Science has a critical role to play in guiding more sustainable development trajectories. Here, we present the Sustainable Amazon Network ( Rede Amazônia Sustentável , RAS): a multidisciplinary research initiative involving more than 30 partner organizations working to assess both social and ecological dimensions of land-use sustainability in eastern Brazilian Amazonia. The research approach adopted by RAS offers three advantages for addressing land-use sustainability problems: (i) the collection of synchronized and co-located ecological and socioeconomic data across broad gradients of past and present human use; (ii) a nested sampling design to aid comparison of ecological and socioeconomic conditions associated with different land uses across local, landscape and regional scales; and (iii) a strong engagement with a wide variety of actors and non-research institutions. Here, we elaborate on these key features, and identify the ways in which RAS can help in highlighting those problems in most urgent need of attention, and in guiding improvements in land-use sustainability in Amazonia and elsewhere in the tropics. We also discuss some of the practical lessons, limitations and realities faced during the development of the RAS initiative so far.

Concatenated sequence analysis with 16S rRNA, rpoB and fusA genes identified a bacterial strain (IRBG74) isolated from root nodules of the aquatic legume Sesbania cannabina as a close relative of the plant pathogen Rhizobium radiobacter (syn. Agrobacterium tumefaciens). However, DNA:DNA hybridization with R. radiobacter, R. rubi, R. vitis and R. huautlense gave only 44%, 5%, 8% and 8% similarity respectively, suggesting that IRBG74 is potentially a new species. Additionally, it contained no vir genes and lacked tumour-forming ability, but harboured a sym-plasmid containing nifH and nodA genes similar to those in other Sesbania symbionts. Indeed, IRBG74 effectively nodulated S. cannabina and seven other Sesbania spp. that nodulate with Ensifer (Sinorhizobium)/Rhizobium strains with similar nodA genes to IRBG74, but not species that nodulate with Azorhizobium or Mesorhizobium. Light and electron microscopy revealed that IRBG74 infected Sesbania spp. via lateral root junctions under flooded conditions, but via root hairs under non-flooded conditions. Thus, IRBG74 is the first confirmed legume-nodulating symbiont from the Rhizobium (Agrobacterium) clade. Cross-inoculation studies with various Sesbania symbionts showed that S. cannabina could form fully effective symbioses with strains in the genera Rhizobium and Ensifer, only ineffective ones with Azorhizobium strains, and either partially effective (Mesorhizobium huakii) or ineffective (Mesorhizobium plurifarium) symbioses with Mesorhizobium. These data are discussed in terms of the molecular phylogeny of Sesbania and its symbionts.

Tropical forests have a high diversity of plant species; are they associated with a correspondingly rich microbial flora? We addressed this question by examining the symbiotic rhizobium bacteria that nodulate a diverse pool of forest legume species in Brazil. The 44 strains studied had been isolated from 29 legume tree species representing 13 tribes including all three subfamilies of the Leguminosae, and were chosen to represent major groups from a larger sample that had previously been characterized by SDS-PAGE of total proteins. Partial 16S rRNA gene sequence was determined, corresponding to positions 44-303 in the Escherichia coli sequence. Fifteen sequences were found, including six novel ones. However, all but one of them could be assigned to a genus because they grouped closely with sequences from previously described rhizobial species. Fast-growing strains had sequences similar to Rhizobium spp., Sinorhizobium spp. or Mesorhizobium spp., while the slow-growing strains had sequences similar to Bradyrhizobium spp. One strain with an intermediate growth rate had a unique sequence which indicated that the strain might belong to the genus Azorhizobium. Although the strains showed a variety of sequences, it was surprising that these strains isolated from taxonomically very diverse host plants in previously unexplored environments were mostly very similar to strains described previously, largely from agricultural systems.

Resumo -O objetivo do trabalho foi avaliar o efeito de doses excessivas de Zn no crescimento e nutrição do eucalipto, em casa de vegetação. Mudas de Eucalyptus maculata e Eucalyptus urophylla foram crescidas em vasos contendo 2 L de solução nutritiva de Clark, adicionando-se doses crescentes de Zn: 0, 400, 800, 1.200 e 1.600 µM fornecidas como ZnSO 4 . Após cinco semanas, as plantas exibiram clorose internerval, escurecimento das raízes e inibição do crescimento, mesmo nas doses mais baixas do metal. A dose crítica de Zn na solução (para redução de 10% na matéria seca da parte aérea) em E. maculata foi de 170,3 µM, e em E. urophylla, 73,0 µM. Os níveis críticos de toxidez de Zn na planta foram de 853 mg kg -1 em E. maculata, e 697,8 mg kg -1 em E. urophylla. Esses resultados indicam que E. maculata é mais tolerante ao Zn do que E. urophylla. Altas doses de Zn reduziram as concentrações de Fe e Ca na matéria seca da parte aérea a níveis considerados deficientes para o crescimento das duas espécies. A translocação de Fe das raízes para a parte aérea foi reduzida, independentemente da espécie, de 21% no controle para apenas 2% em 1.600 µM de Zn, indicando forte relação entre a queda na produção de matéria seca e a ocorrência da deficiência induzida de Fe nas plantas.Termos para indexação: eucalipto, nutrição das plantas, oligoelementos, metais pesados. Zinc toxicity on growth and nutrition of Eucalyptus maculata and Eucalyptus urophylla in nutrient solutionAbstract -The objective of this work was to evaluate the effect of excessive doses of Zn on growth and nutrition of eucalyptus in a greenhouse experiment. Seedlings of Eucalyptus maculata and Eucalyptus urophylla were grown in pots containing 2 L of Clark's nutrient solution, adding increasing levels of Zn, in µM: 0, 400, 800, 1,200 and 1,600 supplied as ZnSO 4 . After five weeks, plants exhibited internerval chlorosis, dark roots and growth inhibition, even in lower metal levels. The critical metal dose in the solution, for 10% reduction of shoot dry matter, was 170.3 µM and 73.0 µM in E. maculata and E. urophylla, respectively. The plant critical toxicity levels were 853 mg kg -1 in E. maculata and 697.8 mg kg -1 in E. urophylla. These results indicate that E. maculata is more tolerant to Zn than E. urophylla. High levels of Zn reduced concentrations of Fe and Ca in shoot dry matter to levels considered deficient for the growth of both species. Translocation of Fe from roots to shoots was also reduced, independently of the species, of 21% in control to 2% in 1,600 µM Zn, indicating strong relationship among dry matter reduction and induced Fe-deficiency in plants.
