Fentanyl analogs constitute a particularly dangerous group of new psychoactive compounds responsible for many deaths around the world. Little is known about their metabolism, and studies utilizing liquid chromatography quadrupole time-of-flight mass spectrometry (LC-QTOF-MS) analysis of hepatocyte incubations and/or authentic urine samples do not allow for determination of the exact metabolite structures, especially when it comes to hydroxylated metabolites. In this study, seven motifs (2-, 3-, 4- and β-OH as well as 3,4-diOH, 4-OH-3-OMe and 3-OH-4-OMe) of fentanyl and five fentanyl analogs, acetylfentanyl, acrylfentanyl, cyclopropylfentanyl, isobutyrylfentanyl and 4F-isobutyrylfentanyl were synthesized. The reference standards were analyzed by LC-QTOF-MS, which enabled identification of the major metabolites formed in hepatocyte incubations of the studied fentanyls. By comparison with our previous data sets, major urinary metabolites could tentatively be identified. For all analogs, β-OH, 4-OH and 4-OH-3-OMe were identified after hepatocyte incubation. β-OH was the major hydroxylated metabolite for all studied fentanyls, except for acetylfentanyl where 4-OH was more abundant. However, the ratio 4-OH/β-OH was higher in urine samples than in hepatocyte incubations for all studied fentanyls. Also, 3-OH-4-OMe was not detected in any hepatocyte samples, indicating a clear preference for the 4-OH-3-OMe, which was also found to be more abundant in urine compared to hepatocytes. The patterns appear to be consistent across all studied fentanyls and could serve as a starting point in the development of methods and synthesis of reference standards of novel fentanyl analogs where nothing is known about the metabolism.
The effect of shifting Spodoptera littoralis larvae from a host plant to another pointed out that pupal weight, growth index, female longevity and fecundity were affected. Host plants considered were okra, tomato and castor oil plants. Larvae were reared for three successive generations under laboratory conditions (25-30˚C, 70%R.H. and 12D:12L) on each of the three chosen plants, and then shifted to each of the other two hosts.Larvae reared continuously on castor oil leaves, okra and tomato leaves then shifted to either one of the other plants induced a highly significant decrease in the mean pupal weight. Shifting from the favourable host (castor oil leaves) to either okra or tomato leaves, highly significantly decrease the growth rate and the mean number of eggs deposited by the female. Shifting from castor oil leaves to either okra or tomato induced insignificant increase in the female longevity. Phytochemical analysis showed that, castor oil leaves, possess a high value of total carbohydrates, total proteins and nitrogen meanwhile the phenolic compounds were present at a low value level. Biochemical analysis showed that both essential and non essential amino acid were higher in the hemolymph of the last larval instars of S. littoralis reared on castor oil leaves. A noticeable decrement was detected in the amino acid level of larvae fed on okra leaves followed by more decrement in larvae fed on tomato leaves.
Unsustainable agricultural practices that minimize soil organic matter can promote the removal of heavy metal pollutants into the food chain. Such polluted soils can release contaminants into the groundwater, which leads to accumulation in plant tissue that is transferred to animals, birds, insects, and humans. Biomonitors of soil pollution with heavy metals can be identified by the ground beetles Calosoma chlorostictum (Coleoptera: Carabidae) as bioindicators of soil quality and its yield sustainability. The experiment was performed on two sites in Zagazig City (30.62° N, 31.44° E), Egypt. The physicochemical parameters indicated that soil moisture and organic matter had the highest differences in the polluted agricultural soil compared to the reference soil. However, there were no significant differences in chloride content. The atomic absorption analysis exhibited the highest concentration recorded for arsenic (As) and the lowest for selenium (Se) in the polluted soil and the insect’s midgut. Meanwhile, the differences between heavy metal concentrations in the total soil and midgut of C. chlorostictum from current sites indicated that the highest differences were in aluminum (Al) and mercury (Hg), while arsenic (As) and cadmium (Cd) were the lowest. Furthermore, the correlation between heavy metal concentrations in the soil and insect midgut was highest in As, while the lowest correlation was noticed in Al. We used transmission electron microscopy (TEM) that showed a more considerable disturbance in the C. chlorostictum midgut epithelial layer collected from the agricultural area than in the insects collected from the reference area. Evident ultrastructural alterations showed a rupture and distortion of microvilli, destruction of the columnar and regenerative cells, large separation between epithelial cells, and stretching of the cellular axis, as a result of which the lumen became very narrow. Moreover, a lot of vacuoles with little enzyme secretion were observed in the columnar epithelial cells. In addition, other manifestations due to pollution with heavy metals such as a pyknotic nucleus with abnormal chromatin, cytoplasmic vacuolization, disruptions, and vacuolation of mitochondria were detected, as well as the appearance of electron-dense vesicles, a lot of lysosomes, large myelin figures, and dilation of the rough endoplasmic reticulum on account of soil contamination. Potential counteractive health influence in such applications could be avoided if the soil was adequately treated.
