BackgroundCardiac troponin-I assays have been validated in horses.’High-sensitivity’ cardiac troponin assays are now the standard in human cardiology. Objective: Appropriately validate the’high-sensitivity’ cardiac Troponin-T (hscTnT) assay for clinical use in horses, establish reference intervals, determine the biological variation, and demonstrate assay utility in selected clinical cases. Methods: Analytical validation of the Roche hscTnT assay included within- and between-run precision, linear dose response, limit of quantitation (LoQ), stability, and comparison with cTn-I (iSTAT). Reference intervals and biological variation were determined using adult, healthy, Non-Competition Horses (N = 125) and Racing-Thoroughbreds (N = 178). HscTnT levels were measured in two horses with cardiac pathology.ResultsThe hscTnT demonstrates acceptable within-run (L1 = 6.5 ng/L, CV 14.9 %, L2 = 10.1 ng/L, CV 8.7 %, L3 = 15.3 ng/L, CV 5.4 %) and between-run precision (L1 = 12.2 ng/L, CV 8.4 %, L2 = 57.0 ng/L, CV 8.4 %, L3 = 256.0 ng/L, CV 9.0 %). The assay was linear from 3 to 391 ng/L. The LoQ was validated at 3 ng/L. Samples demonstrated insignificant decay over freeze-thaw cycle. Comparison with cTnI assay showed excellent correlation (range: 8.0–3535.0 ng/L, R2 = 0.9996). Reference intervals: The upper 95th and 99th percentile of the hscTnT population distribution were 6.8 and 16.2 ng/L in Non-Competition Horses, and 14.0 and 23.2 ng/L in Racing-Thoroughbreds. Between-breed, diurnal effect, and between-day variation was below LoQ. Two clinical cases with presumed cardiac pathology had hscTnT levels of 220.9 ng/L and 5723.0 ng/L.ConclusionsThis benchmark study is the first to comply with CLSI guidelines, thus further establishing the performance characteristics of the hscTnT assay, and reference intervals in healthy horses. Two clinical cases demonstrated further the clinical utility of the assay.

BackgroundThe Heavy Horse Pull is a competition where teams of two horses pull an increasingly heavy sled for a short distance. Similar to human wrestlers, some horses may undergo rapid weight change in order to enter a lower weight category. The objectives were to study the physiological effects of this practice as well as of the pulling competition in draft horses.ResultsFifty horses were divided into light-, middle- and heavyweight categories based on their arrival weights and competed 1–3 days after. Body weight was measured upon arrival and pre-competition. Blood was sampled for chemistry and high sensitivity cardiac troponin T (hscTnT) at arrival, pre- and post-competition in 34, 26 and 20 horses, respectively. Body weight increased significantly between arrival and pre-competition for light (7.2% (Median: 62.8Kg (41.7–77.0)) and middle (8.6% (Median: 80.5Kg (62.7–90.9)) weight categories. Change in body weight was correlated (r = 0.69, p = 0.002) with competition ranking for middleweights. The ratios of weight pulled to team body weight were 2.7 (1.9–2.8), 2.6 (2.5–2.6) and 2.4 (2.2–2.5) for the lightweights, middleweights and heavyweights, respectively. Blood chemistry indicated hemoconcentration on arrival in the middleweight and lightweight horses. Hemoconcentration was not seen on arrival in some horses with marked rapid weight change. Overall, no chemistry parameter changed between pre- and post-competition. The hscTnT stayed within normal range post-competition.ConclusionsWhile horses arrived to the event with indications of hemoconcentration, they appeared to have sufficient time to rehydrate prior to competition, and the effects of the competition were reversible within 3 h.

This publication is a compilation of all Research Abstracts presented at the Ninth International Conference on Equine Exercise Physiology. Unlike previous ICEEP conferences there will not be a conference proceedings of full length manuscripts. These abstracts succinctly summarise a wide array of investigations relevant to the equine athlete, and will be useful to veterinarians and others involved in management of horses used for sport, work and competition. The abstracts encompass the topics of Applied PhysiologyIntroduction: The ability to accurately assess equine oxygen consumption (VO2) under field conditions has been limited by the need for unrestricted gas exchange.Methods: Two variations of a mask and an associated electronics control module (ECM) were designed to enable breath-by-breath measurement of airflows with two 8.0 cm diameter pneumotachometers located 7.5 cm in front of each narus and connected to differential pressure transducers mounted on the outside of the mask. The ECM was comprised of electronics for signal filtering to the flow transducers, an oxygen sensing cell, and an analog-to-digital converter all powered by a lithium-ion battery. The battery also powered a pump connected to gas sampling ports between the nares and pneumotachometers. Airflow and oxygen content of inspired and expired gases were recorded through the ECM and electronically transferred to a notebook. VO2 was determined from these recordings by an operator using a customized software analysis program. One mask encased the lower head (E). The other left the jaw free so horse could wear a bit and be ridden (R). Multiple treadmill exercise tests were undertaken by 6 horses to measure VO2max and blood gases. Each mask was worn twice and results compared to those from an open flow-through system (O) by 2-way RMANOVA (P<0.05). Utility of the system was evaluated using the intraclass correlation coefficient of 4 independent raters.Results: Blood gases and VO2max (152.0 ± 4.0 (mean ± SEM; O), 147.7 ± 4.3 (E), 150.7 ± 3.3 (R) ml/(kg.min) were not different between masks. VO2 measures were reproducible for each mask. Agreement between the 4 raters was excellent (intraclass correlation coefficient = 0.99).Conclusions: Masks capable of measuring VO2 during field exercise were developed, tested and found accurate by multiple users.Ethical Animal Research: Studies performed were approved by the Institution's Animal Care and Use Committee (protocol #3807). BackgroundA “high‐sensitivity” cardiac troponin‐T (hscTnT) assay recently has been validated for use in horses and is a specific biomarker of myocardial damage. Postexercise release kinetics of cTnT utilizing the hscTnT assay have yet to be established in horses.ObjectivesTo determine: (1) cTnT release kinetics in racing Thoroughbreds after a high‐intensity 5/8th mile Chuckwagon race; (2) the effects of age on pre‐ and postrace cTnT concentrations; and (3) sampling guidelines for clinicians evaluating horses presenting after exercise.AnimalsSamples were obtained from 38 Thoroughbred geldings aged 5–16 years before racing and immediately, 2, 3, 4, 6, 12, and 24 hour postrace.MethodsProspective, observational study with convenience sampling. A fifth‐generation hscTnT assay was used for plasma sample analysis, and concentrations were compared at all time‐points. Correlations were determined between cTnT concentrations and age. Biochemistry analysis was performed to assess rhabdomyolysis, renal failure, and exercise‐induced dehydration.ResultsAll horses with measureable cTnT concentrations had significant postexercise increases in cTnT with a median peak (8.0 ng/L) at 3‐hour postrace. All horses had peak postexercise cTnT concentrations 2‐ to 6‐hour postrace ≤ the 99th percentile upper reference limit of 23.2 ng/L, after which all cTnT concentrations decreased until returning to baseline by 12–24 hours. There was no correlation over time between cTnT concentrations and age.Conclusions and Clinical ImportanceIn racing Thoroughbreds completing short‐duration, high‐intensity Chuckwagon races, cTnT concentrations are expected to be increased 2‐ to 6‐hour postrace and to decrease by 12–24 hours while remaining ≤23.2 ng/L throughout. This study contributes to establishing guidelines for clinical use of the hscTnT assay in exercising horses.
