Abstract:In this work, the modifications promoted by alkaline hydrolysis and glutaraldehyde (GA) crosslinking on type I collagen found in porcine skin have been studied. Collagen matrices were obtained from the alkaline hydrolysis of porcine skin, with subsequent GA crosslinking in different concentrations and reaction times. The elastin content determination showed that independent of the treatment, elastin was present in the matrices. Results obtained from in vitro trypsin degradation indicated that with the increase of GA concentration and reaction time, the degradation rate decreased. From thermogravimetry and differential scanning calorimetry analysis it can be observed that the collagen in the matrices becomes more resistant to thermal degradation as a consequence of the increasing crosslink degree. Scanning electron microscopy analysis indicated that after the GA crosslinking, collagen fibers become more organized and well-defined. Therefore, the preparations of porcine skin matrices with different degradation rates, which can be used in soft tissue reconstruction, are viable.
The isolation and identification of bioactive metabolites from complex extracts obtained from microbial growth media is a time consuming, costly, and labor-intensive task. A strategy to rapidly identify secondary metabolites isolated from extracts obtained from the culture media of marine-derived and endophytic fungal strains is described. Identification was achieved by HPLC-UV-MS and 1 H NMR analyses in combination with data obtained from the Dictionary of Natural Products. Among the compounds identified, (-)-naphthoquinoneimine, citreorosein, emodin, pyrenocine A and harzianopyridone displayed moderate to potent antiviral activity. (-)-Naphthoquinoneimine was isolated as the enantiomer of its previously reported dextrorotatory congener, while 6,7-dihydroxy-2,2-dimethyl-4-chromanone is herein reported for the first time as a natural product.

The interest in the use of evaporative light scattering detector (ELSD) for the analysis of different classes of natural products has grown over the years. This is because this detector has become an excellent alternative compared to other types of detectors, such as the refractive index detector and the ultraviolet (UV) detector. This review describes the basic principles of ELSD functioning and discusses the advantages and disadvantages in using an ELSD for the analysis of organic compounds. Additionaly, an overview, covering the last 23 years, of ELSD applications in natural products analysis (saponins, terpenes, carbohydrates, glycosides, alkaloids, steroids, flavonoids, peptides, polyketides, coumarins and iridoids) is presented and discussed.
The aim of this study were characterize acellular collagen matrices derived from porcine pericardium (PP) and to evaluate their properties after sterilization by ethylene oxide and gamma ray. PP matrices were subjected to alkaline hydrolysis (AH), and samples were characterized for biological stability, membrane thickness measurements, differential scanning calorimetry (DSC) and scanning electron microscopy (SEM). Subsequently, the matrices were frozen, lyophilized and sterilized by ethylene oxide or gamma radiation. For in vitro assays, CHO-K1 cell culture was used and evaluated for cytotoxicity, clonogenic survival assay, genotoxicity and mutagenicity. Analysis of variance (ANOVA) was used, followed by Dunnett’s post-test, with a significance level of 5%. After AH, there was no significant change in matrix thickness. The relative biodegradability of the material after implantation was observed. Morphology and dimensions had small changes after AH. As for cell viability, none of the tested matrices showed a statistically significant difference (p > 0.05; Dunnett) regardless of the sterilization method. Furthermore, it was found that PP matrices did not interfere with the proliferation capacity of CHO-K1 cells (p > 0.05; Dunnett). As for genotoxicity, when sterilized with ethylene oxide (NP, P12 and P24), it showed genotoxic potential, but it was not genotoxic when sterilized by gamma radiation. No mutagenic effects were observed in either group. PP-derived collagen matrices hydrolyzed at different times were not cytotoxic. It is concluded that the best method of sterilization is through gamma radiation, since no significant changes were observed in the properties of the PP matrices.

Em casos severos de trauma, os processos naturais de regeneração são insuficientes no reparo dos danos, resultando em lesões cutâneas crônicas. A desvitalização de matrizes homólogas ou heterólogas é uma alternativa para a produção de matrizes dérmicas. A pele porcina é bastante similar à pele humana, podendo ser utilizada como matriz de colágeno na regeneração de tecido mole. Além disso, ela tem como constituinte principal o colágeno tipo I, e, assim, pode ser utilizada em queimaduras de segundo grau. Este trabalho teve como objetivo a preparação e caracterização de matrizes extracelulares de colágeno tipo I por meio de hidrólise alcalina e reticulação com glutaraldeído (GA). As matrizes de colágeno foram obtidas a partir da hidrólise alcalina de pele porcina, com posterior reticulação com GA, em diferentes concentrações (0-0,1%) e tempos de reação (15 e 45 min). As matrizes foram caracterizadas através de determinação do conteúdo de elastina, estabilidade biológica (tripsina), calorimetria exploratória diferencial (DSC), termogravimetria (TG/DTG), microscopia eletrônica de varredura (MEV) e avaliação preliminar da citotoxicidade in vitro. Através da determinação do conteúdo de elastina, foi possível determinar a massa média de colágeno presente nas matrizes, a qual foi de 95,2±0,2% (m/m), e a massa média de elastina, que foi de 4,8±0,2% (m/m), e também verificar que independente do tratamento, a elastina estava presente nas matrizes. O ensaio de estabilidade biológica mostrou que o tratamento com GA diminui a biodegradação do material; sendo obtidas porcentagens de degradação que variaram de 83,6%±1,1 (0% GA) a 46,1%±0,7 (0,085% -45min), indicando, assim, que com o aumento da concentração de GA e do tempo de reação, há uma diminuição da porcentagem de degradação. Pela análise termogravimétrica, foi observado que o colágeno presente nas matrizes tornou-se mais estável termicamente em conseqüência do aumento do grau de reticulação e, portanto, mais resistentes à degradação térmica. Os resultados de DSC confirmam os de termogravimetria devido ao aumento nos valores das temperaturas de desnaturação das matrizes em função do aumento do tempo de reação e da concentração...
