In the period between December 2011 and March 2012, a total of three mycological expeditions were made to the La Paciencia Valley in South Eastern Tierra del Fuego. The obtained data represents the first major record of fungi (macromycetes) of the area. More than 140 species, representing at least 11% of the mycobiota described for the island, were collected and documented. A checklist of 49 identified species, belonging to 25 families, 11 orders and 3 classes of the kingdom Eumycota is presented in this article, including a species previously not described from Chile. The species names are presented along with their taxonomical and ecological information.
