Barriers to Hindered Rotation Around the N-Glycosidic Bond, IV ') 1-( P-D-Ribofuranosyl)isocyanuric AcidsThe 8-ribofuranosyl nucleosides 5 a -c and 6b, c are synthesized. The temperature dependency of the 'Hand I3C-NMR spectra can only be interpreted as hindered rotation around the Nglycosidic bond 'I, The Gibbs activation energies (43 -46 kJmoI ~ I ) are independent of acetylation of the OH groups of the furanose ring or restriction of its flexibility through formation of a 2 ' 3isopropylidene derivative, and are considerably lower than the corresponding data (about 69 kJmol-I) of the glucopyranosyl nucleosides 7b, c. The actibation entropies are small. The results are interpreted.
Barriers to Hindered Rotation Around the N-Glycosidic Bond, I N-GlucopyranosidesThe N-P-D-glucopyranosides 4a-c, 8c, d, 12a, c, 14a, c, and 15 are synthesized, the aglyca of which have a C,-axis along the glycosidic bond. Preparations of the N-glucosides 5a, b and 912, d are also described. From reasons of symmetry the temperature dependency of the 'H-and I3C NMR spectra can only be interpreted as hindered rotation around the glucosidic bond with a twofold rotational potential. Solvents, acylation of the OH-groups of the sugar moiety, intraor intermolecular hydrogen bonds as well as substituents of the aglycon remote from the neighbourhood of the glycosidic bond are of little influence on the parameters to activation of the hindered rotation. But glucosides with planar six-membered cyclic aglyca show higher rotational barriers (e. g. AG* = 69.12 kJ mol-' for 4b) than glucosides with a planar five-membered ring as aglycon (e.g. AG* = 53.4 kJ mol-' for 8d). The results are interpreted as repulsion between orbitals of the aglycon carbonyl oxygens vicinal to the glycosidic bond and ring oxygen 5'-0 and substituent 2'-0 of the sugar moiety.
