With the present work, we aim to provide a better understanding of chromosome evolutionary trends among southern Brazilian species of Iridoideae. Chromosome numbers and genome sizes were determined for 21 and 22 species belonging to eight genera of Tigridieae and two genera of Trimezieae, respectively. The chromosome numbers of nine species belonging to five genera are reported here for the first time. Analyses of meiotic behaviour, tetrad normality and pollen viability in 14 species revealed regular meiosis and high meiotic indexes and pollen viability (> 90%). The chromosome data obtained here and compiled from the literature were plotted onto a phylogenetic framework to identify major events of chromosome rearrangements across the phylogenetic tree of Iridoideae. Following this approach, we propose that the ancestral base chromosome number for Iridoideae is x = 8 and that polyploidy and dysploidy events have occurred throughout evolution. Despite the variation in chromosome numbers observed in Tigridieae and Trimezieae, for these two tribes our data provide support for an ancestral base number of x = 7, largely conserved in Tigridieae, but a polyploidy event may have occurred prior to the diversification of Trimezieae, giving rise to a base number of x2 = 14 (detected by maximum‐parsimony using haploid number and maximum likelihood). In Tigridieae, polyploid cytotypes were commonly observed (2x, 4x, 6x and 8x), whereas in Trimezieae, dysploidy seems to have been the most important event. This feature is reflected in the genome size, which varied greatly among species of Iridoideae, 4.2‐fold in Tigridieae and 1.5‐fold in Trimezieae. Although no clear difference was observed among the genome sizes of Tigridieae and Trimezieae, an important distinction was observed between these two tribes and Sisyrinchieae, with the latter possessing the smallest genome sizes in Iridoideae. © 2014 The Linnean Society of London, Botanical Journal of the Linnean Society, 2014, 177, 27–49.
The polyploid species Herbertia lahue (Molina) Goldblatt is widespread in the grassland areas of southern America. In this study, genotypic and phenotypic variations among hexaploids and octoploids from H. lahue were assessed based on quantitative floral traits and ISSR markers. Additionally, gynoecium and breeding systems were analysed through anatomical analysis and hand-pollination experiments. Cytotypes are self-compatible, and the anatomy of the embryo sac remains similar in hexaploids and octoploids. AMOVA indicates that hexaploid populations are less genetically variable than octoploids (respectively 4 and 22 %, P \ 0.001), thus the largest source of genetic variation for H. lahue lies among populations (82 %, P \ 0.001), probably a combined effect of selfing, changes in population size, and local adaptation. Genetic variation between cytotypes was significant (16 %, P \ 0.001), though unclear segregation among cytotypes was discovered through PCoA. A Mantel test indicated no relationship between geographic and genetic distances (r = 0.04; P = 0.37), possibly as a result of the combined effect of the recurrent origin of polyploids, gene flow restricted by selfing, and poor capacity for seed dispersal. In terms of floral measurements, eight measures of perigonium, androecium, and gynoecium varied significantly between cytotypes, though no clear grouping was defined by PCA. Presumably, complex variation in floral characters across cytotypes may result in the absence of distinct grouping by PCA. The combined use of ISSR markers and morphological characters revealed a complex relationship between H. lahue cytotypes shaped by both intrinsic and extrinsic factors.
Plants of the family Iridaceae are well represented in the grassland vegetation of southern Brazil, occurring in the Pampa and Atlantic Forest biomes. Nevertheless, little is known about the taxonomy and evolution of Iridaceae species in southern Brazil. The main goal of this review is to compile published information about South American Iridaceae, and to discuss the evolution and genetic diversity of the family presenting our own research data in the light of the published literature. The main focus is on the genera Calydorea, Cypella, Herbertia, and Sisyrinchium. Aspects of reproductive system and of pollinator attraction are also discussed.
