-Lipase production by a Brazilian wild strain of Yarrowia lipolytica at different stirring speeds and air flow rates was studied. The relationship among lipid consumption, cell growth and lipase production by this microorganism is presented. The most pronounced effect of oxygen on lipase production was determined by stirring speed. Maximum lipase activity was detected in the late stationary phase at 200 rpm and an air flow rate of 1-2 dm 3 /min (0.8-1.7 vvm) when the lipid source had been fully consumed. Higher stirring speeds resulted in mechanical and/or oxidative stress, while lower stirring speeds seemed to limit oxygen levels. An increase in the availability of oxygen at higher air flow rates led to faster lipid uptake and anticipation of enzyme release into the culture medium. The highest lipase production was obtained at 200 rpm and 1 dm 3 /min (0.8 vvm).
Silver nanoparticles (Ag-NPs) are effective nano-filler agents with antifungal activity able to improve bamboo's durability against fungus. Ag-NPs were used to fill up the bamboo biological matrix to obtain an engineered biocomposite material.
To increase the durability of bamboo it is important to find an easy method to fill the micro and meso structure of the biological matrix using a nanostructural material with an anti-fungical activity. A colloidal solution of silver nanoparticle (Ag-NPs) is a dispersion of metal nanoparticle in water with a diameter between 5-100 nm. Even if the biological mechanism is not completly understood yet, Ag-NPs show a satisfactory bactericidal and antifungical activity. We present a simple and rapid production of Ag-NPs made by a sol-gel synthesis in flow mode by means of microreactor tecnology through a chemical reduction of AgNO3 with NaBH4 in presence of two different organic ligands: sodium/potassium tartrate and trisodium citrate. The synthesis of Ag-NPs in continuous flow compared to the batch technique allowed to reduce the time of synthesis, facilitating the reproducibility of the process and consequently obtaining NPs with more uniform physical and chemical characteristics. The microorganisms of the genus Aspergillus were used for the microbiological tests. The effect of different Ag-NPs on microbial growth was observed daily. In particular, it was shown that the response of the fungus is inversely proportional to the size of the nanoparticles, cell growth is disrupt depending on the proportion between silver and organic ligand and microbialstatic effect, especially in relation to sporulation stage was also observed.
Enzymes are frequently used in biotechnology to carry out specific biological reactions, either in industrial processes or for the production of bioproducts and drugs. Microbial lipases are an important group of biotechnologically valuable enzymes that present widely diversified applications. Lipase production by microorganisms is described in several published papers, however, none of them refer to metrological evaluation and the estimation of the uncertainty in measurement. Moreover, few of them refer to process optimization through experimental design. D-optimal design was used to enhance lipase production in shaken-flasks with Yarrowia lipolytica cells. Analysis of the metrological aspects of this process is presented. It suggests a significant reduction in the cost of the enzyme. Lipase production by Yarrowia lipolytica cells aiming the classification as certified reference material is recommended after further purification and stability studies.
