RESUMO Os resíduos agroindustriais são materiais alternativos e de baixo custo, uma vez que quantidades consideráveis destes são descartadas no ambiente, com potencial uso como materiais adsorventes na remoção de poluentes, por exemplo. Nesse sentido, este trabalho foi realizado com o objetivo de avaliar a utilização da fibra de coco verde, como adsorvente na remoção do corante azul de metileno em meio aquoso. O corante azul de metileno é muito utilizado pela indústria têxtil e encontra-se presente em seus efluentes podendo contaminar o ambiente. A fibra de coco verde foi tratada com HCl, NaOH e hexano. Os parâmetros avaliados foram pH, tempo de agitação, mecanismo cinético e equilíbrio de adsorção. Os dados cinéticos foram ajustados aos modelos de Pseudo primeira ordem, Pseudo segunda ordem, Difusão intrapartícula e Quimissorção, resultando como o mais adequado para o processo o modelo de Pseudo segunda ordem. As isotermas de adsorção foram delineadas aos modelos de Langmuir e Freundlich, sendo este último o que apresentou melhor ajuste para os dados experimentais obtidos. A capacidade máxima adsortiva foi verificada, sendo o tratamento com HCl (0,1 mol L-1) aquele que proporcionou aumento significativo na capacidade adsortiva máxima (166,7 mg g-1) da fibra de coco verde quando comparada a fibra de coco in natura e aos demais tratamentos. Os resultados mostraram que a fibra de coco verde, tratada com ácido clorídrico, pode ser utilizada como material adsorvente na remediação de poluentes orgânicos, como corantes catiônicos em efluentes líquidos.
Urease is an enzyme that catalyzes the hydrolysis of urea into carbon dioxide and ammonia, and its soil activity can be evaluated by monitoring the concentration of the products or reagents in the reaction. This work presents the development of a novel analytical methodology to evaluate the enzymatic activity of the urease in the soil, from the quantification of ammonium ion, product of urea hydrolysis, using flow injection analysis with gas diffusion and conductometric detection. The results were compared with the colorimetric method. The conductivity response was evaluated for flow parameters and NaOH concentration. The method presented a linear range of 0.02 to 4.0 mmol L -1 of NH4 + , limit of detection 0.02 mmol L -1 and theoretical analytical frequency of 28 h -1 samples. The proposed method presented repeatability and accuracy comparable to the colorimetric method. The practicality, the high analytical frequency are great attractions for the use of the method proposed in the evaluation of the enzymatic activity of the urease in the soil.
ResumoUrease é uma enzima que catalisa a hidrólise da ureia em dióxido de carbono e amônia, sendo que sua atividade no solo pode ser avaliada monitorando a concentração dos produtos ou reagentes dessa reação. O presente trabalho objetivou o desenvolvimento de metodologia analítica para avaliação da atividade enzimática da uréase no solo, a partir da quantificação do íon amônio, produto da hidrólise da ureia, utilizando análise por injeção em fluxo com difusão gasosa e detecção condutométrica. Os resultados foram comparados com o método colorimétrico. A resposta de condutividade foi avaliada para os parâmetros de fluxo e concentração de NaOH. O método apresentou faixa linear de 0,02 a 4,0 mmol L -1 de NH4 + , limite de quantificação de 0,07 mmol L -1 e frequência analítica teórica de 28 amostras h -1 . O método proposto apresentou precisão e exatidão comparável ao método colorimétrico. A praticidade, a elevada frequência analítica são grandes atrativos para a utilização do método proposto na avaliação da atividade enzimática da uréase no solo.Palavras-chave: Sistema FIA; enzima; análise colorimétrica.
