Mikroalga diatom dianggap sebagai organisme hidup yang memiliki banyak manfaat dan telah menarik banyak perhatian dari peneliti-peneliti dunia. Dalam beberapa tahun ini, diatom juga telah digunakan sebagai bahan nutrasetikal dan sumber obat dalam industri farmasi. Penduduk lokal di Desa Pabean Udik, Kabupaten Indramayu, sebagian besar memiliki profesi sebagai nelayan pembudidaya, dimana produksi perikanan budidaya berkontribusi cukup besar pada pendapatan daerah. Namun, salah satu kendala yang dialami oleh pembudidaya adalah adanya serangan penyakit yang disebabkan oleh bakteri patogen yang seringkali diatasi dengan penggunaan antibiotik sintetik yang tidak ramah lingkungan. Oleh karena itu, dalam penelitian ini, kami melakukan sosialisasi terhadap peningkatan kesadaran masyarakat lokal di desa Pabean Udik tentang potensi diatom sebagai produk alami dengan multi-manfaat, terutama sebagai agen antibakteri alami dalam praktik budidaya perikanan laut. Dalam studi ini, pendidikan berbasis masyarakat dengan beberapa kegiatan seperti ceramah, diskusi, video interaktif dan sesi doorprize dilakukan. Selain itu, kuesioner pre-test dan post-test dilakukan sebagai alat untuk mengidentifikasi kesadaran peserta dan untuk mengukur indikator tujuan. Hasil survey yang diperoleh menunjukkan peningkatan pemahaman peserta hingga 20%. Kami mengamati bahwa selama diskusi peserta juga menunjukkan kesediaan mereka untuk meningkatkan pengetahuan mereka tentang potensi pemanfaatan diatom untuk meminimalkan risiko infeksi patogen. Oleh karena itu, hasil ini menunjukkan bahwa penelitian ini berhasil meningkatkan pemahaman masyarakat lokal tentang potensi manfaat diatom dan meningkatkan kesadaran masyarakat untuk menggunakan diatom untuk banyak tujuan, terutama dalam menggantikan obat antimikroba yang tidak ramah lingkungan.
Sektor kelautan dan perikanan merupakan salah satu sumber andalan dalam produksi pembangunan perikanan di Indonesia. Udang merupakan salah satu komoditas unggulan utama dalam menunjang produksi pendapatan devisa non migas. Untuk mencapai target produksi sesuai dengan yang diharapkan, berbagai permasalahan menghambat upaya peningkatan produksi tersebut, antara lain kegagalan produksi akibat penyakit oleh bakteri Vibrio, sehingga penyakit ini dikenal sebagai ‘vibriosis’. Salah satu pencarian senyawa yang bersifat antibakteri adalah dengan cara melakukan penapisan terhadap senyawa-senyawa alam. Salah satu sumber senyawa alam adalah dari tumbuhan Tephrosia vogelii. Penelitian ini bertujuan untuk mengidentifikasi dan mengisolasi senyawa dari tumbuhan T. vogelii sebagai sumber senyawa antibakteri terhadap dua bakteri Vibrio yaitu Vibrio alginolitycus dan Vibrio harveyi. Bahan ekstrak dibuat dengan metode maserasi menggunakan pelarut aseton, sementara uji antibakteri dilakukan dengan metode difusi agar secara in vitro. Deguelin dan tefrosin kemudian diisolasi dari ekstrak aseton daun T. vogelii. dengan metode difusi agar senyawa deguelin memberikan nilai inhibisi 6,3 mm terhadap V. harveyi dan 6,2 mm terhadap V alginolitycus. Sedangkan tefrosin memberikan nilai inhibisi 6,3 mm terhadap V. harveyi dan 6,6 mm terhadap V alginolitycus. Ini adalah evaluasi antibakteri pertama dari deguelin dan tefrosin terhadap dua bakteri yang diuji.
Perairan Sungai Citarum banyak memperoleh limpasan limbah antropogenik dari darat yang dapat menimbulkan pencemaran lingkungan. Untuk mengatasi masalah pencemaran tersebut, bakteri dapat dimanfaatkan sebagai agen bioremediasi. Hingga saat ini belum tersedia informasi komunitas bakteri di Perairan Sungai Citarum, khususnya dari wilayah hilir Muara Gembong. Penelitian ini bertujuan untuk mengidentifikasi komunitas, kelimpahan, dan keanekaragaman bakteri dari Perairan Muara Gembong yang telah diperkaya dengan pendekatan metagenomik, serta analisis fungsional dengan melihat profil protein yang dimiliki dari bakteri yang teridentifikasi yang berpotensi sebagai agen bioremediasi. Pengambilan sampel air dilakukan di dua lokasi (A1 dan A2) dengan karateristik perairan yang berbeda. Sampel air estuari diperkaya dengan 0,2x Nutrient Broth. Sekuensing dilakukan dengan metode Next Generation Sequencing (NGS) menggunakan amplikon sekuensing platform Novaseq Illumina, dengan primer 16S rRNA region V3-V4 (341F dan 806R). Hasil identifikasi menunjukkan terdapat 37 genera, 19 famili, 13 ordo, 7 kelas, dan 4 filum bakteri dengan 49 OTU untuk sampel A1 dan 48 OTU untuk sampel A2. Kelimpahan tertinggi diperoleh pada kelas Gammaproteobacteria (99,82%) yang didominasi oleh genus Vibrio. Keanekaragaman bakteri pada kedua sampel diperkaya ini tergolong sedang dengan tingkat dominasi tinggi. Analisis fungsional menunjukkan kemampuan bakteri yang teridentifikasi mampu mendegradasi benzoat, dioxin, asam lemak, metana, naftalen, nitrotoluen, PAH, dan sulfur dengan protein dan enzim yang terlibat dalam metabolisme tersebut. Penelitian ini menunjukkan bakteri yang teridentifikasi pada Perairan Muara Gembong mempunyai potensi bioremediasi untuk cemaran lingkungan, seperti polutan organik dari logam berat.
This research aims to determine the most effective concentration level of basil leaves extract in preserving nile tilapia filet at low temperatures (5 – 10oC) to obtain optimal shelf life. This research was conducted at the Central Laboratory and Fishery Products Processing Laboratory, Faculty of Fisheries and Marine Sciences, Padjadjaran University started from June until October 2022. The research method used was an experimental method with four treatments by duplo. Fish filets were treated by soaking basil leaves extract with concentrations of 0%, 20%, 25%, and 30% for 30 minutes and stored at low temperature (5 – 10oC). Observations were made on days 1st, 3rd, 5th, 7th, 8th, and 9th for nile tilapia filets with a concentration of 0% while treatment of basil leaves extract with concentrations of 20%, 25%, and 30% was carried out on days 1st, 3rd, 5th, 7th, 8th, 9th, 10th, 11th, and 12th. The parameters observed in this research included the calculation of the total number of bacteria (TPC) and degree of acidity (pH). The results showed that the use of basil leaves extract with a concentration of 25% was the most effective concentration because it was able to extend the shelf life until the 11th day with the number of bacteria 4.4 x 105 cfu/g and pH value of 6.65.
