The study aimed at determining the effect of melatonin on the activity of protective antioxidative enzymes in the heart and of lipid peroxidation products in the course of intoxication with doxorubicin (DOX). The rats were categorized into four groups, receiving: 0.9% NaCl i.p. (NaCl control); melatonin [20 mg/kg body weight (b.w.)] s.c. (control Mel); DOX (2.5 mg/kg b.w.) i.p.; melatonin plus DOX in doses as above. All the substances were administered once in a week for four consecutive weeks. Homogenates of heart tissue were examined for activities of glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT), levels of reduced glutathione (GSH) and of lipid peroxidation indices (MDA + 4-HDA). Administration of melatonin alone did not induce alterations in levels of MDA + 4-HDA, GSH, or in activity of GPx, SOD or CAT, as compared to the group receiving 0.9% NaCl. GSH levels decreased following DOX but remained at normal levels following DOX and melatonin. The level of MDA + 4-HDA increased following DOX, as compared with the control, a change prevented by the combination of DOX + melatonin. Activities of GPx, SOD and CAT were higher in groups receiving DOX and/or DOX plus melatonin than in control groups. Activity of CAT and the level of GSH in the group receiving DOX plus melatonin were significantly higher than in the group intoxicated with DOX alone. The obtained results demonstrate that, when given in parallel with DOX, melatonin protects cardiomyocytes from damaging effects of the cytostatic drug (reflected by the levels of MDA + 4-HDA). The protective effect resulted, in part from the augmented levels of GSH and from stimulation of CAT activity by melatonin in cardiomyocytes subjected to the action of DOX.
Fundamental motor skills (FMS) are the basic elements of more complex sport-specific skills and should be mastered at the end of early childhood; however, the relationship between FMS and sport-specific skills has not yet been verified in prepubertal soccer players. Therefore, the aim of this study was to determine the role of FMS in the process of acquiring soccer-specific motor skills (measured using speed dribbling) with regard to physical fitness and biological maturation. Forty male soccer players (11.5 ± 0.3 years of age) at the highest performance level participated in the study. The test of Gross Motor Development – second edition and Unifittest 6–60 were used to assess FMS and physical fitness, respectively. The role of FMS in a complex theoretical model with the relationships between physical fitness, biological maturation and speed dribbling was analyzed by multiple regression path analyses (MRPA). Moderate to strong correlations were found between FMS, physical fitness, and speed dribbling ( r = 0.56–0.66). Biological maturation did not appear to be a significant predictor of physical fitness or speed dribbling. The MRPA model using FMS as mediator variable between physical fitness and speed dribbling showed a significant indirect effect (standard estimation = −0.31, p = 0.001; R 2 = 0.25). However, the direct correlation between physical fitness and speed dribbling was non-significant. Our results showed that FMS significantly strengthened the influence of physical fitness on the performance of speed dribbling, a soccer-specific motor skill, and thus play an important role in the process of acquiring sport-specific motor skills in prepubertal soccer players. When considering the long-term training process, especially during childhood and before puberty, a wide range of FMS activities should be applied for better and possibly faster acquisition of soccer-specific motor skills.
This study aimed to investigate the effects of the 6-week small-sided games training (SSGs) vs. high-intensity interval training (HIIT) on the psychophysiological and performance responses, and technical skills of young basketball players. Thirty-two male players (age: 14.5 ± 0.5 years of age) were randomly divided into SSGs group (n = 16) and HIIT group (n = 16) training methods thrice per week for 6 weeks. The players in the SSGs group performed two 5–8 min of 2 vs. 2 with 2 min rest periods, while the players in HIIT performed 12–18 min of runs at intensities (90 to 95%) related to the velocity obtained in the 30-15 intermittent fitness test (IFT). Pre-testing and post-testing sessions involved assessments of Yo-Yo Intermittent Recovery Test level 1, 30-15 intermittent fitness test, 5 and 30 m sprint times, vertical jump height, repeated sprint ability, defensive and offensive agility, and technical skills. The SSGs group demonstrated significantly higher agility-based technical responses in terms of the control dribbling and shooting skills (d = 1.71 vs. 0.20, d = 1.41 vs. 0.35, respectively) compared with the HIIT group. Conversely, the HIIT induced greater improvements in 30 m sprint times (d = 3.15 vs. 0.68). These findings provided that SSGs in youth basketball players may allow similar positive physical adaptations to HIIT, with an extra advantage of improving technical skills while improving enjoyability.
