Staphylococcus aureus é considerado o principal agente causador da mastite bovina em rebanhos leiteiros em todo o mundo. Afeta seriamente a produção e a saúde da glândula mamária. A persistência da infecção glandular mamária e a inflamação crônica da glândula mamária diminuem o potencial produtivo da vaca leiteira, além de constituir um risco sanitário para o rebanho leiteiro e para o homem devido à possível contaminação do leite com cepas resistentes a antibióticos e afetam a qualidade e a segurança de produtos lácteos frescos e não pasteurizados. O conhecimento da epidemiologia e do desenvolvimento da infecção glandular mamária permite a melhor aplicação de estratégias de prevenção e controle da mastite bovina.
<p><strong>Background</strong>. <em>Staphylococcus aureus</em> is considered the main causal agent of mastitis in dairy herds globally, affecting milk production and constituting a potential risk to public health. The presence of virulence factors such as biofilm formation and slime production favor colonization and persistence of the mammary gland infections. <strong>Objective</strong>. The study aims to characterize the phenotypes of slime-producing and biofilm-forming <em>S. aureus</em> associated with subclinical mastitis with different CMT (California Mastitis Test) degrees<strong> </strong>and their relationship with the persistence of infection. <strong>Methodology</strong>. To characterize the slime-producing and biofilm-forming <em>S. aureus</em> phenotypes, 116 individual milk samples were taken from cows with subclinical mastitis through the CMT, obtaining 80 <em>S. aureus</em> isolates. Slime production on Congo Red agar was determined <em>in vitro</em>, and the conformation of bacterial clusters adherent identified biofilm formation to rigid surfaces and crystal violet staining. <strong>Results</strong>. 68.8% (55) of the isolates produced slime, of which 20% (16) were considered as producers and 48.8% (39) as intermediate producers. 82.5% (66) of the total isolates were related to the formation of biofilm in different degrees; 1.25% (1) very high trainer, 41.25% (33) high trainer, and 40.0% (32) moderately trainer. Highly biofilm-forming isolates were associated with subclinical mastitis degrees 2 and 3 of the CMT. <strong>Implications</strong>. The verification of slime and biofilm formation in most of <em>S. aureus</em> isolations obtained from subclinical mastitis may imply that their presence is related to the condition's persistence and severity. <strong>Conclusion</strong>. The association between the phenotypes of slime-producing and biofilm-forming <em>S. aureus</em> was established with high degrees of the CMT and mammary infection persistence in the cows studied.</p>
A total of 269 cows in small family herds in the central region of México from different municipalities of México State were studied. Composed milk samples were obtained to detect subclinical mastitis and S. aureus infection and for characterization of phenotypes as follows: biotypes, capsular exopolysaccharide 5 and 8, ORSA/MRSA and MRSA strains; and in vitro phagocytosis neutrophil activity and apoptosis by S. aureus serotype 5. Results were evaluated by estimating proportions and chi-square test (p < 0.05). The microbial isolation rate was 46%; S. aureus isolation rate was 23.4-21.0% among cow herds; 39% of microbial isolates were in 1500-2500 cells/mL, with Wisconsin test. The phenotypes of S. aureus were: biotypes A and C are identified frequently that produce alpha and beta-hemolysin toxins, and a smaller proportion other hemolysins types. S. aureus isolates capsular serotypes 5 and 8 show differences in the in vitro neutrophil phagocytosis activity and apoptosis. The ORSA/MRSA isolates show that MRSA strains' mec A gene was confirmed by PCR. The S. aureus infection level in the dairy cow herds shows a wide municipal distribution, identifying different S. aureus pathotypes enclosed to virulence factors and MRSA to establish a potential health risk in small dairy cow herds in México.
Com o objetivo de evidenciar a importância do selênio (Se) na produção ovina, analisa-se a importância e essencialidade deste micronutriente, repercussões por sua deficiência e dados de estudos realizados nos quais se destaca a necessidade de sua aplicação na clínica e capturados em diferentes fases produtivas de ovinos. O Se é encontrado em estado puro ou associado ao enxofre, ferro e cádmio; é essencial para plantas e animais, previne miodistrofias em bovinos, diátese hemorrágica exsudativa em aves, alterações hepáticas em suínos; Intervém no crescimento, na fertilidade e é um agente terapêutico eficaz em doenças graves da glândula mamária. Juntamente com a vitamina E, mantém as funções das paredes celulares e membranas orgânicas; tem efeito positivo no ganho de peso e no aumento da fertilidade das ovelhas, no número de cordeiros nascidos, bem como no maior peso vivo ao nascer, quando são mantidos níveis adequados de Se na dieta ou pela aplicação de selenito de sódio . Pode ser responsável por alterar a resposta imune, em cordeiros que apresentam baixos níveis de Se e vitamina E no sangue, com baixa atividade de GSH-Px, apresentam diminuição dos níveis de imunoglobulinas, baixa atividade fagocitária e maior destruição de macrófagos. Em relatórios para avaliar a condição por bactérias causadoras de gabarro (Dichelobacter nodosus, bactéria que está associada a Fusobacterium necrophorum), em ovinos estabulados e em pastejo, foi demonstrado que a suplementação em doses acima do recomendado com Se-levedura a uma taxa de 24,5 mg Se/kg melhorou o crescimento em cordeiros e a saúde em ovelhas em comparação com os níveis máximos da FDA. Uma avaliação adicional da suplementação de Se é necessária para determinar se ela tem um efeito adicional na resposta imune inata e adaptativa e fornece proteção contra patógenos bacterianos ou virais.
No estado do México contribui com 16,3% da ovinocultura nacional, aqui as poucas fazendas leiteiras utilizam leite cru na elaboração de subprodutos de forma artesanal, o que pode ser um fator de risco para contaminação e para a saúde • público se a qualidade microbiológica do leite não for a ideal; Portanto, o objetivo deste estudo foi realizar a caracterização microbiológica de amostras de leite de um rebanho localizado no Vale do Toluca. De onde foram obtidas amostras de leite de 16 ovelhas na linha de produção, no período de julho a setembro deste ano. O teste da Califórnia foi realizado para determinar a presença de mastite subclínica devido a possível infecção da glândula mamária, amostras de leite foram coletadas para caracterização microbiológica em laboratório. Para a análise bacteriológica, as amostras foram inoculadas em placas com ágar sangue de carneiro para crescimento de bactérias em geral e ágar Mac Conkey para Gram negativo. As placas foram incubadas a 37ºC por 24 a 48 horas, as colônias isoladas foram identificadas por coloração de Gram, hemólise em ágar sangue, exames bioquímicos e sistema API. foram encontradas 17 amostras contaminadas, obtendo-se 20 isolados; dos quais 90% correspondiam a bactérias do gênero Staphylococcus; isolando quatro espécies Staphylococcus aureus 11 isolados correspondentes a 55%, chromogenes 4 isolados 20%, xylosus 2 isolados 10% e haemolyticus 1 isolado 5% e 10% de outros gêneros bacterianos. O estudo denota a importância de bactérias do gênero Staphylococcus sp. na saúde da glândula mamária e como patógenos que causam contaminação do leite de ovelhas de um rebanho leiteiro no Vale de Toluca.
