Vor kurzem haben Lieb und Larnyar 1 ) die von mir angegebene jodometrische Methode zur Bestimmung der Homogentisinsäure im Harn 2 ) in einigen Punkten modifiziert. 8 ) Meine Methode gründet sich auf die leichte Oxydierbarkeit der Homogentisinsäure zu Chinonessigsäure durch Jodlösung nach Bicarbonatzusatz und die vollkommene Reversibilität dieser quantitativ verlaufenden Reaktion in schwefelsaurer Lösung.Einige der von den genannten Autoren vorgenommenen Änderungen, wie z. B. die Benutzung von n/20-statt n/10-Lösungen, erscheinen mir nicht sehr wesentlich. Nur zwei der von Lieb und Larnyar eingeführten Modifikationen sollen hier kurz besprochen werden.Die eine dieser näher zu erörternden Abänderungen besteht darin, daß Lieb und Larnyar die Bestimmung in sehr großer Verdünnung ausführen. Hierdurch wird die Reduktion der Chinonessigsäure zu Homogentisinsäure freilich so stark verlangsamt, daß die genannten Autoren nach dem Ansäuern mit Schwefelsäure 15 Minuten bis zur Titration warten müssen, während bei Innehaltung meiner ursprünglicben Arbeite Vorschrift die Reaktion in wenigen Sekunden beendet ist; nur bei sehr niedrigen Homogentisinsäurewerten kann sie etwas länger (einige Minuten) dauern.Während hiernach das Arbeiten in großer Verdünnung kaum als Verbesserung angesehen werden kann, ist eine solche in dem Zusatz von Jodkali gegeben: denn wenngleich meine ursprüngliche Methodik, die ohne Jodkaliumzusatz arbeitete, immerhin 99°/ 0 der zugesetzten Homogentisinsäure ergab, lieferte Jodkaliumzusatz unter Einhaltung der ursprünglich angegebenen Verdünnungsverhältnisse immer Werte, die weniger als l % von dem theoretischen abwichen. Dies geht aus den 6 Versuchen der untenstehenden Tabelle hervor, bei der die Endtitration sofort nach der Ansäuerung ausgeführt wurde. Man sieht, daß bei genügender Konzentration der Homogentisinsäure längeres Warten, wie es von Lieb und Larnyar empfohlen wird, unnötig ist.
Neben den am Glykogen und verschiedenartigen phosphorsäurehaltigen Substanzen bei der spezifischen Funktion des Muskels sich vollziehenden Abbau-und Aufbauvorgängen konnten in neuerer Zeit auch Änderungen im Kolloidzustande der Muskeleiweißkörper als Tätigkeitsfolge erkannt werden. Nachdem Ah r ah a m und Kahn 1 ) in einer aus dem hiesigen Institut veröffentlichten Arheit eine fortschreitende Verminderung der Fähigkeit des Muskels zur Synthese von Hexosediphosphorsäure unter bestimmten ionalen Bedingungen als Begleiterscheinung des fortschreitenden Absterbens aufgefunden hatten, machten Embden und Jpst 2 ) ganz ähnliche Beobachtungen nach starker Muskelermüdung. Während aber die Verminderung der Synthesefähigkeit des Muskels beim Absterben ein durchaus irreversibler Vorgang ist, erwies sich -die Verringerung der Synthesefähigkeit nach starker Ermüdung als ein unter geeigneten Erholungsbedingungen völlig reversibler Prozeß.Für die in den Arbeiten der genannten Autoren ausgesprochene Vermutung, daß die unter den geschilderten biologischen Verhältnissen beobchtete Verminderung der Synthesefähigkeit durch Veränderung im Zustande jener Kolloide bedingt wäre, die die Einwirkung der Ionen auf den fermentativen Muskelstoffwechsel vermitteln, konnten von H. J. Deuticke 8 ) wichtige experimentelle Anhaltspunkte gewonnen werden.Deuticke fand nämlich, daß sowohl beim Absterben wie bei der Ermüdung der Muskulatur Änderungen in der Löslichkeit der Muskelproteine auftreten, die in ihrer Richtung von der ionalen Zusammensetzung l )
