Correct structural assignment of small molecules and natural products is critical for drug discovery and organic chemistry. Anisotropy‐based NMR spectroscopy is a powerful tool for the structural assignment of organic molecules, but it relies on the utilization of a medium that disrupts the isotropic motion of molecules in organic solvents. Here, we establish a quantitative correlation between the atomic structure of the alignment medium, the molecular structure of the small molecule, and molecule‐specific anisotropic NMR parameters. The quantitative correlation uses an accurate three‐dimensional molecular alignment model that predicts residual dipolar couplings of small molecules aligned by poly(γ‐benzyl‐l‐glutamate). The technique facilitates reliable determination of the correct stereoisomer and enables unequivocal, rapid determination of complex molecular structures from extremely sparse NMR data.

Time is of the essence: The rotational motion of biomolecules depends on intra- and intermolecular interactions and thus on distinct functional states. A new method, called HYCUD accurately predicts rotational correlation times in complex dynamic systems. It gives insights into the motional behavior of multidomain proteins in their free form and in supramolecular complexes.

Die korrekte strukturelle Zuordnung von kleinen Molekülen und Naturstoffen ist für die Wirkstoffentdeckung und die organische Chemie von entscheidender Bedeutung. Die Anisotropie‐basierte NMR‐Spektroskopie ist ein leistungsfähiges Instrument zur strukturellen Zuordnung organischer Moleküle, beruht jedoch auf der Verwendung eines Mediums, das die isotrope Bewegung von Molekülen in organischen Lösungsmitteln stört. Hier stellen wir eine quantitative Korrelation zwischen der Atomstruktur des Ausrichtungsmediums, der Molekülstruktur des kleinen Moleküls und den molekülspezifischen, anisotropen NMR‐Parametern her. Die quantitative Korrelation verwendet ein dreidimensionales molekulares Ausrichtungsmodell, das dipolare Restkopplungen kleiner Moleküle vorhersagt, die durch Poly(γ‐benzyl‐l‐glutamat) ausgerichtet sind. Die Methode erleichtert die zuverlässige Bestimmung des richtigen Stereoisomers und ermöglicht die eindeutige und schnelle Bestimmung komplexer Molekülstrukturen aus sehr spärlichen NMR‐Daten.

Die Rotationsbewegung von Biomolekülen wird durch molekulare Wechselwirkungen und damit durch unterschiedliche funktionelle Zustände beeinflusst. Eine neue Methode mit dem Namen HYCUD ermöglicht die Vorhersage von Rotationskorrelationszeiten in sehr komplexen, dynamischen Systemen und gibt damit Einblicke in das Verhalten von Mehrdomänenproteinen in ihrer freien Form sowie in supramolekularen Komplexen (Bild: flexibler Komplex aus Heterochromatin Protein 1 und Mononukleosom).
