Zusammenfassung:Aus 5-Hydroxymethyl-uracil entsteht glatt 5-Benzyloxymethyl-uracil (VII). Diese Verbindung eignet sich gut zur Darstellung von Nucleosiden mit empfindlichen Zuckern. -Das nach einer verbesserten Methode gewonnene Quecksilbersalz IX von Benzyloxymethyl-uracil kondensiert mit 3.5-Bis-[p-methyl-benzoyl]-2-desoxy-Dribofuranosylchlorid unter milden Bedingungen (Raumtemperatur) in sehr guter Ausbeute und liefert beide anomeren, vollsubstituierten Nucleoside in kristallisierter Form. Das ß-Nucleosid X erhält man quantitativ (Ausbeute 49 %), oc-Nucleosid: 35% Ausbeute. Der Konstitutionsbeweis wird auf 3 voneinander unabhängigen Wegen geführt: Überführung in Thymidin, NMR-Spektroskopie, Messung der optischen Rotationsdispersion (CoT-Summary: Benzylation of 5-hydroxymethyl-uracil to 5-benzyloxymethyl-uracil (VII) readily takes place. This compound is well suited for use in the preparation of nucleosides with labile sugars. -The mercury salt IX can be obtained according to an improved procedure. It condenses with 3,5-di-/?methy 1 -benzoyl -2 -deoxy -D -ribofuranosylchloride under mild conditions (room temperature) to give crystalline, completely substituted, anomeric nucleosides in high yields. The more interesting ß-nucleoside X is quantitatively formed (based on optimal yields in regard to the "trans"-mlo), the oc-nucleoside in 35 % yield. The chemical structure has been proved by 3 independent means: conversion to thymidine, NMR-spectroscopy and measurement XON-Effekt). Katalytische Hydrierung und Entacylierung ergeben die freien Desoxyriboside. Es gelingt so, in präparativem Maßstab 5-Hydroxymethyl-2'-desoxy-uridin kristallisiert mit einer Ausbeute von ca. 45% (bezogen auf IX) darzustellen. Das entsprechende -Anomere isoliert man ebenfalls kristallisiert (Ausbeute 23-32%). Es war, wie die gesamte -Reihe, bisher unbekannt. -Aus den a-und ß-Benzyläther-nucleosiden lassen sich leicht Verbindungen gewinnen, die für weitere Synthesen, z. B. von Nucleotiden geeignet sind. 5-Chlormethyluracil-nucleosid (XX) reagiert mit nucleophilen Substanzen, die so mit dem Thymidinrest als einem beweglichen Träger (mobile carrier) verknüpft werden.of optical rotatory dispersion (CorroN-effect). Catalytic hydrogenation and deacylation produces free deoxyribosides. In this simple manner naturally occuring crystalline 5-hydroxymethyl-2'-deoxyuridine may be obtained on a preparative scale (approx. 45 % yields based on IX). The respective crystalline a-anomer (23-32 %) as well as the whole series were not previously known. -Conversion of and benzylether nucleosides readily leads to valuable synthetic intermediates e. g. for the preparation of nucleotides. 5-Chloromethyl uracil nucleoside XX reacts with nucleophilic compounds, e. g. alcohols, amines. Das Gangliosid aus Rinderhirn ließ sich chromatographisch in zwei kristallisierende Komponenten zerlegen. Das schneller wandernde Gangliosid (G2) ist aus je 1 Mol Stearinsäure, Sphingosin, Glucose, Galaktose, Acetylgalaktosamin und Lactaminsäure (N-Acetyl-neuraminsäure) aufgebaut. Die Verknüpfungsweise der 6 Bausteine wird aus Ergebnissen der Permethylierung mit CH3J und BaO + Ba(OH)2 bzw. SrO + Sr(OH)2 in Dimethylformamid und partieller Acetolysen abgeleitet. Dabei zeigt sich, daß ein aus der Kuhmilch schon früher isoliertes Trisaccharid auch Baustein des Gangliosids G2 aus Rinderhirn ist. Aus Frauenmilch konnten neue saure Oligosaccharide gewonnen werden. Das Gangliosid G1 ist komplizierter gebaut und vermutlich noch nicht einheitlich. — An physiologischen Eigenschaften der Ganglioside G1 und G2 werden besprochen: die Steigerung der Körpertemperatur; die Erhöhung der Resistenz gegenüber Coli-Infektionen; das Bindungsvermögen für Tetanustoxin; die Spaltbarkeit durch Influenzavirus und durch das receptor destroying enzyme (RDE)
