To understand the functional significance of skeletal muscle anatomy, a method of quantifying local shape changes in different tissue structures during dynamic tasks is required. Taking advantage of the good spatial and temporal resolution of B-mode ultrasound imaging, we describe a method of automatically segmenting images into fascicle and aponeurosis regions and tracking movement of features, independently, in localized portions of each tissue. Ultrasound images (25 Hz) of the medial gastrocnemius muscle were collected from eight participants during ankle joint rotation (2° and 20°), isometric contractions (1, 5, and 50 Nm), and deep knee bends. A Kanade-Lucas-Tomasi feature tracker was used to identify and track any distinctive and persistent features within the image sequences. A velocity field representation of local movement was then found and subdivided between fascicle and aponeurosis regions using segmentations from a multiresolution active shape model (ASM). Movement in each region was quantified by interpolating the effect of the fields on a set of probes. ASM segmentation results were compared with hand-labeled data, while aponeurosis and fascicle movement were compared with results from a previously documented cross-correlation approach. ASM provided good image segmentations (<1 mm average error), with fully automatic initialization possible in sequences from seven participants. Feature tracking provided similar length change results to the cross-correlation approach for small movements, while outperforming it in larger movements. The proposed method provides the potential to distinguish between active and passive changes in muscle shape and model strain distributions during different movements/conditions and quantify nonhomogeneous strain along aponeuroses.
Skeletal muscle contains many muscle fibres that are functionally grouped into motor units. For any motor task there are many possible combinations of motor units that could be recruited and it has been proposed that a simple rule, the 'size principle', governs the selection of motor units recruited for different contractions. Motor units can be characterised by their different contractile, energetic and fatigue properties and it is important that the selection of motor units recruited for given movements allows units with the appropriate properties to be activated. Here we review what is currently understood about motor unit recruitment patterns, and assess how different recruitment patterns are more or less appropriate for different movement tasks. During natural movements the motor unit recruitment patterns vary (not always holding to the size principle) and it is proposed that motor unit recruitment is likely related to the mechanical function of the muscles. Many factors such as mechanics, sensory feedback, and central control influence recruitment patterns and consequently an integrative approach (rather than reductionist) is required to understand how recruitment is controlled during different movement tasks. Currently, the best way to achieve this is through in vivo studies that relate recruitment to mechanics and behaviour. Various methods for determining motor unit recruitment patterns are discussed, in particular the recent wavelet-analysis approaches that have allowed motor unit recruitment to be assessed during natural movements. Directions for future studies into motor recruitment within and between functional task groups and muscle compartments are suggested.
SUMMARY Motor units are generally considered to follow a set, orderly pattern of recruitment within each muscle with activation occurring in the slowest through to the fastest units. A growing body of evidence, however, suggests that recruitment patterns may not always follow such an orderly sequence. Here we investigate whether motor unit recruitment patterns vary within and between the ankle extensor muscles of the rat running at 40 cm s-1 on a level treadmill. In the past it has been difficult to quantify motor unit recruitment patterns during locomotion; however, recent application of wavelet analysis techniques has made such detailed analysis of motor unit recruitment possible. Here we present methods for quantifying the interplay of fast and slow motor unit recruitment based on their myoelectric signals. Myoelectric data were collected from soleus, plantaris and medial gastrocnemius muscles representing populations of slow, mixed and fast fibres, respectively, and providing a good opportunity to relate myoelectric frequency content to motor unit recruitment patterns. Following wavelet transformation, principal component analysis quantified signal intensity and relative frequency content. Significant differences in signal frequency content occurred between different time points within a stride (P<0.001). We optimised high- and low-frequency wavelets to the major signals from the fast and slow motor units. The goodness-of-fit of the optimised wavelets to the signal intensity was high for all three muscles (r2>0.98). The low-frequency band had a significantly better fit to signals from the soleus muscle (P<0.001), while the high-frequency band had a significantly better fit to the medial gastrocnemius(P<0.001).
Background: Three commonly used non-invasive protocols are implemented to estimate the timing at which PHV most likely occurs. Accurate estimation of circumpubertal years can aid in managing training load of adolescent athletes. Aim: Three protocols were compared against observed age at PHV: an estimate of 13.8 ±1.0 year -generic age at PHV (from longitudinal measures); an estimate based on the maturity offset equation, predicted age at PHV ±1.0 year; a window of PHV based on 85 -96% of predicted adult height at time of observation. Subjects and Methods: A final sample of 23 (from 28) adolescent male participants were selected from the academy of an English Premier League club. Anthropometric measures were collected across five playing seasons; age at PHV was estimated with Super-Imposition by Translation and Rotation (SITAR). The three protocols were compared based on measures at 13.0 years. Results and Conclusions: An age window based on predicted maturity offset did not improve estimation of PHV compared to generic age method; however, the percentage of predicted adult height window showed improvement in performance shown by the following results. Predicted age at PHV correctly assigned 15 participants (65%) as experiencing PHV, while the percentage height correctly assigned 17 participants (74%). Generic age and predicted age at PHV correctly predicted observed age at PHV for 14 participants (61%), percentage of adult height window correctly predicted 22 participants (96%).
