High-sucrose and high-fat diets induce deregulation in the metabolism of lipids and carbohydrates. This study aimed to detect the initial consequences on lipogenesis, gluconeogenesis and insulin signaling in the livers of rodents fed high-fat and/or high-sucrose diets for a short period of time. Male mice received a standard chow (SC), high-fat (HF), high-sucrose (HSu) or high-fat, high-sucrose (HFHSu) diet for 4 weeks. At euthanasia, blood was collected and the liver was removed for histomorphometrical and molecular analysis. The HF, HSu and HFHSu groups presented glucose intolerance, hepatomegaly, liver steatosis and lipid profile alteration when compared to the SC group (p < 0.0005). Additionally, there was an elevation in protein levels involved in lipogenesis (SREBP-1c), gluconeogenesis (PEPCK and G6Pase) and insulin signaling (IRS-1 and Akt) in the livers from the experimental groups compared to the SC group (p < 0.0005). Thus, we conclude that a short-term HF and/or HSu diet promotes glucose intolerance and liver damage in adult male mice. Surprisingly, the short exposure to excess sucrose in the diet promoted glucose intolerance and liver damage even in the absence of an increase in body mass or changes in serum insulin, cholesterol and triacylglycerol levels.
The plasma filtration and formation of the urine is a very complex process necessary for the elimination of metabolites, toxins, and excessive water and electrolytes from the body. The initial process of urine formations is done by the glomerular filtration barrier inside the glomeruli. This specialized barrier consists of three layers, fenestrated endothelium, basement membrane, and podocytes, which ensure that water and small molecules pass through while cells and large molecules are retained. The glomerular filtration barrier is found with abnormal morphology in several diseases and is associated with renal malfunction; thus, it is interesting to study these structures in different experimental and clinical conditions. The normal glomerular barrier and its alterations in some conditions (hypertension, diabetes, and fetal programming) are discussed in this chapter. Furthermore, some methods for studying the glomerular filtration barrier by electron microscopy, both by qualitative and quantitative methods, are present.
RESUMO -As manifestações neurológicas associadas à deficiência de vitamina B12 incluem polineuropatia, mielopatia, demência e neuropatia óptica. O diagnóstico laboratorial é feito através da dosagem sérica de cianocobalamina ou homocisteína e da excreção urinária de ácido metilmalônico. No estudo anatomopatológico observa-se na microscopia a destruição da mielina e de axônios vistos na substância branca. A região mais comumente afetada é o cordão posterior cervical e/ou torácico. O acometimento da coluna lateral é raro, ocorrendo em casos graves e avançados. O tratamento consiste na reposição de vitamina B12 e a resposta depende da gravidade do quadro e do tempo transcorrido entre o inicio dos sintomas e inicio do tratamento. Relatamos o caso de um paciente que apresentou, como manifestação de deficiência de vitamina B12, mielite transversa. The diagnosis is established by vitamin B12, homocysteine and methylmalonic acid measurements. Myelin and axon destruction in the white matter of the spinal cord are observed. The posterior column of the cervical and thoracic level is the most common involved area. The involvement of the anterior column is restricted to advanced and relatively severe cases. Treatment is based on vitamin B12 injections, and the prognosis depends on the stage of vitamin deficiency and deterioration at treatment onset. We report a case with transverse myelitis due to vitamin B12 deficiency. This picture is relatively uncommon, however, we believe patients with transverse myelitis should have vitamin B12 studies as part of the diagnosis work up. Apresentamos o caso de um paciente com quadro de mielite transversa secundária a deficiência de vitamina B12. CASOHomem, 49 anos, com diabetes mellitus tipo 2 e hipotireoidismo decorrente de tireoidectomia por doença de Graves, com história familiar de anemia perniciosa. Iniciou sintomas neurológicos há um ano caracterizados por quadro progressivo de parestesias nos quatro membros, paraplegia flácida e arreflexa, anestesia completa abaixo de T8 e incontinência urinária e fecal.