High-intensity interval training (HIIT) is frequently utilized as a method to reduce body mass. Its intensity of work results in a number of beneficial adaptive changes in a relatively short period of time. Irisin is a myokine and adipokine secreted to the blood during exercise and it takes part in the regulation of energy metabolism. It is a vital issue from the prophylaxis point of view as well as treatment through exercise of different diseases (e.g., obesity, type-2 diabetes). The aim of this study was to evaluate changes in irisin concentration, body composition, and aerobic and anaerobic performance in men after HIIT. Eight weeks of HIIT following the Tabata protocol was applied in the training group (HT) (n = 15), while a sedentary group (SED) (n = 10) did not participate in fitness activities within the same time period. Changes of irisin, body composition, and aerobic and anaerobic performance were evaluated after graded exercise test (GXT) and Wingate anaerobic test (WAnT) before and after eight weeks of training. Training resulted in an increased of blood irisin concentration (by 29.7%) p < 0.05), VO2max increase (PRE: 44.86 ± 5.74 mL·kg−1·min−1; POST: 50.16 ± 5.80 mL kg−1·min−1; p < 0.05), reduction in percent body fat (PRE: 14.44 ± 3.33%; POST: 13.61 ± 3.16%; p < 0.05), and increase of WAnT parameters (p < 0.05) in the HT group. No changes were observed in the SED group. HIIT resulted in beneficial effects in the increase in blood irisin concentration, physical performance, and reduced fat content. The HIIT may indicate an acceleration of base metabolism. This effect can be utilized in the prevention or treatment of obesity.
Adipose tissue is an organ that performs a lot of significant physiological functions, which is why its excess in the body results in pathological states in many of its organs and systems. Adipose tissue is not only a tissue which stores fat and plays a protective role. It is an important endocrine organ where signals sent from different tissues are generated and integrated. Adipose tissue is both morphologically and physiologically differentiated. However, due to their plasticity, adipocytes can undergo a transformation and change their structure and metabolism depending on the physiological state of the organism and the conditions to which it is exposed.
The study determined the effects of sprint interval training on the acute and chronic changes of serum brain-derived neurotrophic factor (BDNF) and aerobic capacity. Twenty-six cyclists were divided into experimental (E) and control groups. Both groups executed a 6-month exercise intervention involving high-intensity interval training (HIIT) and continuous endurance training (CET) with group E replacing HIIT and CET sessions with sprint interval training (SIT) that was executed twice a week. Two exercise tests were administered prior to the intervention and at 2 and 6 months after study outset. Incremental exercise test assessed aerobic capacity by measuring maximal oxygen uptake and work output; the sprint interval exercise test (SIXT) comprises 3 sets of four 30-s all-out repetitions interspersed with 90 s of rest with sets separated by 25–40 min of active recovery. Oxygen uptake, work output, BDNF, and vascular endothelial growth factor A (VEGF-A) concentrations (baseline, 10 min after first set, and 10 and 60 min after third SIXT set) were taken during the SIXT. Significant decreases in BDNF relative to baseline values were observed 10 min after the first set and 60 min after the third set in group E at the 2- and 6-month assessments. Increases in baseline VEGF-A after 2 and 6 months of training and increases in maximal oxygen uptake after 2 months of training were also observed only in group E. The inclusion of SIT with HIIT and CET shows positive long-term effects, including increased maximal oxygen uptake and baseline VEGF-A and a reduction in BDNF below baseline levels during and after SIXT.
This study examined the effects of a nine-week intervention of four different high-intensity training modalities [high-intensity functional training (HIFT), high-intensity interval training (HIIT), high-intensity power training (HIPT), and high-intensity endurance training (HIET)] on the resting concentration of brain-derived neurotropic factor (BDNF). In addition, we evaluated the BDNF responses to Graded Exercise Test (GXT) and Wingate Anaerobic Test (WAnT) in men. Thirty-five healthy individuals with body mass index 25.55 ± 2.35 kg/m2 voluntarily participated in this study and were randomly assigned into four training groups. During nine-weeks they completed three exercise sessions per week for one-hour. BDNF was analyzed before and after a GXT and WAnT in two stages: (stage 0—before training and stage 9—after nine weeks of training). At stage 0, an increase in BDNF concentration was observed in HIFT (33%; p < 0.05), HIPT (36%; p < 0.05) and HIIT (38%; p < 0.05) after GXT. Even though HIET showed an increase in BDNF (10%) this was not statistically significant (p > 0.05). At stage 9, higher BDNF levels after GXT were seen only for the HIFT (30%; p < 0.05) and HIIT (18%; p < 0.05) groups. Reduction in BDNF levels were noted after the WAnT in stage 0 for HIFT (− 47%; p < 0.01), HIPT (− 49%; p < 0.001), HIET (− 18%; p < 0.05)], with no changes in the HIIT group (− 2%). At stage 9, BDNF was also reduced after WAnT, although these changes were lower compared to stage 0. The reduced level of BDNF was noted in the HIFT (− 28%; p < 0.05), and HIPT (− 19%;p < 0.05) groups. Additionally, all groups saw an improvement in VO2max (8%; p < 0.001), while BDNF was also correlated with lactate and minute ventilation and selected WAnT parameters. Our research has shown that resting values of BDNF after nine weeks of different forms of high-intensity training (HIT) have not changed or were reduced. Resting BDNF measured at 3th (before GXT at stage 9) and 6th day after long lasting HITs (before WAnT at stage 9) did not differed (before GXT), but in comparison to the resting value before WAnT at the baseline state, was lower in three groups. It appears that BDNF levels after one bout of exercise is depended on duration time, intensity and type of test/exercise.
