The purpose of this study was to investigate the possible molecular pathways through which ghrelin accelerates in vitro oocyte maturation. Bovine cumulus-oocyte complexes (COCs), after 18 or 24 h maturation in the absence or the presence of 800 pg ml-1 of acylated ghrelin were either assessed for nuclear maturation or underwent in vitro fertilization in standard media and putative zygotes were cultured in vitro for 8 days. In a subset of COCs the levels of phosphorylated Akt1 and ERK1/2 (MAPK1/3) were assessed at the 0th, 6th, 10th, 18th and 24th hours of in vitro maturation (IVM). At 18 and 24 h no difference existed in the proportion of matured oocytes in the ghrelin-treated group, while in the control group more (P < 0.05) matured oocyte were found at 24 h. Oocyte maturation for 24 h in the presence of ghrelin resulted in substantially reduced (P < 0.05) blastocyst yield(16.3%) in comparison with that obtained after 18 h (30.0%) or to both control groups (29.3% and 26.9%, for 18 and 24 h in maturation, respectively). Ghrelin-treated oocytes expressed lower Akt1 phosphorylation rate at the 10th hour of IVM, and higher ERK1/2 at the 6th and 10th hours of IVM compared with controls. In cumulus cells, at the 18th and 24th hours of IVM Akt1 phosphorylation rate was higher in ghrelin-treated oocytes. Our results imply that ghrelin acts in a different time-dependent manner on bovine oocytes and cumulus cells modulating Akt1 and ERK1/2 phosphorylation, which brings about acceleration of the oocyte maturation process.
Plasminogen activators/Plasmin system plays pivotal role in regulating reproductive functions of mammals. Here, we examined the effects of modification of in vitro fertilization medium (IVF medium) with the addition of tissue-type plasminogen activator (t-PA), on bovine embryo development and quality, assessed by quantification of expression of various genes related to metabolism, oxidation, implantation and apoptosis. In addition, plasminogen activator activity (PAA) and plasminogen activator inhibition (PAI) were measured in the spent media. After conventional IVM, 2016 cumulus-oocyte complexes (COCs) were divided into four groups with modified composition of the IVF medium containing t-PA and/or its inhibitor epsilon-aminocaproic acid (control, t-PA, t-PA+ε-ACA, ε-ACA). Presumptive zygotes were cultured for 8 days in synthetic oviductal fluid (SOF) medium; gene expression studies were carried out on morulae and blastocysts. t-PA alone significantly suppressed cleavage and blastocyst formation rates, but this effect was neutralized by the addition of ε-ACA. PAA in the treated group was significantly reduced by ε-ACA, but without total elimination. Significant differences were detected in the expression of genes related to apoptosis and/or cell cycle arrest (BAX, BCL2L1, KAT2B) between embryos produced in t-PA-modified media and controls, giving an overall notion that the inferior developmental competence of treated embryos may be attributed to apoptotic phenomena induced by t-PA. In conclusion, it appears that excessive t-PA content in the IVF media, suppresses blastocyst formation rate, possibly due to induction of apoptotic phenomena.
The effects of modification of the in vitro embryo culture media (IVC) with the addition of urokinase-type plasminogen activator (u-PA) on the yield and/or quality of bovine embryos were examined in two experiments. In Experiment 1, denuded embryos were cultured in semi-defined synthetic oviductal fluid (SOF) for seven days, while in Experiment 2 embryos were co-cultured with cumulus cell monolayer in a serum-containing SOF medium. Plasminogen activator activity (PAA) and plasminogen activator inhibition (PAI) were determined in all spent IVC media. At the activity used (5 IU/ml), u-PA had no effect either on in vitro embryo production rates or on embryo quality as revealed by gene expression analysis of 10 important mRNA transcripts related to apoptosis, oxidation, implantation and metabolism. PAA and PAI analysis indicated the need for wellbalanced plasminogen activators and inhibitors as a culture environment for embryo development. However, more research is needed to unveil the mechanism by which u-PA is involved in in vitro embryo production systems.