Aus 5-Hydroxymethyl-uracil (1) entsteht bei der Chlorierung mit einer Mischung von Phosphoroxidchlorid und Phosphorpentachlorid in guter Ausbeute 2.6-Dichlor-5-chlormethyl-pyrimidin (2). Dieses reagiert mit den verschiedensten Verbindungen, die nucleophile Gruppen enthalten. Die drei Chlor-Atome besitzen eine deutlich abgestufte Reaktionsfähigkeit: Cl-aliphatisch (allylständig) > Cl an C-6 > Cl an C-2. So lassen sich gezielt mono-, di- und trisubstituierte Derivate, z. B. Alkyläther, Phenoläther, Thioäther, Amine und Azide darstellen. Einige dieser Verbindungen besitzen beträchtliche bakteriostatische und fungistatische Wirksamkeit. Aus Rhodizonsäure-1.4-diimid wurde durch katalytische Hydrierung in schwefelsaurer Lösung das Sulfat eines 1.4-Diamino-2.3.5.6-tetrahydroxy-cyclohexans erhalten. In gleicher Weise ließ sich Triamino-phloroglucin in ein 1.3.5-Triamino-2.4.6- trihydroxy-cyclohexan überführen
dieses Aza-Analogen des 6-Dimethyl-amino-fulvens [?] hin. Die Hydrolyse von (3) mit 2 N H2S04 fuhrt uber (4) unmittelbar zum dimeren Cyclopentadienon (5) [3]. Elektrophile Agenzien substituieren (3) im Fiinfring. Beispielsweise liefert die Vilsmeier-Formylierung von (3) mit Dimethylformamid und Phosphoroxychlorid iiber das isolierbare Iinmonium-Salz (6) (Pcrchlorat: orange Nadeln, F p = 135 'C, Zersetzung) den gleichfalls bestandigen Aldehyd (7) (orange Nadeln. Fp = 65°C; hma, = 251.8 (4,18); 3733 (1,35 mil) (Ig E) (in Methanol)).Max-Planck-lnstitut fur Medizinische Forschung, Heidelberg lnstitut fur Chemie 5-Hydroxymethyl-2'-desoxycytidin (9) ersctzt in der DNS verschiedener Bakteriophagen das gesamte Desoxycytidin und ist daher von besonderem Interesse. -Benzylierung von 5-Hydroxymethylcytosin zum Benzylather (I) (Fp = 250 OC (Zers.), Ausb. 90%) und anschlieDende Acylierung, z. B. mit Benzoylchlorid in Pyridin, fiihrt zu (2) (Fp = 174-175T; > 80%). Die Auftrennung gelingt durch Loslichkeitsunterschiede. F-Anomer (4) (Fp = 170-171 "C; [a]: = -52" in Chloroform; 48%). a-Anomer (5) (Fp = 128-129°C; [a]'," = -114" in Chloroform; 35%). Kochen von (4) in Methanol gibt (6) (Fp = 163-164°C; VERSAMMLUNGSBERICHTE[.1]?," = 42,7 " in Chloroform; 80-85 7:). WPhrend katalytische Hydrierung von (4) zu 86% (7) (Fp = 151-152°C.[a]';' = -64,5 in Chloroform) liefert, gewinnt man aus ( 4 ) und 16) durch Abspaltung der Acylreste mit Na-Methylat-Losung (8) (Fp = 198-199'C; [a]: = +33O in Dimethylformamid; 91 :J. Katalytische Hydrierung von (8) und Behandlung von (7) mit Na-Methylat-Losung fuhrt zum identischen freien (9) (Fp = 203 "C (Zers.); [a]' ," = +51 ' in H2O; Ausb. 35 % bez. auf (3), RF 0,16-0,18 (FlieRmittel: n-Butanol/HZO (gesatt.), 0,3 I -0,33 n-Propanol/HzO (5:l). aufsteigende Methode, UV-Absorption bei pH 1 : Amax 283 mp. 212 m!L, E~~~ 12,6.103, 11,9.103, Amin 243 m p , Emin 1,6.103, A 2501260 = 0,43, A 2801260 = 2,68).f 4) I (61 (7) (81 R1= H Rt = Benzoyl R I = R 3 = H RZ = Benzyl R z = H RZ = Benzyl R' = p-Toluyl R3 = p-Toluyl Der Konstitutionsbeweis von (4) wurde durch Abbau zu 8-Thyrnidin erbracht. Die Zuordnung der Konfiguration gelang zusltzlich durch Analyse des N MR-Spektrums. Legt man lediglich Wert auf die Darstellung von unsubstituiertem (9), so kann man von dem zweifach acylierten (z. B. benzoylierten) (20) (Fp = 154-155°C; 80%) ausgehen, dessen Kondensation zum geschiitzten Nucleosid (Fp = 181 --I82 "C) ebenfalls glatt verlauft. Abspaltung aller Acyle liefert in einem Schritt (9). Wir berichten demnachst uber die Darstellung von anderen Hydroxymethylcytosin-nucleosiden und VOn -nucleotiden.Eingegangen am 15.
