ABSTRACT. The genus Theobroma found in the Amazon region is composed of 22 species, including Theobroma speciosum, better known as cacauí. These species are constantly threatened by forest fragmentation caused by human activities and require conservation strategies and management aimed at preserving them in their natural environments. The main objective of this study was to analyze the population structure and genetic diversity within and between natural populations of T. speciosum by using ISSR molecular markers to understand the population structure of the species. Four natural populations belonging to the Amazon rainforest (BAC, CRO, FLA, and PNA), located in the State of Mato Grosso, were selected. Amplification reactions were performed using 15 ISSR primers. The BAC population showed higher genetic diversity (H = 0.095 and I = 0.144) and higher percentage of polymorphism (28.71%). The populations showed an F ST value of 0.604, indicating marked genetic differentiation. The highest genetic variation was found between populations. Gene flow was low between populations, indicating genetic isolation between populations.
RESUMOO objetivo deste estudo foi avaliar 50 genótipos de teca com base em marcadores moleculares ISSR. Dentre os genótipos estudados, 12 eram árvores candidatas, 36 vizinhas às candidatas e 02 árvores consideradas superiores com base no fenótipo, segundo a avaliação dos produtores. Esses genótipos foram propagados via seminal e possuem de 10 a 12 anos em campo. Para tal estudo foram coletadas em campo folhas jovens e expandidas de cada genótipo. O DNA total foi extraído e as amplificações foram feitas via PCR com 12 primers ISSR, previamente selecionados. As amostras foram aplicadas em gel de agarose 1,5% e submetidas à corrida de eletroforese. Para visualização dos produtos amplificados, o gel foi corado com brometo de etídeo e fotodocumentados em transiluminador UV. Os dados foram analisados utilizando-se o programa POPGENE 1.31 e os parâmetros de diversidade foram estimados. As análises de dissimilaridade foram estimadas pelo Índice de Jaccard e a construção do dendrograma foi feita com base no método UPGMA. Paralelamente, foi realizada a análise fenotípica de dados morfológicos referentes às árvores candidatas pela análise multicategórica. Todas as análises foram realizadas com auxílio do Programa Genes. Os 12 primers amplificaram 56 fragmentos. Para o índice de conteúdos polimórfico, os iniciadores UBC 841, UBC 857 e UBC 807 proporcionaram maiores valores, 0,329, 0,327 e 0,303, respectivamente. A diversidade genética das árvores candidatas (H = 0,1601 e I = 0,2301) foi similar à das vizinhas (H = 0,1507; I = 0,2178). O dendrograma gerado pelo método UPGMA formou 14 grupos, enquanto que no agrupamento com base em Tocher, 15 grupos foram formados. Quanto à análise fenotípica, a variável que mais contribuiu para a diversidade foi a formação de catana. Esses resultados refletem que há variabilidade entre os genótipos. Nota-se, portanto, a necessidade da ampliação da variabilidade do material, através da introdução de genótipos de diferentes procedências e não relacionados geneticamente. Brazil nut is a native Amazon species of high commercial value classified as vulnerable in terms of extinction risk due to marked illegal-burning activity and agricultural-frontier expansion processes occurring in the region. This study was undertaken to analyze the genetic diversity within and between two natural Brazil-nut populations occurring spontaneously in the Southern Amazon region spaced 50 km apart, both of which were located in the municipality of Alta Floresta, northern Mato Grosso state, Brazil. These are rural areas and samples were from native forest patches. Leaf samples were collected from 86 plants from distinct areas; 36 were from population AGRO (Agrocondor II Farm, geographic coordinates 55º30' W and 9º00' S) and 50 from population CAR (Carolina Farm, geographic coordinates 57º00' W and 11º00' S). A molecular-diversity study was conducted using 11 microsatellite loci developed for the species. To determine the level of genetic diversity between and within subpopulations, we applied principal coordinate analysis, analysis of molecular variance, observed and expected heterozygosity, polymorphic information ©FUNPEC-RP www.funpecrp.com.br Genetics and Molecular Research 18 (2): gmr18174 F.S. Vieira et al. 2 content (PIC), UPGMA-based clustering, and Bayesian inference structuring. Seventy alleles were found with the SSR markers, with an average PIC of 0.72. Average Ho and He were 0.43 and 0.82, respectively. AMOVA revealed that 81% of the variability is within populations, as found in other studies of Brazil-nut in the states of Pará, Acre and Amazonas. The dendrogram obtained by the UPGMA method and the clustering provided by Bayesian inference resulted in two and four groups formed, respectively. All 36 individuals of the AGRO population were allocated in Group I, and the 50 individuals of population CAR were allocated in Groups II, III, and IV. The two subpopulations have sufficient genetic variability for the composition of an in situ germplasm bank that can be used in breeding programs and in programs for the conservation of the species.
In view of the economic and environmental importance of Bertholletia excelsa Bonpl. for the Amazon and the reducing natural habitat area of this species, the present study proposes to examine genetic variability among accessions of Brazil nut from the Amazon rainforest. Seventeen native Brazil nut genotypes were sampled from Sinop-MT and the genomic DNA were amplified using 15 inter-simple sequence repeat (ISSR) markers. The percent polymorphism and the polymorphism information content (PIC) of each primer were determined. The primers amplified 84.62% of the polymorphic bands and 15.38% of the monomorphic bands. The PIC for each primer ranged from 0 to 0.68. The Unweighted Pair Group Method with Arithmetic Mean (UPGMA) was the most efficient in group representation, as it showed the highest cophenetic correlation coefficient (CCC = 0.92), the lowest stress (12.31%) and the lowest distortion (1.51%). The use of ISSR markers was an efficient tool in the study of genetic diversity among Brazil nut genotypes, and the genetic diversity found can be used for conservation and pre-breeding programs for this crop.
Após a decadência do ciclo econômico da borracha, a castanheira constituiu-se no principal produto extrativo. A crescente destruição da floresta Amazônica influi negativamente na ecologia da castanheira. Padrões de distribuição espacial é uma ferramenta muito utilizada para entender o comportamento ecológico da espécie. Foram demarcadas duas áreas no Parque Nacional do Juruena, nas quais continham um módulo do PPBio em cada, e nelas amostradas todas as castanheiras com DAP>10 cm. Os indivíduos amostrados foram divididos em cinco classes: Jovens; adultos jovens; adultos produtivos; adultos maduros; e adultos idosos. Na área I, 1200 ha, foram amostrados 143 indivíduos, e na área II, 500 ha, 18 árvores. A densidade foi de 0,119 e 0,032 indivíduos ha-1 para a área I e área II respectivamente. 82,52% dos indivíduos da área I estão na classe adultos produtivos e 72,22% estão nesta mesma classe na área II. As árvores amostradas na área I possuem idade média de 255 anos, e na área II de 208. O índice de Morisita, para a área I e área II foi de 4,133 e 7,320 respectivamente. A distribuição espacial das castanheiras amostradas em ambas as áreas foi agregada. A área I possui maior densidade, alta concentração de árvores em período produtivo e maior média de idade do que a área II.