B-mode ultrasound is often used to quantify muscle architecture during movements. Our objectives were to 1) systematically review the reliability of fascicle length (FL) and pennation angles (PA) measured using ultrasound during movements involving voluntary contractions; 2) systematically review the methods used in studies reporting reliability, discuss associated challenges, and provide recommendations to improve the reliability and validity of dynamic ultrasound measurements; and 3) provide an overview of computational approaches for quantifying fascicle architecture, their validity, agreement with manual quantification of fascicle architecture, and advantages and drawbacks. Three databases were searched until June 2019. Studies among healthy human individuals aged 17–85 yr that investigated the reliability of FL or PA in lower-extremity muscles during isoinertial movements and that were written in English were included. Thirty studies ( n = 340 participants) were included for reliability analyses. Between-session reliability as measured by coefficient of multiple correlations (CMC), and coefficient of variation (CV) was FL CMC: 0.89–0.96; CV: 8.3% and PA CMC: 0.87–0.90; CV: 4.5–9.6%. Within-session reliability was FL CMC: 0.82–0.99; CV: 0.0–6.7% and PA CMC: 0.91; CV: 0.0–15.0%. Manual analysis reliability was FL CMC: 0.89–0.96; CV: 0.0–15.9%; PA CMC: 0.84–0.90; and CV: 2.0–9.8%. Computational analysis FL CMC was 0.82–0.99, and PA CV was 14.0–15.0%. Eighteen computational approaches were identified, and these generally showed high agreement with manual analysis and high validity compared with phantoms or synthetic images. B-mode ultrasound is a reliable method to quantify fascicle architecture during movement. Additionally, computational approaches can provide a reliable and valid estimation of fascicle architecture.
It has recently been shown that motor units in human medial gastrocnemius (MG), activated during standing, occupy relatively small territories along the muscle’s longitudinal axis. Such organisation provides potential for different motor tasks to produce differing regional patterns of activity. Here, we investigate whether postural control and nerve electrical stimulation produce equal longitudinal activation patterns in MG. Myoelectric activity, at different proximal–distal locations of MG, was recorded using a linear electrode array. To ensure differences in signal amplitude between channels did not result from local, morphological factors two experimental protocols were completed: (i) quiet standing; (ii) electrical stimulation of the tibial nerve. Averaged, rectified values (ARVs) were calculated for each channel in each condition. The distribution of signals along electrode channels was described using linear regression and differences between protocols at each channel determined as the ratio between mean ARV from standing: stimulation protocols. Ratio values changed systematically across electrode channels in seven (of eight) participants, with larger values in distal channels. The distribution of ARV along MG therefore differed between experimental conditions. Compared to fibres of units activated during MG nerve stimulation, units activated during standing may have a tendency to be more highly represented in the distal muscle portion.
SUMMARYMammalian skeletal muscles are composed of a mixture of motor unit types, which contribute a range of mechanical and physiological properties to the muscle. For a muscle to effectively contribute to smooth, co-ordinated movement it must activate an appropriate number and combination of motor units to generate the required force over a suitable time period. Much evidence exists indicating that motor units are activated in an orderly fashion, from the slowest through to the fastest. A growing body of evidence, however, indicates that such a recruitment strategy does not always hold true. Here we investigate how motor unit recruitment patterns were influenced by changes in locomotor velocity and incline. Kinematics data and myoelectric signals were collected from three rat ankle extensor muscles during running on a treadmill at nine velocity and incline combinations. Wavelet and principal component analysis were used to simultaneously decompose the signals into time and frequency space. The relative frequency components of the signals were quantified during 20 time windows of a stride from each locomotor condition. Differences in signal frequency components existed between muscles and locomotor conditions. Faster locomotor velocities led to a relative increase in high frequency components, whereas greater inclines led to a relative increase in the low frequency components. These data were interpreted as representing changes in motor unit recruitment patterns in response to changes in the locomotor demand. Motor units were not always recruited in an orderly manner, indicating that recruitment is a multi-factorial phenomenon that is not yet fully understood.
Denervation of selected ankle extensors in animals results in locomotor changes. These changes have been suggested to permit preservation of global kinematic characteristics of the hindlimb during stance. The peak ankle joint moment is also preserved immediately after denervation of several ankle extensors in the cat, suggesting that the animal’s response to peripheral nerve injury may also be aimed at preserving ankle mechanical output. We tested this hypothesis by comparing joint moments and power patterns during walking before and after denervation of soleus and lateral gastrocnemius muscles. Hindlimb kinematics, ground reaction forces and electromyographic activity of selected muscles were recorded during level, downslope (–50%) and upslope (50%) walking before and 1–3 weeks after nerve denervation. Denervation resulted in increased activity of the intact medial gastrocnemius and plantaris muscles, greater ankle dorsiflexion, smaller knee flexion, and the preservation of the peak ankle moment during stance. Surprisingly, ankle positive power generated in the propulsion phase of stance was increased (up to 50%) after denervation in all walking conditions (p < 0.05). The obtained results suggest that the short-term motor compensation to denervation of lateral gastrocnemius and soleus muscles may allow for preservation of mechanical output at the ankle. The additional mechanical energy generated at the ankle during propulsion can result, in part, from increased activity of intact synergists, the use of passive tissues around the ankle and by the tendon action of ankle two-joint muscles and crural fascia.
