β-Thalassemia merupakan suatu kelainan genetik yang bersifat herediter dengan prevalensi tinggi di Asia Tenggara. Di Indonesia, diperkirakan sejumlah 200.000 bayi pembawa thalassemia lahir setiap tahun sehingga diperlukan upaya deteksi dini yang sederhana, cepat, dan murah. NESTROFT (Naked Eye Single Tube Red Cell Osmotic Fragility Test) sebagai metode deteksi berdasarkan fragilitas sel darah merah memiliki sensitivitas uji pada carrier β-thalassemia sebesar 99,8%. Oleh karena itu, pengembangan metode tersebut untuk deteksi pembawa β-Thalassemia di populasi Indonesia perlu dikembangkan. Penelitian dilakukan secara eksperimental menggunakan 102 sampel individu dewasa. Dilakukan uji NESTROFT dengan mencampurkan sampel uji dalam dua larutan, yakni buffer saline 0,36% sebagai reagen uji dan akuades steril sebagai kontrol. Hasil positif ditunjukkan dengan garis hitam pada latar belakang kertas tidak dapat dilihat secara kasat mata pada sampel uji dibandingkan kontrol akibat adanya penurunan osmotic fragility pada eritrosit. Selanjutnya, pemeriksaan indeks hematologis juga dilakukan sebagai data konfirmasi. Hasil penelitian menunjukkan bahwa uji NESTROFT dapat mendeteksi sebesar 88,9% dari total subyek pembawa sifat β-thalassemia. Hasil pemeriksaan hematologis juga menunjukkan kondisi sampel positif dengan ciri sebagai pembawa β-thalassemia seperti kondisi mikrositik, hipokromik, anisositosis, poikilositosis, serta nilai MCV maupun MCH rendah. Kesimpulan dari penelitian menunjukkan bahwa uji NESTROFT dapat dijadikan sebagai langkah awal diagnosis (preliminary detection) pembawa β-thalassemia yang efektif dan efisien. Selain itu, uji NESTROFT dapat digunakan pula untuk deteksi dini kelainan hemoglobin lainnya.
