Many quantitative trait loci (QTLs) have been identified by molecular genetic studies which control grain size by regulating grain width, length, and/or thickness. Grain width 2 (GW2) is one such QTL that codes for a RING-type E3 ubiquitin ligase and increases grain size by regulating grain width through ubiquitin-mediated degradation of unknown substrates. A natural variation (single-nucleotide polymorphism at the 346th position) in the functional domain-coding region of OsGW2 in japonica rice genotypes has been shown to cause an increase in grain width/weight in rice. However, this variation is absent in indica rice genotypes. In this study, we report that reduced expression of OsGW2 can alter grain size, even though natural sequence variation is not responsible for increased grain size in indica rice genotypes. OsGW2 shows high expression in seed development stages and the protein localizes to the nucleus and cytoplasm. Downregulation of OsGW2 by RNAi technology results in wider and heavier grains. Microscopic observation of grain morphology suggests that OsGW2 determines grain size by influencing both cell expansion and cell proliferation in spikelet hull. Using transcriptome analysis, upregulated genes related to grain size regulation have been identified among 1,426 differentially expressed genes in an OsGW2_RNAi transgenic line. These results reveal that OsGW2 is a negative regulator of grain size in indica rice and affects both cell number and cell size in spikelet hull.

Paralysis is the loss of the ability of one or more muscles to move, due to disruption of signaling between the nervous system and muscles. The most common causes of paralysis are stroke, head injury, spinal cord injury (SCI) and multiple sclerosis. The search for cure of paralysis is yet to be found. Many ethnobotanical surveys have reported the use of medicinal plants by various ethnic communities in treating and curing paralysis. The present review discusses the use of medicinal plants in India for ameliorating and curing paralytic conditions, as well as discuses some of the important developments in future possible applications of medicinal plants in treatment of paralysis. This review reports the use of 37 medicinal plants for their application and cure of ailments related to paralysis. Out of the 37 plants documented, 11 plants have been reported for their ability to cure paralysis. However, the information on the documented plants were mostly found to be inadequate, requiring proper authentication with respect to their specificity, dosage, contradictions etc. It is found that despite the claims presented in many ethnobotanical surveys, the laboratory analysis of these plants remain untouched. It is believed that with deeper intervention on analysis of bioactive compounds present in these plants used by ethic traditional healers for paralysis, many potential therapeutic compounds can be isolated for this particular ailment in the near future.

Climate change impacts crop production through the imposition of different abiotic and biotic stresses, and by altering the prevalence of pests and diseases. The wild relatives of crop plants exhibit enhanced tolerance to environmental stresses due to reduced severity of selection of agriculturally important traits. Wild bananas represent a largely untapped wealth of genetic diversity. Although some wild relatives of the banana crop have been screened for their tolerance to different biotic and abiotic stresses, many remain to be explored. The wild bananas show more hardiness and higher resilience to different stressors in comparison to their cultivated counterparts. They have been harnessed in banana improvement programmes to enhance stress tolerance and productivity. To utilise wild bananas for crop improvement, they need to be readily available to breeders. This warrants devising conservation strategies and the development and maintenance of centres from which different accessions can be procured. In this article, we have discussed some important biotic and abiotic stresses including banana wilt disease, Black Sigatoka disease, viral diseases, salt stress and drought stress where wild bananas are used for imparting tolerance. The conservation of wild bananas, related challenges and contemporary limitations related to their use for crop improvement has also been outlined. Bananas being most important food crop in the world and generally understudied, here, we present a comprehensive review of the use of wild relatives of banana and their related germplasm for the improvement of biotic and abiotic stress tolerance.
