Response surface optimization was applied for the extraction of antibacterial substances from Rhubarb (ASR) against aquatic pathogenic . Based on the experimental results of single factors, the optimal extraction conditions were determined by Box-Behnken design combined with response surface methodology with conditions: 100% ethanol as extraction solvent, liquid/material ratio of 29 mL/g and extraction temperature at 88 °C for 148 min. The factual value of inhibition zones can reach 21.31 ± 0.95 mm and is not different from the predicted value (21.74 mm), which showed that the response surface methodology applied to the extraction optimization of antibacterial substances from Rhubarb against. is feasible. Moreover, the yield of ASR was 30.29 ± 2.27%. Five compounds, namely, aloe-emodin, rhein, emodin, chrysophanol and physcion, were identified in ASR by comparing the HPLC chromatogram of the reference mixtures and the sample. Contents of the five compounds were 0.68 ± 0.02, 0.24 ± 0.05, 0.78 ± 0.07, 6.68 ± 0.97 and 0.58 ± 0.15%, respectively. The minimal inhibitory concentration (MIC) values of ASR, aloe-emodin, rhein, emodin, chrysophanol and physcion were 0.625, 0.125, 0.015,> 1, > 1, and > 1 mg/mL, respectively, which indicated that aloe-emodin and rhein are the main antibacterial compounds of Rhubarb.

Fifteen compounds, including two tetramic acid derivatives, penicillenol A1 (1) and penicillenol A2 (2), six polyphenols containing both phenolic bisabolane sesquiterpenoid and diphenyl ether units, expansols A-F (3-8), together with six phenolic bisabolane sesquiterpenoids (9-14) and diorcinol (15), were isolated from the fermentation broth of the marine-derived fungus ZSDS1-F11 isolated from the sponge Phakellia fusca Thiele collected in the Yongxing island of Xisha. Their structures were elucidated mainly by using extensive NMR spectroscopic and mass spectrometric analyses. Compounds 3-5, 7 and 8 showed potent COX-1 inhibitory activity with IC50 values of 5.3, 16.2, 30.2, 41.0 and 56.8 μM, respectively. Meanwhile, compounds 3-8 showed potent COX-2 inhibitory activity with IC50 values of 3.1, 5.6, 3.0, 5.1, 3.2 and 3.7 μM, respectively. In addition, compound 1 exhibited antituberculosis activity with 96.1% inhibition at concentration of 10 μM.

The aim of this study was to perform bioassay-guided fractionation and identification of active substances against the aquatic pathogen Vibrio anguillarum from the fungus Aspergillus tubingensis SZX-6 associated with the crab Portunus trituberculatus. Based on the antibacterial activity against V. anguillarum, three compounds were isolated and purified from the culture broth of strain SZX-6 by sequential column chromatography over silica gel, Sephadex LH-20 and RP-18. The isolated compounds were identified on the basis of extensive spectroscopic data analysis. They were methyl 6-acetyl-5,7,8-trihydroxy-4-methoxy-2-naphthoate (1), daidzein (2) and genistein (3). Compound 1 showed moderate antibacterial activity against V. anguillarum (minimum inhibitory concentration 0.063 mg/mL). The obtained results could serve as a basis for further research towards design of active antibacterial agents against aquatic pathogens.

The aim of this study was to investigate the extraction of antibacterial substances from Galla chinensis (ASGC) that act against the aquatic pathogen Vibrio harveyi, to identify the chemical composition and explore the mechanism of action. The optimal extraction process was as follows: anhydrous methanol, liquid/material ratio (mL/g) 26:1, extraction temperature 98 C and extraction time 157 min. The diameter of the inhibition zones (21.31 ± 0.51 mm) under the optimal conditions agreed with the predicted value by experimental rechecking. The yield of ASGC was 39.39% ± 0.69%, and two compounds including gallic acid (GA) and penta-O-galloyl-b-D-glucose (5GG) were identified in ASGC. The minimal inhibitory concentration (MIC) values of ASGC and GA were 0.50 and 0.25 mg/mL, respectively. Meanwhile, 5GG and other chemical constituents did not exert antibacterial activities against V. harveyi (MICs > 1 mg/mL). Further investigation of the mechanisms of action of ASGC and GA against V. harveyi revealed their strong destructive effects on the growth of bacterial cells, the integrity and permeability of the cell membrane, and morphological alterations. The report provides a favorable prospect of GA as a potential control agent against V. harveyi in aquaculture.

A novel sesquiterpenoid was isolated from the whole plant of Ecdysanthera rosea Hook. et Arn. Its structure was elucidated as tricyclo [5,3,1,0 2,4 ]-3-aldehyde-3,11-dimethyl-8-methanoic acid on the basis of spectral evidence (1D and 2D NMR) and was named ecdysanthblic acid. Ecdysanthera rosea Hook. et Arn. is distributed in Xishuangbanna, Yunnan province, P. R. China. It was a traditional herb for the Dai nationality [1] and has been used to treat garget and rheumatism; the chemical constituents of this plant have been reported; C 21 steroids [2] and triterpenoid [3] were isolated. The present work describes the isolation and structural elucidation of a novel sesquiterpenoid (1) from this plant, designated as tricyclo[5,3,1,0 2,4 ]-3-aldehyde-3,11-dimethyl-8-methanoic acid. The plants were collected from Xishuangbanna, Yunnan province, P. R. China, in August 2005. Compound 1 was obtained as a white powder; positive FABMS showed a molecular ion peak at m/z 251 [M+H] + , and the HRESIMS of 1 gave an [M+Na] + peak at m/z 273.1466; combine with analysis of its NMR data, it possesses the molecular formula of C 15 H 22 O 3 . The IR spectrum (KBr) showed the presence of carbonyl (2731, 2822, and 1701 cm −1 ), carboxy (1729 cm − 1 ), and tricycle (2990 cm −1 ). The 13 C NMR and DEPT exhibited 15 carbon signals for two methyls (δ C 7.4 and 15.7),four methylenes, seven methines, and two quaternary carbons, including one carbonyl group (δ C 203.0) and one carboxy group (δ C 177.5). The methyl (δ C 7.4) has a singlet signal (δ 1.17) in the 1 H NMR spectrum and an upfield shift in the 13 C NMR spectrum, suggesting that it is probably connected with a quaternary carbon and shielded by the carbonyl group, which means that C-13 (δ C 7.4) and C-12 (δ C 203.0) are both attached to C-3 (δ C 35.0), as proved by the correlation of C-3 in the HMBC spectrum; H-2 (δ H
