Allophylus edulis (A.St.-Hil., Cambess. and A. Juss.) Radlk., commonly known as cocum, belongs to the Sapindaceae family. This species is of great medicinal interest, with studies showing that its fruits have antioxidant, anti-cholinesterase, and cytotoxic activity. In addition, it is used in traditional medicine as an antidiarrheal, anti-inflammatory and antihypertensive. The objective of this study was to perform somatic embryogenesis in vitro from leaf and root explants of Allophylus edulis, using different 6-benzylaminopurine (BAP) concentrations combined with naphthalene acetic acid (NAA). All treatments exhibited 100% callus formation, except for the treatment without supplementation of growth regulators. The calluses developed in treatments from leaf explants showed up to two colors (brown and brown/cream), and the highest fresh and dry mass was observed in the treatment with 0.5 mg L-1 of BAP with 0.1 mg L-1 of NAA. There was no shoot formation from the leaf explants. The callogenesis in treatments from root segments showed callus formation with up to three colors (brown, brown/cream, and cream/green), and the highest fresh and dry mass was obtained when cultivated with 2.0 mg L-1 of BAP combined with 0.1 mg L-1 of NAA. These auxin and cytokinin concentrations also showed a higher number of shoots. The interaction between auxin and cytokinin is recommended to obtain somatic embryogenesis in root segments and callus with morphological characteristics suitable for organogenesis. ********* In press - Online First. As espécies do gênero Paullinia são utilizadas na medicina popular por suas propriedades antipiréticas, antinevrálgicas e anti-diarréicas, e como analgésico similar à aspirina. A micropropagação vem sendo usada com sucesso na conservação de germoplasma e propagação clonal e massal de diversas espécies florestais. Neste aspecto, um grande desafio a ser superado é no estabelecimento in vitro, uma vez que para o cultivo é necessária obtenção de tecidos livres de contaminações provocadas por microorganismos. Uma das etapas mais importantes na micropropagação é a desinfestação superficial do material vegetal a ser utilizado, tal procedimento pode ser realizado utilizando álcool 70 % (v/v) e hipoclorito de sódio em diferentes concentrações, além de outros produtos desde que não prejudique o material a ser regenerado. A concentração da solução desinfetante e o tempo de exposição influencia significativamente no índice de contaminação in vitro, tornandose necessário o estabelecimento de um protocolo eficiente para cada espécie. Este trabalho teve como objetivo estabelecer um protocolo eficiente de descontaminação de explantes foliares de pananá. Foram testadas três concentrações de hipoclorito de sódio em diferentes períodos de tempo (1,0% nos tempos 27; 30 e 33 minutos; 1,5% nos tempos 19, 21 e 24 minutos e 2,5% nos tempos 10; 13 e 16 minutos) combinada com álcool 70% (v/v) com tempo de exposição de 2 minutos. Como resultados, observou-se que as concentrações assim como o tempo de exposição do hipoclorito de sódio se diferenciam entre si em relação ao índice de contaminação fúngica ao nível de 1% de probabilidade de erro. Ao utilizar cloro ativo a 1,0% com tempo de exposição de 33 minutos foi obtido 95,6% de explantes livres de contaminação. Podemos concluir que a concentração (1,0%) e o tempo de exposição (33 minutos) da solução desinfetante) influenciam significantemente na descontaminação do material vegetal para estabelecimento in vitro.
