Our results demonstrate that intravenous treatment is a superior method for reducing colon inflammation compared with intraperitoneal therapy.
The oxidative stress is a pathomechanism underlying the pathophysiology of colitis and MSC play an important role in preventing the impairment of antioxidants defenses in inflamed colon.
Glucocorticoids and immunosuppressive drugs are commonly used to treat inflammatory disorders, such as inflammatory bowel disease (IBD), and despite a few improvements, the remission of IBD is still difficult to maintain. Due to their immunomodulatory properties, mesenchymal stem cells (MSCs) have emerged as regulators of the immune response, and their viability and activation of their migratory properties are essential for successful cell therapy. However, little is known about the effects of immunosuppressant drugs used in IBD treatment on MSC behavior. The aim of this study was to evaluate MSC viability, nuclear morphometry, cell polarity, F-actin and focal adhesion kinase (FAK) distribution, and cell migratory properties in the presence of the immunosuppressive drugs azathioprine (AZA) and dexamethasone (DEX). After an initial characterization, MSCs were treated with DEX (10 μM) or AZA (1 μM) for 24 hrs or 7 days. Neither drug had an effect on cell viability or nuclear morphometry. However, AZA treatment induced a more elongated cell shape, while DEX was associated with a more rounded cell shape (P < 0.05) with a higher presence of ventral actin stress fibers (P < 0.05) and a decrease in protrusion stability. After 7 days of treatment, AZA improved the cell spatial trajectory (ST) and increased the migration speed (24.35%, P < 0.05, n = 4), while DEX impaired ST and migration speed after 24 hrs and 7 days of treatment (-28.69% and -25.37%, respectively; P < 0.05, n = 4). In conclusion, our data suggest that these immunosuppressive drugs each affect MSC morphology and migratory capacity differently, possibly impacting the success of cell therapy.
7 -INTRODUÇÃOAssim como outros grãos, o milho é constantemente exposto a possíveis contaminações fúngicas, as quais podem se iniciar ainda no campo, durante o seu desenvolvimento. Neste caso, a contaminação pode ocorrer pela presença de esporos e fragmentos de micélio presentes no solo, de restos de plantas e sementes, ou ainda serem transportados pelo vento, chuva ou insetos (MILLS, 1989).Os efeitos do crescimento fúngico incluem diminuição do poder de germinação, emboloramento visível, descoloração, odor desagradável, perda de matéria seca, aquecimento, mudanças químicas e nutricionais, perda de qualidade e produção de compostos tóxicos -as micotoxinas (POMERANZ, 1982). Esta contaminação pode fazer com que os grãos tornem-se impróprios para o consumo humano e animal, resultando em grandes perdas econômicas (PASTER & BULLERMAN, 1988). FUNGOS E FUMONISINAS NO PERÍODO PRÉ-COLHEITA DO MILHO RESUMOA contaminação fúngica e por micotoxinas no milho pode ocorrer antes mesmo da colheita, resultando em grandes perdas econômicas. Fusarium spp. é o principal contaminante do milho no campo, e muitas espécies deste gênero são responsáveis pela produção de fumonisinas. As fumonisinas são tóxicas para animais e humanos, estando envolvidas na doença da leucoencefalomalácea eqüina e associadas à síndrome de edema pulmonar em suínos e câncer de esôfago em humanos. O objetivo deste trabalho foi identificar os pontos críticos de contaminação fúngica e a produção de fumonisinas durante o período pré-colheita do milho. Os resultados mostraram um crescimento fúngico já na fase de grão farináceo, com nenhuma diferença significativa nas fases seguintes: grão farináceo duro e maturação fisiológica. Fungos não esporulados foram predominantes na fase de florescimento (100%) e na fase de grão farináceo (95%). Espécies saprófitas foram identificadas na fase de grão farináceo duro (23,25%). Fusarium spp. já foi evidente na fase de grão farináceo (5%), aumentando consideravelmente nas fases de grão farináceo duro (62,5%) e maturação fisiológica (90%). Todas as cepas de Fusarium spp. testadas mostraram-se potencialmente