Estuaries are subjected to continual environmental impacts from activities in the catchment areas. This research assessed the quality of two estuarine habitats located in Ilha do Maranhão, Brazil, through histological and genotoxic biomarkers in Centropomus undecimalis, comparing the data obtained to metal, physical, and chemical concentrations of water samples. The gill histological alterations were analyzed by the histological alteration index and genotoxic lesions in erythrocytes were detected by the Micronucleus Test. The analysis of metals revealed that all water samples contained at least two elements with concentrations higher than that allowed by the current Brazilian law. For gill histological analysis, snook of both areas assessed exhibited moderate lesions, indicating that the local fish are affected by environmental stress. Micronucleus analysis of snook showed that the Bacanga river basin is the most affected. In addition to assessing the health of commercial fish populations, the information about the biomarkers used for the species can serve to contribute to the preparation and/or application of health assessment models and implementation of environmental recovery policies for coastal aquatic environments.
Esta pesquisa adaptou a técnica do Ensaio Cometa na espécie Centropomus undecimalis, aplicando-a na avaliação da genotoxicidade de dois ambientes estuarinos na Ilha do Maranhão, rio Bacanga e estuário do Pau Deitado. Os peixes foram coletados no período chuvoso de 2015 e o sangue foi retirado por punção branquial para a padronização. Os parâmetros físico-químicos da água foram mensurados no local. Para a avaliação dos danos do DNA, os nucleoides foram classificados em classes de 0 a 3, segundo o grau de danos. A padronização da técnica teve alguns desafios, sendo o principal a fixação da agarose após o banho na solução Lise. Com isso, o tempo de exposição na solução foi alterado, bem como o tempo de corrida eletroforética. Os parâmetros físico-químicos da água dos pontos amostrados apresentaram valores dentro do permito pela legislação nacional, exceto a concentração de oxigênio dissolvido em ambos os pontos. Foram observados danos de todas as classes no DNA de C. undecimalis de ambos os pontos, os peixes do rio Bacanga apresentaram maior média de DNA com nível alto de danos (Classe 3) e os peixes do estuário do Pau Deitado apresentaram maior média de DNA com nível médio de danos (Classe 2). O valor médio do Score de danos no DNA foi maior nos peixes do Rio Bacanga, indicando que esses animais estão expostos a substâncias genotóxicas que induzem alterações e lesões no DNA. A técnica do Ensaio Cometa em C. undecimalis foi adaptada com poucas alterações e os danos no DNA detectados pela técnica mostraram-se como metodologias eficazes na avaliação da qualidade ambiental dos estuários amostrados. Dessa forma, a metodologia padronizada pode subsidiar programas de biomonitoramento nas áreas. Além disso, é necessário expandir a diversidade de organismos estudados por essa técnica na região, para serem usados em estudos de biomonitoramento.
