Proliferation of cells under hypoxia is facilitated by metabolic adaptation, mediated by the transcriptional activator Hypoxia Inducible Factor-1 (HIF-1). HIF-1α, the inducible subunit of HIF-1 is regulated by oxygen as well as by oxygen-independent mechanisms involving phosphorylation. We have previously shown that CK1δ phosphorylates HIF-1α in its N-terminus and reduces its affinity for its heterodimerization partner ARNT. To investigate the importance of this mechanism for cell proliferation under hypoxia, we visually monitored HIF-1α interactions within the cell nucleus using the in situ proximity ligation assay (PLA) and fluorescence recovery after photobleaching (FRAP). Both methods show that CK1δ-dependent modification of HIF-1α impairs the formation of a chromatin binding HIF-1 complex. This is confirmed by analyzing expression of lipin-1, a direct target of HIF-1 that mediates hypoxic neutral lipid accumulation. Inhibition of CK1δ increases lipid droplet formation and proliferation of both cancer and normal cells specifically under hypoxia and in an HIF-1α- and lipin-1-dependent manner. These data reveal a novel role for CK1δ in regulating lipid metabolism and, through it, cell adaptation to low oxygen conditions.
Colorectal cancer usually develops in clearly defined stages, with distinct molecular alterations characterizing each transition. This often slow process makes colorectal cancer an ideal target for early detection programs. The blossoming of global, -omics approaches in recent years has led to greatly increased expectations for novel diagnostic and prognostic tools. Despite many early disappointments and the resulting skepticism, real progress has been made with exciting new prospects for cancer research. This review summarizes currently available proteomic tools for identifying novel biomarkers and drug targets, as well as an overview of their application in research on molecular mechanisms of carcinogenesis. Emphasis is given to novel sample preparation methods, protein separation and identification techniques, and advanced mass spectrometry tools for quantitative proteomic. The most important applications of these technologies in colorectal cancer research are discussed.
Hemoglobinopathies affect patients in the wider Mediterranean area and consist of 4 distinct subgroups: beta thalassemia major (TM), beta thalassemia intermedia (TI), sickle cell disease syndromes (SCD) (homozygous SCD, SCD/beta thalassemia trait) and hemoglobinopathy H (alpha thalassemia). The clinical spectrum of these syndromes varies from mild to severe. Complex interactions between genes and environmental factors form the clinical manifestations in hemoglobinopathies. There is an unmet need to clarify these multifactorial mechanisms. This is the first Greek study, describing mutational alleles (variants in the HBB and HBA1/HBA2 genes, type of mutation and prevalence) in 217 patients with hemoglobinopathies of two large centers in Greece (Larissa and Athens) and associating particular genotypes or gene variants with clinical manifestations (transfusion frequency, complications). Thus, the complex interplay between corresponding genotypes and phenotypes was investigated. The present study results are in accordance with previous national studies with limited variations, due to regional prevalence of specific gene variants, as expected. The type and prevalence of variants in beta and alpha globin genes differ significantly among countries. In the beta thalassemic or SCD patients of our cohort, co-inheritance of variants in the alpha globin genes, leading to absence or reduction of alpha globin synthesis were associated with milder clinical course, whereas the inheritance of additional alpha genes (triplication) led to a more severe clinical phenotype. In cases in whom the genotype and phenotype did not correlate, other factors such as the function or modification of possible regulatory genes or additional nutritional or environmental effects should be investigated.
Οι μεταλλάξεις στα γονίδια RAS είναι από τις πιο συχνές γενετικές μεταβολές που παρατηρούνται στην ανάπτυξη και την πρόοδο του καρκίνου. Ειδικότερα στον καρκίνο του παχέος εντέρου, το ογκογονίδιο KRAS μεταλλάσσεται στα αρχικά στάδια της νόσου και αφού έχουν προηγηθεί μεταλλάξεις και απενεργοποίηση ογκοκατασταλτικών γονιδίων, όπως το APC. Οι RAS πρωτεΐνες ανήκουν στην οικογένεια των μικρών GTPασών και εναλλάσσονται μεταξύ μιας ανενεργής (που προσδένει το GDP) και μιας ενεργής (που προσδένει το GTP) μορφής. Οι μεταλλάξεις του γονιδίου οδηγούν συνήθως σε πρωτεΐνη που βρίσκεται συνεχώς στην ενεργή μορφή και ενεργοποιεί σηματοδοτικά μονοπάτια που ελέγχουν τον πολλαπλασιασμό, την απόπτωση και άλλες κυτταρικές λειτουργίες. Στην παρούσα μελέτη, αναζητήθηκαν οι αλλαγές που προκαλούν τα ογκογονίδια KRASV12 και HRASV12 στο πρωτεομικό προφίλ της Caco2, μιας κυτταρικής σειράς αδενοκαρκινώματος του παχέος εντέρου που δεν φέρει μεταλλάξεις στους γενετικούς τόπους των RAS. Η έκφραση των δύο ογκογονιδίων οδήγησε στη δημιουργία δύο νέων κυτταρικών σειρών, Caco2-KRASV12 και Caco2-HRASV12. Η σύγκριση των πρωτεομικών προφίλ των τριών κυτταρικών σειρών έγινε με ηλεκτροφόρηση δύο διαστάσεων και στη συνέχεια ταυτοποιήθηκαν περίπου 200 διαφορικά εκφραζόμενες πρωτεΐνες με διαδοχική φασματομετρία μάζας. Οι περισσότερες από τις πρωτεΐνες αυτές ήταν μοναδικές για κάθε ογκογονίδιο. Υπήρξε όμως και σημαντικός αριθμός πρωτεϊνών ρυθμιζόμενων με κοινό τρόπο από τα δύο ογκογονίδια. Στα Caco2-KRASV12 κύτταρα, διαπιστώθηκε ότι ο μετασχηματισμός από το ογκογονίδιο KRASV12 οδηγεί σε αύξηση των επιπέδων και της ενεργότητας της ενδογενούς HRAS πρωτεΐνης. Η γονιδιακή αποσιώπηση της τελευταίας προκάλεσε αναστροφή των πρωτεομικών αλλαγών που παρατηρήθηκαν στα Caco2-KRASV12 κύτταρα. Η διαπίστωση αυτή ενισχύει την υπόθεση ότι η μεταλλαγμένη και συνεχώς ενεργή KRAS λειτουργεί, τουλάχιστον εν μέρει, μέσω της ενεργοποίησης της 107 ΠΕΡΙΛΗΨΗ ενδογενούς HRAS. Με χρήση χημικών αναστολέων, δείχτηκε επιπλέον ότι το σηματοδοτικό μονοπάτι των MAP κινασών παίζει καθοριστικό ρόλο καθοδικά της ανωτέρω διαδικασίας. Τέλος, δείχτηκε ότι η γονιδιακή αποσιώπηση της HRAS στα Caco2-KRASV12 κύτταρα επαναφέρει την έκφραση κυτταροσκελετικών πρωτεϊνών και συσχετίζεται με τον περιορισμό των μεταστατικών ιδιοτήτων των κυττάρων αυτών. Τα αποτελέσματα αυτά αναδεικνύουν μια πιθανή συνέργια της ογκογόνου ΚRAS και της ενδογενούς HRAS στην καρκινογένεση του παχέος εντέρου
