The alfalfa plant, after harvesting, was washed, dried, and grinded to get fine powder used in water treatment. We used the alfalfa plant with ethanol to make the alcoholic extract characterized by using (GC-Mass, FTIR, and UV) spectroscopy to determine active compounds. Alcoholic extract was used to prepare zinc nanoparticles. We characterized Zinc nanoparticles using (FTIR, UV, SEM, EDX Zeta potential, XRD, AFM). Zinc nanoparticle with Alfalfa extract and alfalfa powder were used in the treatment of water polluted with inorganic elements such as Cr, Mn, Fe, Cu, Cd, Ag by (Batch processing). The batch process with using alfalfa powder gets treated with Pb (51.45%), which is the highest percentage of treatment. Mn (13.18%), which is the lowest percentage of treatment. The batch process with using Zinc nanoparticles gets the result treated with Pb(98.822%), which is the highest percentage of treatment, and Mn (10.31%), which is the lowest percentage of treatment. When comparing alfalfa powder and zinc nanoparticle, it has been found that the treatment with zinc nanoparticle is more efficient in the removal of inorganic pollutants.
An isolated Lactobacillus from several various sources were identified depending on morphological, microscopically and biochemical tests in vitro analysis of probiotic properties that included: an ability to tolerate in different concentration of bile salt, survival in acidic conditions, their antimicrobial activity, and S-layer characterizations were carried out. It was noticed that isolates of Lactobacillus rhamnosus and L. delbrueckii have a broad activity of antimicrobial and found the isolate L. rhamnosus represented with a survival percentage 6.9% at pH 4.5 and 5.1% at pH 2.0) also L. rhamnosus (5.7% at pH 4.5 and 4.9% at pH 2.0) tolerated acidic media, Lactobacillus spp. has antimicrobial activity against all gram-positive and negative tested isolates. 70 kDa of S-layer protein bands were detected with whole-cell SDS-PAGE analysis, and it's predominant in cells of isolates which grown in MRS broth anaerobically. It was noticed that the collected Lactobacillus isolates could be used as probiotic.
The Catharanthus roseus plant was extracted and converted to nanoparticles in this work. The Soxhlet method extracted alkaloid compounds from the plant Catharanthus roseus and converted them to the nanoscale. Chitosan polymer was used as a linking material and converted to Chitosan nanoparticles using Sodium TriPolyPhosphate (STPP). The extracted alkaloids were linked with Chitosan nanoparticles CSNPs by maleic anhydride to get the final product (CSNPs- Linker- alkaloids). The synthesized (CSNPs- Linker- alkaloids) was characterized using SEM spectroscopy UV–Vis., Zeta Potential, and HPLC High-Performance Liquid Chromatography. Scanning electron microscope (SEM) analysis shows that the Chitosan nanoparticles (CSNPs) have small dimensions with regular spherical and nanotube shapes of a diameter range of (49 - 70) nm. The final product (CSNPs- Linker- alkaloids) has two shapes (spherical particles and tubes) in nano dimensions and is close to each other compared to normal Chitosan. The absorption peaks for Chitosan (CS), Chitosan nanoparticles (CSNPs), Chitosan nanoparticles (CSNPs), and maleic anhydride revealed that converting Chitosan to Chitosan nanoparticles and mixing it with the plant extract, led to an increase in the absorption value and wavelength range. Also, the appearance of two peaks at 222 nm and 402 nm nano instead of the peak of Chitosan at 289.9 nm. Zeta Potential results of CSNPs- Linker- alkaloids showed that the extract of the nano-alkaloids bound to chitosan nanoparticles carries a positive charge of 54.4 mV. This surface charge is essential in maintaining the colloidal solution's stability in its natural form without changing. High-Performance Liquid Chromatography (HPLC) was used to estimate qualitative and quantitative plants extracted from Catharanthus roseus. Quantitative HPLC results show that Catharanthus roseus contains a good and acceptable concentration of Vinblastine, Vincristine, Vinorelbine, Vincamine, and Vintafolide (66.75, 242.91, 0.7, 83.77, 42.34) ppm respectively. The qualitative results show a good match for the influential groups of pure standard vincristine and alcoholic extract and dry powder of the Catharanthus roseus plant. The successful synthesis of nanoparticles from the Catharanthus roseus plant can be used in biosensors and biomedical applications.
Objectives: To identify and to characterize of methicillin-resistant Staphylococcus aureus (MRSA) isolates collected from food (n=43), food handler (n=8), and clinical samples (n=15). Methods: Genotyping was applied for the purposes of epidemiological investigation and source tracking of these isolates. Pulsed field gel electrophoresis (PFGE) was performed using the restriction endonucleases SmaI. Findings: PFGE analysis showed that pulse types of isolates from clinical samples differed that from food and food handler isolates. However, a similarity was observed between pulse types of isolates from food and food handlers, suggesting an epidemiological association between isolates from these two sources and that MRSA may be transmitted from food handlers to foods. Novelty : PFGE genotyping of MRSA isolates was useful, as a first study of its type in Iraq, for investigating epidemiological relatedness and even contaminated food sources.
