The objective of this study was to evaluate the hepatoprotective effect of the honey bee Apis mellifera ethanolic extract of the red propolis, obtained in four municipalities of the Rio Grande do Norte semi-arid region, through an in vitro evaluation of the antineoplastic potential in human hepatic carcinoma (HepG2) and normal cell lines (L929), and from the comet assay in hepatic cell lines (ZF-L hepatocytes) to evaluate the genoprotective potential of the extract. The hepatoprotective effect was also evaluated in vivo by the induction of chronic experimental hepatic lesions in rodents (Rattus norvegicus Berkenhout, 1769), Wistar line, by intraperitoneal administration of thioacetamide (TAA) at the dose of 0.2g/kg. The animals were distributed in the following experimental groups: G1 (control), G2 (treated with 500mg/kg ethanolic extract of propolis), G3 (treated with 500mg/kg of ethanolic extract and TAA) and G4 (treated with TAA). All rats were submitted to serum biochemical, macroscopic, histological and stereological biochemical exams of the liver. It was verified the genoprotective effect of red propolis since the mean damages promoted to DNA in cells tested with the extract were significantly lower than the mean of the positive control damage (hydrogen peroxide). The red propolis extract did not present cytotoxic activity to the tumor cells of human liver cancer, as well as to normal ones. The absence of cytotoxicity in normal cells may indicate safety in the use of the propolis extract. The results of the serum biochemical evaluation showed that the serum levels of the aminotransferase enzymes (AST) did not differ significantly between G1, G2 and G3 when compared to each other. G4 showed significant increase in levels compared to the other groups, indicating that the administration of the extract did not cause liver toxicity, as well as exerted hepatoprotective effect against the hepatic damage induced by TAA. The G3 and G4 animals developed cirrhosis, but in G3 the livers were characterized by the presence of small regenerative nodules and level with the surface of the organ, whereas in G4 the livers showed large regenerative nodules. The livers of the G1 and G2 animals presented normal histological appearance, whereas the livers of the G3 animals showed regenerative nodules surrounded by thin septa of connective tissue, and in G4 the regenerative nodules were surrounded by thick septa fibrous connective tissue. The analysis of the hepatic tissues by means of stereology showed that there was no statistical difference between the percentage of hepatocytes, sinusoids, and collagens in G1 and G2. In G3 the percentage of hepatocytes, sinusoids, and collagen did not differ significantly from the other groups. It was concluded that the ethanolic extract of the red propolis exerted a hepatoprotective effect, because it promoted in vitro reduction of the damage to the DNA of liver cells, antineoplastic activity in human hepatocellular carcinoma cell line (HepG2) and did not exert cytotoxic effect in normal cells or was able to reduce liver enzyme activity and the severity of cirrhosis induced by TAA in vivo.
Considering that neem (Azadirachta indica A. Juss.) causes deleterious effects on fertility, the objective was to evaluate the effect of feeding quail (Coturnix coturnix japonica Linnaeus, 1758) of 90 days old with neem seeds on seminal quality, biometric parameters, and testicular morphometrics, as an experimental model. After the 30 day adaptation period to the experimental conditions, birds were randomly distributed in five treatments of neem seeds incorporated into feed, with 10 replicates: T1 (0%), T2 (5%), T3 (10%), T4 (20%) and T5 (40%). After 60 days of receiving experimental rations, their semen was collected to evaluate the sperm quality, the birds were euthanized, and testes were submitted to biometric, histological, and histomorphometric evaluation. The data were evaluated by analysis of variance and later by Tukey test with a significance level of 5%. The mean values of testicle weight, length, and width significantly decreased in T4 and T5, due degeneration of germinal epithelial cells. In these treatments, it occurred a significant increase (P>0.05) in the average values of the tunica albuginea thickness and a significant reduction in the values of the seminiferous tubule diameter and the height of the germinal epithelium. In addition, the mean values of seminal volume, sperm concentration, sperm motility, vigor, and sperm viability were significantly reduced. Thus, the use of neem in bird feeding, as well as the use for therapeutic purposes, is questionable, considering its negative impact on reproduction.
