The inhibition of dihydropteroate synthase extracts by sulfones and sulfonamides has been determined. Structural dependency of the observed I50‐values has been quantitatively explained by multiple regression and principal component analysis. New highly active sulfones have been synthesized guided by the derived QSAR. The analysis leads to the conclusion that electronic and steric substituent effects are decisive and that sulfones and sulfonamides are binding to the same receptor site. The LFE‐descriptors used are correlated with the minimal conformational entropy. The results are also compared with the results obtained in cell‐free and whole cell systems of bacteria. The additional influence of cell‐wall permeability on inhibitory efficacy is demonstrated.
Aus N-Acetyl-D-glucosamin läßt sich kristallisiertes Chromogen III in einer Ausbeute von 40% d. Th. gewinnen. Dieselbe Verbindung wird, wie es die Theorie verlangt, auch aus N-Acetyl-D-galaktosamin erhalten. Es handelt sich um D(+)-5-Dihydroxyäthyi-3-acetamino-furan (I). Diese Konstitution wird gesichert durch eine Synthese der DL-Verbindung, die von Äpfelsäure (4 kg) ausgeht und 17 Reaktionsschritte erforderlich gemacht hat. Das synthet. Racemat stimmt mit dem Chromogen III, das wir aus einem künstlichen Gemisch von 50% D- und 50% L-Acetylglucosamin erhalten haben, genau überein. Aus dem Chromogen III wurde durch katalytische Hydrierung, Abspaltung der Acetylgruppe und Umsetzung mit Methyljodid ein quartäres Ammoniumsalz erhalten, das sich vom Muscarin durch Austausch der end-ständigen CH3-Gruppe gegen CH2OH unterscheidet; die sterischen Verhältnisse an 2 C-Atomen sind noch unbekannt
Ausgehend vom chromatographisch gereinigten Gemisch der Chromogene, die bei der Einwirkung von verd. Alkali auf NAcetyl‐D‐glucosamin entstehen und mit p‐Dimethylasmino‐benzaldehyd Rotfärbung geben, ließ sich eine schön kristallisierende optisch inaktive Substanz der Formel C6H7O2N gewinnen, welche die Farbreaktion mit Ehrlichs Aldehyd intensiv gibt. Es handelt sich, wie eine von Furancarbonsäure‐(3) ausgehende Synthese zeigt, um das noch unbekannte 3‐Acetamino‐furan.
Die einleitenden Abschnitte handeln von der Entdeckung des Glucosamins, von der Verbreitung der Aminozucker in der Natur und von Einblicken, die man in die Biosynthese dieser Verbindungen gewonnen hat. Am Beispiel der Blutgruppensubstanzen wird auf die Bedeutung für Fragen der Genetik hingewiesen. Im Anschluß daran wird über eigene Untersuchungen berichtet: Synthese von D-Glucosamin, D-Galaktosamin, N-Acetyl-lactosamin und von weiteren 2-Desoxy-2-aminozuckern durch katalytische Halbhydrierung von Aminonitrilen; Isolierung und Konstitutionsaufklärung von N-haltigen Oligosacchariden der Frauenmilch; Darstellung von D-Isoglucosamin durch katalytische Hydrierung von Amadori-Verbindungen; Isolierung der O-Acetyl-lactaminsäure-lactose aus Milch (Colostrum) und deren Hydrolyse durch Influenza-Virus sowie durch ein Enzym aus Cholera-Vibrionen; Abbau der D(-)-Lactaminsäure zu N-Acetyl-D-glucosamin; Bildung von 3-Acetaminofuran und verwandter Verbindungen aus 2-Acetaminozuckern
Während d-Isoglucosamin beim Erhitzen mit Diäthylamin das bereits auf anderem Wege hergestellte „Fructosazin” der Formel III liefert, erhält man durch Wasserabspaltung aus d-Glucosamin in heißem Eisessig ein um 1 Sauerstoffatom ärmeres „Desoxyfructosazin”. Für dieses wird durch hydrierend-hydrolytische Spaltung zu 1 Mol. d-Isoglucosamin und 1 Mol. 3-Desoxy-d-isoglucosamin die Struktur VII bewiesen
