La antracnosis, causada por Colletotrichum spp., es una de la enfermedades poscosecha más importantes en papaya. En este estudio, frutas de papaya, híbrido Pococí, recolectados en plantaciones ubicadas en el cantón de Guácimo, provincia de Limón, presentaron 7 síntomas de antracnosis, luego de un período de almacenamiento. De cada síntoma se obtuvieron aislados de Colletotrichum en medio de cultivo PDA (papa-dextrosa-agar). Los aislamientos se clasificaron en 3 grupos según la similitud de los síntomas y la morfología de la colonia. El análisis de la secuencia de la región espaciadora interna transcrita (ITS) del ADN ribosomal reveló que los aislamientos pertenecieron a 3 especies del género Colletotrichum: C. gloeosporioides sensu lato, C. magnum y C. capsici (C. truncatum). La identificación de las especies fue confirmada con imprimadores taxón específico para C. gloeosporioides e imprimadores especie-específicos para C. magnum. Las 3 especies identificadas mostraron ser patogénicas sobre frutas de papaya. Las especies C. magnum y C. capsici (C. truncatum) se registran por primera vez en Costa Rica, como agentes causales de la antracnosis en papaya.
Introducción. La antracnosis causada por Colletotrichum spp., es la principal enfermedad poscosecha de la papaya en todo el mundo. Diferentes países han identificado por técnicas moleculares nuevas especies del género asociadas a frutas tropicales. Objetivo. Identificar, mediante técnicas morfológicas y moleculares, las especies de Colletotrichum asociadas a la antracnosis en papaya en Costa Rica. Materiales y métodos. Durante el 2014 y 2015, se establecieron 129 aislamientos monospóricos de Colletotrichum spp. aislados a partir de lesiones de antracnosis en frutos recolectados en Parrita, San Carlos y Guácimo, Costa Rica. Todos los aislamientos se caracterizaron por morfología de colonia y PCR con imprimadores taxón especie específicos para las especies C. gloeosporioides s.l. y C. acutatum s.l., respectivamente; así como con imprimadores especie específicos para C. magnum y C. truncatum. La identificación de las especies se confirmó mediante análisis multilocus, con base en la secuencia parcial de los genes actina (ACT), β-tubulina (TUB2), gliceraldehído-3-fosfato deshidrogenasa (GAPDH), quitina sintasa 1 (CHS) y la región ITS. El análisis filogenético se basó en los criterios de máxima verosimilitud y análisis Bayesiano. Resultados. Seis especies: C. fructicola, C. tropicale, C. theobromicola del complejo C. gloeosporioides, C. simmondsii del complejo C. acutatum, C. truncatum y C. magnum s.l., se asociaron a la antracnosis en frutos de papaya. C. magnum s.l. fue la especie más frecuente. Temperaturas superiores a los 18 °C permitieron el desarrollo de cinco especies, C. theobromicola, C. fructicola, C. tropicale y C. magnum s.l., las cuales presentaron un crecimiento acelerado, mientras que C. simmondsii tuvo un crecimiento lento. Conclusión. El análisis multilocus permitió identificar a especies pertenecientes de los complejos C. gloeosporioides y C. acutatum. Este es el primer informe de C. fructicola, C. tropicale, C. theobromicola y C. simmondsii asociadas a la antracnosis en frutos de papaya en Costa Rica.
Introducción. El moho del pedúnculo de la piña es un complejo de microorganismos, que se compone por varios hongos, bacterias y levaduras, que afectan a la fruta en su etapa poscosecha. Objetivo. Caracterizar las levaduras asociadas al moho del pedúnculo de la piña en poscosecha, y evaluar el efecto de diferentes temperaturas sobre el crecimiento de las levaduras durante el almacenamiento. Materiales y métodos. Se realizó la caracterización morfológica de 24 aislamientos; posteriormente, se seleccionaron 9 aislamientos representativos para la caracterización molecular. Para evaluar el crecimiento de las levaduras a diferentes temperaturas, se colocaron suspensiones de estas en concentraciones de 10-6 unidades formadoras de colonias (UFC) sobre placas Petri con el medio PDA (papa-dextrosa-agar). Las placas se almacenaron en una cámara con temperaturas a 4°C, 8°C, 12°C, 16°C y 20°C, y se contó el número de colonias a los 2, 3, 4 y 5 días, de iniciado el experimento. Resultados. Para la descripción microscópica, las células presentaron formas redondas, ovaladas y cilíndricas. En la descripción macroscópica, las formas de las colonias de las levaduras fueron redondas y/o ovaladas. La consistencia de las colonias de las levaduras no pigmentadas fue mantecosa o cremosa, mientras que en las pigmentadas fueron mucoides. En la descripción molecular, las levaduras pigmentadas se clasificaron en las siguientes especies: Rhodotorula mucilaginosa, R. toruloides, R. diobovata, Sporidiobolus pararoseus y S. carnicolor, y las levaduras no pigmentadas en Candidata intermedia, Meyerozyma guilliermondii y M. caribbica. Para el ensayo de temperaturas, se observó que conforme la temperatura de almacenamiento disminuyó en 4°C, las colonias de las levaduras tardaron un día más en hacerse visibles. Conclusión. Se encontró similitud entre la morfología de los aislamientos, por lo que la caracterización molecular permitió identificar la presencia de especies con potencial de biocontroladores para los patógenos asociados al moho.
