Mixotrophic cultivation is a potential approach to produce microalgal
biomass that can be used as raw materials for renewable biofuels and animal feed,
although using a suitable, cost-effective organic carbon source is crucial. Here, we
used a Box–Behnken design with three factors, the glucose and sodium acetate
concentrations, and the percentage of Bold’s basal medium (BBM), to evaluate the
effects of different carbon sources on biomass productivity and the protein and
lipid contents of Neochloris oleoabundans
(UTEX#1185). When grow at optimal levels of these factors, 100 % BBM plus
7.5 g L−1 each of glucose and sodium acetate, N. oleoabundans yielded
1.75 g L−1 of dry biomass, with 4.88 ± 0.09 % N,
24.01 ± 0.29–30.5 ± 0.38 % protein, and 34.4 % ± 0.81 lipids. A nuclear magnetic
resonance spectrum (1H-NMR) of a lipid extract showed
that the free fatty acid content was 11.25 %. Thus, combining glucose and sodium
acetate during the mixotrophic cultivation of N.
oleoabundans can yield greater amounts of biomass, proteins, and lipids
for biofuel production.
O principal objetivo deste estudo foi gerar tecnologias para a extração de óleo de pinhão manso, avaliando a quantidade e qualidade com diferentes métodos de extração. Para a extração do óleo foram utilizados dois tipos de extratores (fator A), prensa tipo expeller e prensa hidráulica, temperatura (fator B) frio (ambiente) e quente (80ºC) e diferentes níveis de descascado das sementes (fator C). Foi utilizado um delineamento inteiramente casualizado com arranjo fatorial AxBxC. As variáveis estudadas foram: quantidade e qualidade (acidez, teor de fósforo e umidade), do óleo bruto e filtrado, e a quantidade de torta. A prensa tipo expeller apresentou as maiores médias de desempenho de óleo bruto. A partir de três kg de sementes com 40% de descascado foi obtido 37,18% de óleo bruto. No índice de acidez, as menores médias foram observadas por prensa hidráulica, com 0% de cascas, com 4,11 mg KOH/g na acidez do óleo bruto e 5,17 mg KOH/g na acidez do óleo filtrado. Foram obtidos menores teores de fósforo, usando prensa hidráulica sem pré-aquecimento, com valores de 72,53 ppm no óleo bruto e 10,32 ppm no óleo filtrado. Foi alcançado maior teor de óleo com a prensa tipo expeller, mas de melhor qualidade na prensa hidráulica.Palavras Chave: bioenergia, biodiesel, teor de óleo.
Para conocer la diversidad genética del piñón (Jatropha curcas L.) existente en Ecuador, se realizó un estudio mediante el uso de marcadores moleculares tipo microsatélites. Fue analizada la variabilidad genética de la colección del Instituto Nacional de Investigaciones Agropecuarias (INIAP) conformada por 158 accesiones, de las cuales 151 fueron colectadas en seis provincias del Ecuador, 4 provinieron de Perú y 3 de Brasil. Como muestras para el estudio se tomaron 15 gramos de brotes tiernos de una planta de cada accesión y se mantuvieron en 50 gramos de sílica gel para su secamiento. Una vez secas, las muestras se maceraron con una solución de metabisulfito de sodio como antioxidante y se almacenaron a temperatura ambiente hasta la extracción del ADN genómico. De los 10 primers SSRs analizados, solo dos mostraron patrones polimórficos o informativos (J-26 y J-28), evidenciando la formación de dos grupos genéticos principales y dos subgrupos. Se determinó que la variabilidad entre genotipos multilocus fue del 13,92%, representada en 22 grupos de accesiones. Los genotipos 20 y 21 presentaron 44,30% y 15,82% de accesiones duplicadas.
