Hippocampal atrophy is reported in several neuropathological disorders. The hippocampal dentate gyrus (DG) is a brain region where adult neurogenesis constitutively occurs. There are some reports suggesting the ability of endogenous neurogenesis to initiate neuronal repair in the hippocampus in response to neuropathological conditions, but its capacity to compensate for neuronal loss is limited. Among strategies to enhance adult hippocampal neurogenesis are enriched environment and lithium. This study aimed to assess whether both strategies could interact to potentiate the generation of new cells in the adult DG. Healthy adult male C57BL/6 mice were divided into four treatment groups for 28 days: control, lithium, enriched environment, enriched environment plus lithium. The animals were injected with BrdU (cell proliferation marker) shortly before the start of the treatments and killed 28 days later for analysis of newly generated cells. Two-way ANOVA followed by post hoc test revealed a significant synergistic interaction between enriched environment and lithium in the total number of BrdU(+) cells in the entire DG (p = 0.019), a trend towards significant synergistic interaction in the dorsal DG (p = 0.075), and a significant additive effect in the ventral DG (p = 0.001). These findings indicate that the combination of enriched environment and lithium has both synergistic and additive effects on the generation of new cells in the healthy adult DG (these effects being possibly segregated along the dorso-ventral axis of the hippocampus), and suggest that it might be worth investigating whether this combination would have a similar effect in neuropathological conditions.

RESUMO:Com a descoberta nas últimas décadas de que neurogênese (nascimento de novos neurônios) persiste no cérebro adulto, surgiu a hipótese de que doenças neurodegenerativas como a doença de Alzheimer poderiam ser superadas, ou pelo menos melhoradas, uma vez que a geração de novos neurônios poderia ajudar a compensar a perda de neurônios nessas doenças. De fato, a neuroproliferação está aumentada no cérebro de sujeitos com doença de Alzheimer, mas os novos neurônios falham em sobreviver e se diferenciar em células maduras. O objetivo deste trabalho foi examinar o efeito isolado bem como o efeito combinado de carbonato de lítio (2,0 g/kg de ração) e enriquecimento ambiental (com estimulação cognitiva e física) sobre a sobrevivência de novas células geradas no hipocampo de camundongos adultos. Camundongos C57Bl/6 de 3 meses de idade no início do estudo foram submetidos por 4 semanas à uma das seguintes condições de tratamento: (a) Ambiente Convencional sem Lítio (n = 7); (b) Ambiente Convencional mais Lítio (n = 7); (c) Ambiente Enriquecido sem Lítio (n = 7); e (d) Ambiente Enriquecido mais Lítio (n = 6). Imediatamente antes do início dos tratamentos, os animais receberam injeção intraperitoneal de BrdU, um marcador de proliferação celular. Vinte e seis dias após o início dos tratamentos, foi coletado sangue da cauda dos animais para medidas de litemia. Vinte e oito dias após a injeção de BrdU, os animais foram sacrificados através de perfusão intracardíaca seguida por fixação com paraformaldeído 4%, para verificação da sobrevivência de células proliferativas. Os cérebros fixados foram cortados em criostato em cortes seriais adjacentes de 60 �m, e os cortes foram processados para marcação imuno�m, e os cortes foram processados para marcação imunom, e os cortes foram processados para marcação imunohistoquímica para BrdU. O número de células BrdU-positivas (BrdU+) foi contado no giro denteado do hipocampo com o uso de um microscópio de luz e objetiva de imersão (aumento 100x). O tratamento com carbonato de lítio por 4 semanas resultou em litemia média de 0,65 mmol/L, portanto, dentro da faixa terapêu-tica que é entre 0,5 e 1,0 mmol/L.