Resumo: O programa de extensão universitária "consCiência na Ciência" do Instituto de Saúde de Nova Friburgo (ISNF) da Universidade Federal Fluminense (UFF) visa aproximar os estudantes do ensino médio à Universidade, assim como divulgar informação acadêmico-científica à população do município de Nova Friburgo e municípios vizinhos situados na região serrana do estado do Rio de Janeiro (RJ). O objetivo deste artigo é relatar esta estratégia de aproximação de estudantes do ensino médio da região serrana do RJ à universidade por meio de cursos de férias na temática Nutrição. Os cursos de férias "Alimentação e Nutrição" e "Nutrição e Boas Práticas de Manipulação de Alimentos” foram oferecidos nos anos de 2014 e 2015, respectivamente, no ISNF/UFF, em dois turnos, com carga horária de 3-4h, e 20-25 vagas/turno. A divulgação do curso foi feita por meio de folder, apresentação nas escolas e website. A inscrição foi realizada em formulário jotform disponibilizado no website do Programa. Todos os estudantes mostraram-se interessados e com vontade de ingressar no ensino superior, vendo no curso de férias uma oportunidade de vivenciar o ambiente da Universidade. Projetos envolvendo estudantes do ensino médio ou fundamental representam um elo primordial entre a universidade e a comunidade. É por meio de ações como o curso de férias, que a comunidade científica pode divulgar o conhecimento produzido na academia, e despertar nos jovens a vontade de ingressar na universidade. Palavras-chave: Ensino Médio, Extensão Universitária, Popularização da Ciência. “ConsCIÊNCIA na CIÊNCIA” Program: Promoting science in high school by a vacation course on nutritional education Abstract: The University Extension Program "Awareness in Science" is part of the Health Institute of Nova Friburgo (HINF) at Universidade Federal Fluminense (Federal Fluminense University - UFF). This initiative aims at narrowing the gap between high school students and the University, as well as to disseminate academic and scientific information to Nova Friburgo and its neighboring population, located in the mountainous region of Rio de Janeiro State. The objective of this paper is to offer an account of the strategy to approach high school students in the aforementioned region through vacation courses in Nutrition. The courses "Eating and Nutrition" and "Nutrition and Good Food Handling Practices" were offered throughout years of 2014 and 2015, respectively, at HINF/UFF, in two turns, with a workload of 3 to 4h, and 20 to 25 vacancies per turn. The initiative was promoted by the distribution of brochures, lectures at schools and on websites. The registration was made with JotForm form available on the Project’s website. All students were interested and willing to apply to higher education courses. These students saw the vacation course as an opportunity to experience the university environment. Projects involving high school and elementary school students represent a major link between the university and the community. By means of these actions, this vacation course can promote the knowledge produced in the university, and thus foster in the students the desire to go to College. El objetivo de este trabajo es dar a conocer la estrategia usada para promover ese acercamiento de estudiantes de la escuela secundaria de la región montañosa de RJ a la universidad por medio de cursos de vacaciones en Nutrición. Los cursos de vacaciones en "Alimentación y Nutrición" y "Nutrición y Buenas Prácticas de Manejo de los Alimentos" se ofrecieron en los años 2014 y 2015, respectivamente, en ISNF/UFF, en dos turnos, con una carga horaria de 3-4h, y 20-25 vacantes/turno. La divulgación del curso fue hecha por prospecto, presentación en las escuelas y el sitio web. La inscripción se llevó a cabo en formulario JotForm disponible en el sitio web del programa. Es por medio de acciones tales como el curso de vacaciones que la comunidad científica puede difundir el conocimiento producido en la academia, y despertar en los jóvenes el deseo de ir a la universidad. -Therapeutic occlusion of the internal carotid artery is the main option for the treatment of the symptomatic intracavernous internal carotid artery aneurysms, but the issue of the best way of doing the balloon test occlusion (BTO) regarding prediction of future ischemic events remains debatable. Single photon emission computerized tomography (SPECT) has been offered as one of the best option of monitoring regional cerebral blood flow (rCB.) during the BTO, where severe asymmetry is predictive of delayed ischemia. We describe a case of important SPECT asymmetry during BTO clinically negative and its complete reversal with carotid occlusion after extra-intracranial bypass with high flow safenous vein bypass between the cervical carotid artery and the middle cerebral artery.KEY WORDS: giant intracavernous aneurysm, cerebral revascularization, carotid artery occlusion, saphenous vein graft, SPECT.Reversão de importante assimetria ao SPECT após bypass venoso de alto fluxo extra-intracraniano em paciente submetido a oclusão terapêutica da carótida interna: relato de caso RESUMO -Oclusão terapêutica carotídea ainda é a principal escolha para o tratamento dos aneurismas sintomáticos da artéria carótida interna ao nível do seio cavernoso.Existem controvérsias em relação à obtenção de testes com maior valor preditivo em relação à complicações isquêmicas futuras durante a realização do teste de oclusão carotídeo por balão. A tomografia computorizada de emissão de fóton único (SPECT) tem sido defendida como uma opção para a melhor avaliação do fluxo sanguíneo regional cerebral, onde evidente assimetria é indicativa de isquemia tardia. Descrevemos um caso de importante assimetria ao SPECT durante o teste clinicamente negativo e sua reversão completa após anastomose extra-intracraniana com interposição de veia safena magna (alto fluxo) entre a artéria carótida cervical e a artéria cerebral média.
