo isquiático. Dessa forma, os dados obtidos foram compilados em tabelas e expressos em desenhos esquemáticos e fotografias. Os pares de nervos isquiáticos originaram-se de raízes ventrais de L6L7S1 (70%) e de L7S1S2 (30%) e distribuíram-se pelos músculos glúteo profundo, bíceps femural, semitendinoso e semimembranoso.
The study investigated the gross and microscopic anatomy of the genital organs of 20 agoutis at different stages of the estrous cycle and four in the final trimester of pregnancy. Specimens were euthanized and their reproductive organs were fixed in a 4% paraformaldehyde or 2.5% glutaraldehyde solution and submitted to routine histological techniques for light and scanning electron microscopy. In the ovary, during the proestrus phase, we observed developing follicles and corpus luteum (CL) in regression; during estrus, there were Graafian follicles; during metestrus, there was a hemorrhagic corpus, whereas in diestrus, there was a mature CL. The uterus was partially double because the cervix was cranially septate but caudally, the septum disappeared, forming a single ostium that opened into the vagina. Changes occurred along the estrous cycle in the uterine and vaginal epithelia, that is, an increase in the uterine epithelium height accompanied by an increase of thickness of the vaginal epithelium during the follicular phase and a decrease of thickness of both epithelia during the luteal phase. The endometrial lining was composed of a simple cuboidal epithelium to simple columnar epithelium with basal nuclei. The vaginal mucosa consisted of epithelium that varied from nonkeratinized stratified squamous (luteal phase) to keratinized stratified squamous (follicular phase). The clitoris was external to the vagina. It presented two protruding lateral keratinized spicules and a centralized urethra, with no common parts between the urinary and genital tracts. Anatomical and histological changes were observed mainly in the cervix, vagina and spicules of the clitoris during the EC.
A cutia é um roedor, que se distribui geograficamente por quase toda América Latina, sendo apreciada pelo sabor da sua carne. Neste trabalho objetivou-se descrever a vascularização arterial da base do encéfalo de cutias, caracterizando o comportamento, origem e distribuição das artérias componentes. Foram utilizados 10 animais do Centro de Multiplicação de Animais Silvestres (CEMAS/UFERSA), pesquisa aprovada pela Comissão de Ética no Uso de Animais (CEUA/ UFERSA- n° 02/2010) e pelo SISBIO (N°32413-1). Após eutanásia, os animais foram incisados na cavidade torácica para injeção de Neoprene látex 650 corado na cor vermelha e posterior abertura da calota craniana. Os encéfalos foram extraídos do crânio para análise da sua face ventral e logo fixados em solução aquosa de formaldeído a 10%. A vascularização arterial do encéfalo da cutia apresenta dois componentes principais, o sistema carótico e o vértebro-basilar. O sistema carótico na cutia é responsável pela vascularização do cérebro anterior em quase sua totalidade. O sistema vértebro-basilar é responsável pela vascularização do cérebro posterior (medula oblonga, pirâmide, corpo trapezóide, o cerebelo e ponte e parte do terço caudal do cérebro anterior), através das artérias cerebrais caudais, originárias dos ramos terminais da artéria basilar. As principais artérias presentes na superfície do encéfalo incluem a basilar que é ímpar, e as pares: artérias vertebrais, cerebelar caudal, trigemial, cerebelar rostral, ramo terminal da basilar, cerebral caudal, ramo comunicante caudal da carótida cerebral, carótida cerebral, ramo comunicante rostral da carótida cerebral, corióidearostral, ramo medial do ramo comunicante rostral, oftálmica interna, cerebral média e artéria cerebral rostral.
ResumoOrigem do plexo lombossacral em Galea spixii (Wagler, 1831) (Rodentia, Caviidae). O preá silvestre é um roedor pertencente à família Caviidae presente na região semiárida do Nordeste do Brasil. Foram utilizados 20 preás que vieram a óbito por causas naturais, obtidos no Centro de Multiplicação de Animais Silvestres (CEMAS) da Universidade Federal Rural do Semi-Árido (UFERSA). Os animais foram ixados em solução aquosa de formaldeído a 10% e, após 48 h, foram dissecados para visualização do plexo. As relações topográicas do plexo lombossacral foram agrupadas em tabelas e, posteriormente, submetidas a análises estatísticas (teste do qui-quadrado com correção de Yates). O plexo lombossacral originou-se das raízes ventrais dos três últimos nervos lombares e dos três primeiros nervos sacrais, correspondendo a 65% dos animais estudados (L 5 L 6 L 7 S 1 S 2 S 3 ). Originam-se do plexo os nervos: femoral, obturatório, isquiático, glúteo cranial, glúteo caudal e pudendo. We used 20 cavies that had died of natural causes, obtained from the Reproduction Center of Wild Animals (CEMAS) of the Federal Rural University of the Semi-Arid (UFERSA). The animals were ixed in aqueous solution of 10% formaldehyde and, after 48 hours, they were dissected to visualize plexus. The topographic relations of the lumbosacral plexus were grouped into tables and then they underwent statistical analysis (chi-square test with Yates' correction). The lumbosacral plexus stemmed from the ventral roots of the last 3 lumbar nerves and the irst 3 sacral nerves, corresponding to 65% of the animals under study (L 5 L 6 L 7 S 1 S 2 S 3 ). These nerves stemmed from plexus: femoral, obturator, ischiatic, cranial gluteal, caudal gluteal, and pudendal.
The current study aimed to determine if characteristics observed in vaginal cytology during the estrous cycle of female SYT cavies corresponded with proliferation of the vaginal epithelium, characterized by proliferating cell nuclear antigen (PCNA) immunolocalization, and with follicular development at different phases of the estrous cycle. After determining estrous cycle phases by vaginal cytology, females were euthanized at metestrus, diestrus, proestrus, and estrus. Histological study of the vaginal epithelium and ovary were then performed. Immunohistochemistry for PCNA in vaginal tissue at each cycle phase was also performed. Superficial cornified cells and early post-ovulatory follicles were found at estrus. Few nuclei below the enucleate superficial cells were immunoreactive to PCNA. At metestrus, the vaginal epithelium underwent desquamation and lost the superficial cornified cells; basal and intermediate cells appeared, and the post-ovulatory follicle formed an early corpus luteum. No PCNA immunoreactivity was observed. At diestrus, the corpus luteum was developed, and the vaginal epithelium contained basal and intermediate cells. There was PCNA immunoreactivity in the cellular nucleus in the germinative stratum of the epithelium. Because of the growth and maturation of ovarian follicles, the vaginal epithelium suffered intense proliferation at proestrus. Vaginal cytology revealed large intermediate cells and nucleated and enucleated superficial cornified cells. In the ovary, mature follicles were present. More apparent immunoreactivity of PCNA in the germinative layer was found. In summary, we inferred that vaginal exfoliative findings matched the proliferation process of the vaginal epithelium. PCNA immunolocalization occurred as well as corresponding follicular development in the ovaries.