Ocorrência de Ascaridia galli e Heterakis gallinarum em galinha-d'angola (Numida meleagris) no estado de Rondônia, BrasilOccurrence of Ascaridia galli and Heterakis gallinarum in Guinea Fowl (Numida meleagris) in the State of Rondônia, Brazil ABSTRACT Background: Parasitic diseases are among the problems that most affect birds. The Ascaridia spp. is one of the genera which is responsible for the main infections, and the Ascaridia. galli is the most frequent species found in birds. The Ascaridia spp. egg is oval and present a smooth shell which could be mistaken with the Heterakis gallinarum eggs, once they are the most frequent parasites in birds. Therefore, this case report describes the occurrence of Ascaridia galli and Heterakis gallinarum in a guinea fowl (Numida meleagris) feces and gut. Case: In January 2017, rain season in northern Brazil, a visit was performed to a free range avian breeding property in the municipality of São Miguel do Guaporé in the state of Rondônia, Brazilian Amazonian Region. The property breeds two chicken species: Gallus gallus domesticus and Numida meleagris as well as one quail species. The problem was related to the guinea fowl breeding which was composed by 50 adults and 45 young birds about two or three weeks old. All 45 young guinea fowl were presenting various clinical signs, which included: yellowish catarrhal diarrhea, isolation, loss of appetite, body weight, ruffled feathers, drooping wings and sternal prominence. All animals that presented the clinical signs as ruffled feathers, drooping wings and sternal prominence died on an average of 24 h and the mortality was of 65.9% (29/45). Subsequently, a three-week old bird corpse was necropsied, and it was observed that its intestines presented a large amount of endoparasites besides presenting thickening. As there were a lot of helminths, they were collected along with the intestinal content for examination. The parasitological examinations of the feces were performed by Willys Molay's technique. As a result of the fluctuation technique, were observed and identified large quantities of Heterakis gallinarum and Ascaridia galli eggs. The adult worms of H. gallinarum and A. galli were identified using dichotomous key. Discussion: Ascaridia galli and Heterakis gallinarum infection were reported in guinea fowl in a free range chicken breeding property in the municipality of São Miguel do Guaporé in the state of Rondônia, Brazilian Amazonian Region. Ascaridia galli and Heterakis gallinarum eggs have already been identified in free-range chickens (Gallus gallus domesticus) by flotation techniques, as Willys Mollay and Faust and they are the main species of helminths housed by chickens. However, these parasites are not commonly reported in guinea fowl worldwide, especially in the Numida meleagris species. Regarding necropsy, intestinal wall thickening is generally a common finding which has been reported in chickens, however it is not commonly reported in guinea fowl. Additionally, the presence of clinical signs of nematode parasitos...
The aim of this study was to determine the occurrence of tick-borne pathogens (Ehrlichia canis, Babesia vogeli, Hepatozoon spp. and Rickettsia spp.) in dogs in Vila de Jericoacoara, coastal region of Ceará, Brazil. Blood samples were collected from 153 animals and analyzed using molecular and serological methods. Sixty animals were found to be infected or exposed to at least one of the pathogens studied. Babesia vogeli was the most prevalent pathogen (15%), followed by E. canis (13.7%) and Hepatozoon spp. (11.8%), which was identified as Hepatozoon canis through sequencing. Twenty dogs (13%) were seroreactive to Rickettsia spp. Rhipicephalus sanguineus sensu lato was observed on 11.8% of the animals. There were associations between age (< 3 years old) and positivity for B. vogeli, and between habitation (stray dogs) and positivity for H. canis. There were also associations between anemia and infection with H. canis, and between leukopenia and exposure to Rickettsia spp. No association was detected between clinical alterations and infection with or exposure to the pathogens studied. The results confirmed that pathogens of veterinary importance are circulating in northeastern Brazil and showed that dogs are exposed to Rickettsia species with zoonotic potential, thus indicating a need for vector control measures.
O citoplasma Extramedullary plasmacytoma in a captive collared peccary (Pecari tajacu) An extramedullary plasmacytoma case in a captive collared peccary (Pecari tajacu) is reported. The animal, a female aging three years old, had a medical history of diffusely distributed skin and mucocutaneous junction lesions, associated with swollen lymph nodes. Clinical examination and complementary exams (complete blood count, biochemical analysis, skin scraping to search mites and fungal culture) were performed. Thirty days after examination, the animal died. At necropsy, multiple consistent nodules, aseptic pustules and swollen lymph nodes were found. On histological exams of the skin and oral mucosa, we observed a large number of round cells forming masses organized in nests, sheets, and cords of cells in a well-vascularized fibrovascular tissue. Neoplastic plasma cells infiltrated between the fibers and the lamina propria of smooth muscle. Spaces among the cell masses were filled with some eosinophil and fluid. Most of the cells were well differentiated, presenting a perinuclear clear zone. In some points, the cells were pleomorphic. The plasma cells presented eccentric, basophilic and spherical nuclei, showing a dense to organized chromatin with distinct nucleoli. Binucleate cells were observed, but multinucleated giant cells were rare. Oral mucosa and lymph nodes tested by immunohistochemical analyses were positive for Mb-1, with a multifocal distribution. In regard to Bcl-2, the neoplastic cells were intermittent weakly positive. So, an extramedullary plasmacytoma was diagnosed in the collared peccary considering the location, the histopathological and immunohistochemical findings. finamente granular de anfifílico para basofílico. As células foram interpretadas como plasmócitos neopl...