Resumo Esse artigo apresenta uma revisão sobre o uso de marcadores moleculares e citogenéticos aplicados à detecção da biodiversidade vegetal. São citados exemplos de aplicação destas ferramentas na investigação de táxons pertencentes às famílias de Monocotiledôneas, Iridaceae e Poaceae, que são alvo de pesquisa científica dos autores desse artigo. RESUMO -Para que as estratégias de melhoramento genético em espécies arbóreas tenham sucesso, o conhecimento da biologia reprodutiva da espécie envolvida é indispensável. Um dos fatores críticos ao sucesso reprodutivo das espécies do gênero Acacia é a presença de vetores para a polinização. Os objetivos deste trabalho foram identificar os visitantes florais em um plantio comercial de Acacia mearnsii De Wild. e quantificar as políades aderidas à superfície corporal dos insetos. As observações foram realizadas em uma torre localizada no plantio comercial durante o período de floração de 2002 e 2003. Os visitantes florais foram capturados no período diurno com o auxílio de rede entomológica e de um cesto aéreo instalado em um trator. No período noturno, os insetos foram capturados com o auxílio de armadilhas luminosas instaladas entre as copas florescidas. Foram observados insetos pertencentes às ordens Coleoptera, Hymenoptera, Diptera, Hemiptera e Lepidoptera. Os besouros da espécie Macrodactylus suturalis foram considerados como dispersores do pólen de acácia-negra, pela alta frequência de indivíduos presentes no povoamento estudado e pela grande quantidade de políades aderidas no corpo dos insetos capturados (X=229,36 políades/inseto). As abelhas da espécie Apis mellifera, embora tenha sido quantificada uma grande quantidade de políades nos indivíduos capturados (X=448,50 políades/ inseto), não foram observadas com frequência na área analisada. Uma das formas recomendadas para aumentar a frequência de abelhas e vespas nos plantios comerciais de acácia-negra é a manutenção de fontes de néctar e a introdução de caixas com abelhas A. mellifera.Palavras-chave: Acácia-negra, polinização e Macrodactylus suturalis. Evolutionary analyses have been widely used for evaluation of genetic diversity of natural populations and correlate these data to the fitness of the species, especially in the case of threatened species. Calydorea crocoides occurs in a restricted area at altitudes from 800 to 1500 m in southern Brazil and is considered endangered. A study assessing genetic diversity, cytogenetic features and ecological niche was performed aiming to characterize C. crocoides by multidisciplinary approaches. Molecular data highlighted that most of the total variation (76%; p < 0.001) was found within populations and the parameters of genetic diversity were high at the species level (PPB = 98.97%; I = 0.4319; h = 0.2821). Gene flow (Nm) was estimated in 0.97 individuals per generation. Cytogenetically, C. crocoides presents a bimodal karyotype and low asymmetry. DAPI banding pattern was uniform, but the CMA-signal evidenced a pericentric inversion in the population ESC688. The species presents high pollen viability and two different morphologies of pollen grains. Our data showed high levels of polymorphism maintained in this species that could ensure conservationist practices in which the main goal is to preserve the evolutionary potential of the species through the maintenance of genetic diversity.
RESUMO -O objetivo deste trabalho foi analisar a receptividade de estigmas de Acacia mearnsii De Wild. em 20 árvores da Área de Produção de Sementes (APS) da Fazenda Locatelli, da empresa SETA S.A., no município de Butiá (RS) e comparar os períodos de receptividade de estigmas e de viabilidade polínica em diversas fases da abertura floral. Foi utilizado como método de colorimetria o reagente de Baker para indicar a receptividade e a viabilidade. As análises foram conduzidas em delineamento inteiramente casualizado e a estatística χ 2 foi utilizada para verificar a significância dos eventos estudados. Foi observada receptividade no início da abertura floral (71%), alcançando a total receptividade (100%) em plena antese. A viabilidade polínica foi detectada no início da antese (77%) e em total abertura floral (88%). Na fase de senescência foram observados estigmas receptivos (50%) e políades viáveis (23%). Há sobreposição da fase de viabilidade masculina e de receptividade feminina, aumentando as chances de autopolinização. Na maioria das árvores analisadas (85%) foi observada a total receptividade (100%) nos estigmas amostrados. Em três árvores (15%) observou-se uma média inferior a 95% de estigmas receptivos. Não foi observada diferença significativa pelo teste do χ 2 na receptividade dos estigmas no germoplasma analisado, sugerindo que o genótipo não interfere diretamente para a receptividade dos estigmas.Palavras chave: acácia-negra, antese, polinização, germoplasma.
