Das Kreatinphosphat nimmt in der Muskelphysiologie eine zentrale Stellung ein. Auch in den neueren Arbeiten über die Totenstarre spielt es eine bedeutende Rolle, da der Kreatinphosphatgehalt des Muskels den zeitlichen Eintritt der Starre wesentlich mitzubestimmen scheint. In diesem Zusammenhang suchten wir nach einem möglichst schnellen, empfindlichen und spezifischen Verfahren zur Bestimmung des Kreatinphosphatgehaltes in biologischem Material.In der Literatur ist eine Reihe von Methoden zur Kreatinphosphat-Bestimmung angegeben, die jedoch meist sehr umständlich und wenig spezifisch sind. Dementsprechend unterscheiden sich die Angaben über den Kreatinphosphatgehalt der Muskeln je nach Art des angewandten Verfahrens weitgehend voneinander.Die älteren Analysenmethoden beruhen auf einer Bestimmung des Phosphats bzw. des Kreatins vor und nach Hydrolyse des Kreatinphosphats. So wurde meist die Differenz zwischen dem mit der Molybdänblau-Methode 1 "direkt" nachweisbaren Phosphat und dem durch Fällung 2 isolierten oder gesondert bestimmten 3 "wahren" anorganischen Phosphat erfaßt. 4 Bei der durch Molybdat katalysierten Hydrolyse werden neben dem Kreatinphosphat noch andere labile Phosphatverbindungen gespalten 5 ' 6 . This study showed that there is a relationship between acid phosphatase levels and life cycle stages in the nematode, Panagrellus silusiae. Ten different isozymes of acid phosphatase were separated electrophoretically. Relative activity peaked at different stages in the life cycle for the different isozymes. Later in the life cycle, there is a general decrease in the relative activity of acid phosphatase itself, while there is a concurrent increase in the number of isozymes present. At least 97% of the acid phosphatase in P. silusiac is soluble (unbound). Acid phosphatase appears to be present in large quantities in the entire gastrointestinal tract, the excretory canals, and the reproductive system of mature Panagrellus silusiae.
