Con el objetivo de determinar el pH óptimo al que cultivos celulares de cochinilla (Dactylopius coccus Costa) registran la máxima producción de ácido carmínico in vitro, se evaluaron varios niveles de pH (4.5, 5.0, 5.5, 6.0 y 6.5) y de fracciones de centrifugado (0, 2, 5 y 10 min) de células maceradas de embriones de cochinilla establecidos en medio nutritivo de Schneider. El contenido de ácido carmínico presente en cada muestra fue determinado mediante cromatografía líquida de alta resolución. La extracción del pigmento de las muestras se realizó con una solución de HCl 2M. Los límites de detección del pigmento fueron de 1.0 mg L-1 a 120.0 mg L-1. No se observó un efecto significativo del pH en el contenido de ácido carmínico ni en el número de líneas celulares, aunque el pH intermedio (5.5) tendió a dar mejores resultados.
Se determinó el efecto del tratamiento estacional con amitraz contra Varroa destructor sobre la población y reservas alimenticias de colonias de abejas melíferas durante las cuatro estaciones del año en el altiplano central de México. Se utilizaron 48 colonias con reinas hermanas, homogéneas en población, reservas de alimentos y niveles de Varroa. 12 colonias recibieron tratamiento en verano, 12 en invierno, 12 en verano e invierno y 12 no fueron tratadas. Los niveles de V. destructor en abejas adultas, en cría y la caída de ácaros en el piso de las colmenas se determinaron durante un año. También se evaluó la población de abejas, áreas de cría, miel, polen y peso de las colonias. Hubo diferencias en los niveles de Varroa entre tratamientos (P<0.05). Finalizando el experimento (primavera), el nivel de infestación en colonias tratadas en verano (602±114) y no tratadas (416±86) fueron superiores (P=0.0002) que en las tratadas en verano e invierno (109±50) o solo en invierno (100±42), entre las cuales no hubo diferencias. Sin embargo, no hubo efecto significativo de los tratamientos sobre la población, reservas de alimentos y peso de las colonias. El tratamiento invernal fue suficiente para controlar Varroa en colonias del altiplano central de México.
Recently, the commercial breeding of Dactylopius coccus in confined environments has been developed not to depend on seasonal production and ensure the supply chain. Previous studies have shown the technical viability of their breeding of i Opuntia in a repository also called "Nopaloteca". However, considering the genetic diversity of Opuntia in each region, it is necessary to evaluate alternative cultivars to maximize the efficiency of that production system. The evaluated cultivars were Esmeralda and Villanueva (Opuntia ficus-indica (L.) Mill.). Four height levels (m) within the “Nopaloteca” (N1: 0.5; N2: 1.0; N3: 1.5 and N4: 2.0) and two harvest indices (pre and post oviposition) were evaluated regarding their effects on the carminic acid concentration (CAC, %) and fresh weight (FW, g) of D. Coccus. The results showed no significant difference in the CAC of the D. coccus colonies in both cultivars (18 %); however, the highest FW was reached in the cv. Esmeralda with 6.3 g per cladode (p ≤ 0.05). The highest CAC was found in the N4 treatment (2.0 m) with 18.6 % (p ≤ 0.05); while the highest average FW values (p ≤ 0.05) were in the intermediate levels of the N2 (1.0 m) and N3 (1.5 m) treatments, with 6.4 and 6.1 g per cladode, each. The post oviposition phase harvest of D. coccus resulted in a higher CAC concentration (20.4 %); meanwhile, in the pre-oviposition phase, 15.6 % was harvested (p ≤ 0.05). Due to the productivity and quality obtained, the Esmeralda cultivar could be used as an alternative host for the intensive breeding of D. coccus in confinement, in north-central Mexico.
A nivel nacional e internacional, la cochinilla silvestre Dactylopius opuntiae es considerada el enemigo principal del nopal (Opuntia spp.)debido a los daños severos y a las pérdidas económicas que ocasiona. En otros países se han realizado estudios para conocer su biología, comportamiento y crecimiento poblacional; pero a pesar de su importancia socioeconómica, en México se han realizado pocas investigaciones sobre esta especie. El objetivo de este estudio fue estimar la supervivencia y reproducción de D. opuntiae en condiciones semicontroladas, a fin de generar información básica que permita implementar una estrategia de manejo y reducir su impacto nocivo. Para conocer el ciclo de vida y determinar los parámetros poblacionales de D. opuntiae se establecieron cohortes en cladodios individuales de Opuntia ficus-indicacv. Villanueva; éstas se monitorearon de agosto a diciembre del 2017. Durante su ciclo de desarrollo se registraron supervivencia (lx), proporción sexual ♀/♂ y fertilidad de hembras adultas (mx). Los resultados mostraron que la tasa neta de reproducción (Ro) fue de 26.09, expresada en el número de hembras recién nacidas por hembra; el tiempo de generación (T) fue de 63.61 d y la tasa intrínseca de crecimiento poblacional (rm) fue de 0.05 hembras∙d-1. Esta estrategia reproductiva y otras características permiten a D. opuntiae colonizar rápidamente la planta; debido a ello, la especie puede ser un fitófago de importancia económica en zonas donde el género Opuntiaes silvestre o cultivo.