Στην παρούσα διατριβή μελετάται η επίδραση της τροποποίησης των in vitro υποστρωμάτων της γονιμοποίησης (IVF) και της καλλιέργειας των εμβρύων (IVC) με την προσθήκη του ενεργοποιού του πλασμινογόνου του ιστικού τύπου (t-PA) και του τύπου της ουροκινάσης (u-PA) αντίστοιχα, στα ποσοστά παραγωγής των βλαστοκύστεων, αλλά και στην ποιότητά τους, αναλύοντας την έκφραση μιας σειράς γονιδίων που συνδέονται με την ικανότητα ενός εμβρύου να επιβιώσει ή να διακόψει την ανάπτυξή του. Στα υποστρώματα της IVF και της IVC προσδιορίσθηκε η δραστηριότητα των ενεργοποιών του πλασμινογόνου (PAA) και των αδρανοποιών τους (PAI) με τη βοήθεια φασματοφωτομετρικής μεθόδου. Στο 1ο πείραμα χρησιμοποιήθηκαν 2016 συμπλέγματα ωαρίων-κυττάρων του ωοφόρου δίσκου (COCs), τα οποία χωρίστηκαν στην ομάδα των μαρτύρων [control], στην ομάδα της οποίας τα υποστρώματα τροποποιήθηκαν με προσθήκη t-PA, τελική δραστηριότητα 500 IU/ml [t-PA], στην ομάδα που εκτός από t-PA περιείχε και τον αναστολέα ε-αμινοκαπροϊκό οξύ (ε-ACA) [t-PA+ε-ACA], και στην ομάδα που περιείχε μόνο ε-ACA [ε-ACA]. Το 2ο πείραμα περιλαμβάνει δύο υποκατηγορίες πειραμάτων, όπου η επίδραση της προσθήκης του u-PA, σε τελική δραστηριότητα 5 IU/ml, μελετήθηκε σε απογυμνωμένα έμβρυα, τα οποία καλλιεργήθηκαν σε ημι-καθορισμένες συνθήκες (πείραμα 2.α, συνολικά 1631 πιθανά ζυγωτά) και σε έμβρυα, τα οποία καλλιεργήθηκαν σε μη-καθορισμένες συνθήκες και με την παρουσία μίας μονοστιβάδας κυττάρων του ωοφόρου δίσκου (πείραμα 2.β, συνολικά 629 πιθανά ζυγωτά). Η αξιολόγηση της ποιότητας των παραγόμενων βλαστοκύστεων ημέρας 7 έγινε με τη συγκριτική μελέτη της έκφρασης των γονιδίων PLAC8, AKR1B1, BIRC5 ή survivin, BBC3 ή PUMA, PΤGS-2 ή COX-2, BCL2L1, SLC2A5 ή GLUT-5, MnSOD, PLG και PLAUR, των οποίων η έκφραση μεταβάλλεται ανάλογα με την ποιότητα των εμβρύων. Για την γονιδιακή ανάλυση των μοριδίων ημέρας 3 και 4 ελέγχθηκαν τα γονίδια BAX, BCL2L1, PIPOX, G6PD, SLC2A5, MnSOD και KAT2B ή PCAF. Tα αποτελέσματα της παρούσας διατριβής δείχνουν ότι η προσθήκη εξωγενούς t-PA στο υπόστρωμα της IVF προκαλεί μείωση των ποσοστών αυλάκωσης και παραγωγής εμβρύων βοοειδών. Όταν η εξωγενής αυτή PAA μειώνεται με την προσθήκη του αναστολέα ε-ACA, τα αποτελέσματα της IVP επανέρχονται στα φυσιολογικά επίπεδα. Από την ανάλυση της έκφρασης των επιλεγμένων γονιδιακών μαρτύρων αποκαλύφθηκε ότι, η τροποποίηση με την προσθήκη εξωγενούς t-PA ενδεχομένως επηρεάζει αρνητικά και την ποιότητα των παραγόμενων πρώιμων εμβρύων, καθώς σε αυτά βρέθηκαν να επάγονται φαινόμενα, όπως η απόπτωση και η διακοπή του κυτταρικού κύκλου. Από την άλλη πλευρά, στο 2ο πείραμα, η τροποποίηση του υποστρώματος καλλιέργειας των εμβρύων με προσθήκη u-PA, δεν επηρέασε την εξωσωματική παραγωγή και την ποιότητα των εμβρύων βοοειδών, είτε αυτά καλλιεργήθηκαν απογυμνωμένα από τη στιβάδα των κυττάρων του ωοφόρου δίσκου και σε υπόστρωμα ημι-καθορισμένων συνθηκών, είτε σε συγκαλλιέργεια σε μονοστιβάδα κυττάρων του ωοφόρου δίσκου και παρουσία ορού. Μολονότι η προσθήκη των δύο ενεργοποιών του πλασμινογόνου στα υποστρώματα της IVP δεν ευνόησε τα ποσοστά παραγωγής των εμβρύων βοοειδών, καθώς και την ποιότητα αυτών, αλλά αντιθέτως τα δυσχέραινε στην πρώτη πειραματική διαδικασία, περισσότερα πειράματα απαιτούνται για τη διαλεύκανση του επακριβούς ρόλου του συστήματος στην IVP. Σε κάθε περίπτωση προκύπτει ότι, οι δυο ενεργοποιοί αποτελούν παράγοντες που συμμετέχουν στη γονιμοποίηση και στην ανάπτυξη των εμβρύων, καθιστώντας ιδιαίτερα ενδιαφέρουσα την αναζήτηση των πιθανών μηχανισμών δράσης και των αλληλεπιδράσεων τους κατά τη διάρκεια των παραπάνω διαδικασιών.
