The Eocene-Oligocene transition (EOT) is a period of ∼790 kyr (34.2-33.5 Ma) that encompasses the chronostratigraphic Eocene-Oligocene Boundary at 33.9 Ma (Coxall & Pearson, 2007;Gradstein & Ogg, 2012;Hutchinson et al., 2021). The EOT marks the shift from the greenhouse conditions of the Eocene to the icehouse conditions of the Oligocene. A two-step increase in benthic foraminiferal oxygen isotopes (δ 18 O benthic ) of ∼1.5‰ (Coxall et al., 2005;Zachos et al., 2001), the latter and larger of which is now referred to as the Earliest Oligocene Isotope Step (EOIS; Hutchinson et al., 2021) reflects a 2.5°C cooling of bottom waters and growth of a continent-wide ice sheet on Antarctica (Bohaty et al., 2012;Lear et al., 2008). The cooling of bottom waters (Lear et al., 2008) leaves 0.6‰ of the δ 18 O benthic shift as an increase in seawater (δ 18 O seawater ), suggesting an ice sheet 60%-130% of modern East Antarctic Ice Sheet at 33.7 Ma (Bohaty et al., 2012).
Sierra Mixe maize is a geographically remote landrace variety grown on nitrogen-deficient fields in Oaxaca, Mexico that meets its nutritional requirements without synthetic fertilizer by associating with free-living diazotrophs comprising the microbiota of its aerial root mucilage. We selected nearly 500 diazotrophic (N 2 -fixing) bacteria isolated from Sierra Mixe maize mucilage and sequenced their genomes. Comparative genomic analysis demonstrated that isolates represented diverse genera and composed three major diazotrophic groups based on nitrogen fixation gene content. In addition to nitrogen fixation, we examined deamination of 1-amino-1-cyclopropanecarboxylic acid, biosynthesis of indole-3-acetic acid, and phosphate solubilization as alternative mechanisms of direct plant growth promotion (PGP). Genome mining showed that isolates of all diazotrophic groups possessed marker genes for multiple mechanisms of direct plant growth promotion (PGP). Implementing in vitro assays corroborated isolate genotypes by measuring each isolate’s potential to confer the targeted PGP traits and revealed phenotypic variation among isolates based on diazotrophic group assignment. Investigating the ability of mucilage diazotrophs to confer PGP by direct inoculation of clonally propagated potato plants in planta led to the identification of 16 bio-stimulant candidates. Conducting nitrogen-stress greenhouse experiments demonstrated that potato inoculation with a synthetic community of bio-stimulant candidates, as well as with its individual components, resulted in PGP phenotypes. We further demonstrated that one diazotrophic isolate conferred PGP to a conventional maize variety under nitrogen-stress in the greenhouse. These results indicate that, while many diazotrophic isolates from Sierra Mixe maize possessed genotypes and in vitro phenotypes for targeted PGP traits, a subset of these organisms promoted the growth of potato and conventional maize, potentially through the use of multiple promotion mechanisms.
Terrestrial climate records for Antarctica, beyond the age limit of ice cores, are restricted to the few unglaciated areas with exposed rock outcrops. Marine sediments on Antarctica's continental shelves contain records of past oceanic and terrestrial environments that can provide important insights into Antarctic climate evolution. The SHALDRIL II (Shallow Drilling on the Antarctic Continental Margin) expedition recovered sedimentary sequences from the eastern side of the Antarctic Peninsula of late Eocene, Oligocene, middle Miocene, and early Pliocene age that provides insights into Cenozoic Antarctic climate and ice sheet development. Here, we use biomarker data to assess atmospheric and oceanic temperatures and glacial reworking from the late Eocene to the early Pliocene. Analyses of hopanes and n‐alkanes indicate increased erosion of mature (thermally altered) soil biomarker components reworked by glacial erosion. Branched glycerol dialkyl glycerol tetraethers from soil bacteria suggest similar air temperatures of 12°C ± 1°C (1σ, n = 46) for months above freezing for Eocene, Oligocene, and Miocene timeslices but much colder (and likely shorter) periods of thaw during the Pliocene (5°C ± 1°C, n = 4) on the Antarctic Peninsula. TEX86‐based (Tetraether index of 86 carbons) sea surface temperature estimates indicate ocean cooling from 7°C ± 3°C (n = 10) in the Miocene to 3°C ± 1°C (n = 3) in the Pliocene, consistent with deep ocean cooling. Resulting temperature records provide useful constraints for ice sheet and climate model simulations seeking to improve understanding of ice sheet response under a range of climate conditions.