toxigênicas. Fumonisinas foram evidentes nas fases finais de desenvolvimento, aumentando consideravelmente da fase de grão farináceo duro (0,2 ppm) para a maturação fisiológica (2,5 ppm). Os autores sugerem atenção especial e adoção de medidas preventivas na fase de grão farináceo, onde Fusarium spp. começa a ser evidente. Palavras-chave: fungos, fumonisinas, pré-colheita. Cyclodextrin glycosyltransferase (CGTase; E.C. 2.4.1.19) is an industrially important enzyme, which is used to produce cyclodextrins (CDs). In this research, we report the use of experimental factorial design to find the best conditions of pH and temperature for CGTase production by Bacillus circulans var. alkalophilus. The optimized calculated values for the tested variables were, respectively, pH 9.7 and temperature 36 degrees C, with a CGTase activity of 615 U mL(-1). The CGTase production was further studied with the optimized process parameters on submerged cultivations (SC) and solid-state cultivations (SSC) using soybean industrial fibrous residue (SIFR). The maximum CGTase activity obtained on SC was 1,155 U mL(-1) under aerobic conditions. Cell growth and CGTase synthesis in SSC using SIFR as substrate was excellent, with CGTase activity of 32,776 U g ((SIFR))(-1). These results strongly support the use of SIFR for CGTase production since it is a non-expensive residue.
Lipoma arborescens is a benign tumor, but it may be a reactive process to other disorders, and its clinical, analytical, radiological and ultrasound presentation may be redundant to any synovial tumor. Despite the characteristic feature on magnetic resonance imaging (MRI), the correct differential diagnosis in atypical presentation, and the need for timely removal of the lesion to prevent joint damage, forces, ultimately, to invasive procedures. The clinical case reported here, fourth described in English language publications on the polyarticular form, also presented other specificities related to one of the swellings, in the knee. Because of its atypical location in the popliteal fossa, recurrent episodes of joint effusion, personal history of knee trauma, pulmonary tuberculosis, and family history of rheumatoid arthritis required particular attention. This process was hampered by the refusal of knee (and ankle) surgery by the patient. He accepted surgical removal of the swellings of the wrists, for aesthetical reasons, with pathologic confirmation of the diagnosis, and clinical success in that location. MRI of the knee showed the typical image of lipoma arborescens, but also other changes that compromise the prognosis.
Objetivos: O presente estudo teve como objetivo promover ações educativas e traçar o perfil do estilo de vida individual de um grupo de pacientes hipertensos ou diabéticos. Métodos: Caracteriza-se como descritivo-exploratório, por meio de pesquisa-ação, utilizando o instrumento Pentáculo do Bem-Estar de Nahas, com uma amostra de conveniência de nove indivíduos — pacientes de uma Unidade de Estratégia de Saúde da Família em Sorocaba, São Paulo — que participaram de um grupo para atividades educativas envolvendo dinâmicas direcionadas aos temas alimentação saudável e prática de atividade física. A coleta de dados aconteceu em encontros semanais durante os meses de agosto e setembro de 2016. Resultados: O grupo caracterizou-se, predominantemente, por indivíduos do sexo feminino, com idades de 43 a 65 anos, com Ensino Fundamental, aposentados e portadores de hipertensão arterial sistêmica. Os indivíduos foram categorizados em três grupos (satisfatório, moderadamente satisfatório e insatisfatório), que estão relacionados aos cinco componentes do pentáculo. Entre os fatores que dificultam a adesão ao tratamento e o controle da hipertensão e da diabetes evidenciam-se a falta de interesse na participação de grupos educativos, a baixa instrução e a desvalorização das informações e das atividades desenvolvidas no grupo. Conclusão: Constatamos que as informações obtidas no grupo, o apoio e a motivação dada pelos profissionais não foram suficientes para mudar o comportamento dos indivíduos para a adoção de hábitos saudáveis.
