A drug–drug and drug–excipient interactions and compatibilities study was conducted for two fixed-dose combination (FDC) products containing olmesartan medoxomil (OLM)/hydrochlorothiazide (HCT) 20/12.5 mg and OLM/HCT 40/12.5 mg during their development including storage. The study consisted of the evaluation of samples retrieved during all stages of a real manufacturing process. Powder X-ray diffraction (PXRD), differential scanning calorimetry (DSC), thermogravimetry (TGA), Fourier transform infrared spectroscopy (FT-IR), and contact angle techniques were applied to the samples to determine interactions and incompatibilities. Dissolution tests and long-term stability studies were conducted to evaluate dosage form performance. Results showed weak solid–state interactions able to obtain a eutectic mixture of OLM and HCT while microcrystalline cellulose (MC) impacted the thermal stability of both drugs. Reliable dissolution and long-term stability tests confirmed that the interactions observed were not considered incompatibilities because they were not influenced by the performance of the final products.
Este proyecto de investigación busca contribuir con el desarrollo de una metodología pionera para la caracterización de tabletas farmacéuticas durante el proceso de recubrimiento por el método de aspersión aleatoria, utilizando diferentes técnicas. Se procura apoyar el desarrollo de una opción viable para sistemas de mayor complejidad, alto volumen de producción y gran procesamiento de información. Los resultados parciales obtenidos, permiten identificar patrones de comportamiento que pueden ser utilizados para facilitar el desarrollo de dispositivos y metodologías automatizables, aplicables a procesos similares, que puedan impactar en los procesos de recubrimiento como factores clave de la calidad y de la gestión productiva.
El avance tecnológico de las últimas décadas ha propiciado el desarrollo de nuevas metodologías para la caracterización de materiales de origen biológico. La espectroscopía Raman es utilizada principalmente en la caracterización de sustancias orgánicas e inorgánicas puras, sin embargo, recientemente se está utilizando para el estudio de organismos y estructuras moleculares de interés biológico. La principal dificultad en el análisis de estos materiales mediante la espectroscopía Raman es la fluorescencia que presentan las muestras al ser iluminadas con el haz de luz láser, pero mediante la técnica SERS (Surface Enhanced Raman Spectroscopy), es posible aumentar la señal Raman y reducir la interferencia de la fluorescencia. Para la aplicación de las técnicas SERS se pueden utilizar nanopartículas metálicas, principalmente oro y plata, de una adecuada distribución de tamaños. En este trabajo se presentan los resultados de la aplicación de técnica SERS a la caracterización espectroscópica del ADN de dos organismos diferentes, de un ser humano y de una bacteria del género Rickettsia, utilizando nanopartículas de oro y plata para la aplicación de la metodología.
Se realizó una caracterización de la fluorescencia, reflectancia y respuesta fisiológica al estrés hídrico en plántulas de Gmelna arborea Roxb. Para lo cual, se desarrolló un experimento durante 31 días con dos tratamientos: plantas testigo (se aplicó un régimen hídrico normal) y plantas en condición de estrés hídrico (a las que se les limitó en su totalidad el agua por el periodo del experimento). Para ambos métodos se implementaron clones de G. arborea y se evaluaron cada siete días hasta el final del experimento, se analizó la fotosíntesis, conductancia estomática, turgencia foliar, contenido relativo de clorofila, fluorescencia y reflectancia. Los resultados mostraron que la biomasa en las plantas con estrés se redujo en un 26,9 % siendo la cobertura foliar la que mostró mayor afectación (disminución del 29,1 %), la reducción de la fotosíntesis fue del 60,1 % (5,8 µmol m-2 s-1) con respecto a la planta testigo (14,6 µmol m-2 s-1). Para las variables conductancia estomática, turgencia y contenido relativo de clorofila el comportamiento fue de disminuir sus valores de un 20 a 60 % conforme el periodo de estrés se incrementó. Con respecto a la fluorescencia se determinaron dos patrones: de los 400 a 550 nm la fluorescencia se incrementó en las plantas estresadas, mientras que a partir de los 600 a 725 nm fueron las plantas testigo las que presentaron mayor excitación, en ambos rangos se presentaron valores máximos en los puntos correspondientes a clorofila a y b. Finalmente, la reflectancia mostró diferenciación entre los 460 y 770 nm siendo las plantas en condiciones de estrés las que presentaron una reflectancia mayor. El patrón obtenido de degradación fisiológica de las plantas estresadas se debe a la pérdida de agua del organismo que incide en cierre de estomas, aumento de producción de ácido abscísico, disminución de la síntesis de pared celular y limitaciones en la síntesis de proteínas.
<p class="p1">En muchos accidentes de tránsito es difícil establecer la responsabilidad por los daños materiales, las lesiones a las personas o las muertes, debido a que los conductores de los vehículos responsables abandonan el lugar de los hechos. Por eso, es fundamental, como parte de la recolección de la evidencia, intentar recuperar fragmentos de pintura automotriz. El estudio de las características físicas de estos fragmentos puede permitir la identificación del vehículo del que provienen. Sin embargo, con frecuencia, esta evidencia presenta un tamaño reducido, del orden de milímetros cuadrados o menor, lo cual requiere que el análisis de las diferentes propiedades físicas no ofrezca riesgo de alteración de la muestra. Para ello, resulta conveniente utilizar métodos como los que ofrecen las técnicas microscópicas y espectroscópicas. En el presente trabajo se muestran los resultados de los análisis de varios fragmentos de pintura automotriz, realizados con microscopio óptico y microscopía Raman, que podrían ser utilizados como evidencia forense.</p>
